DESIGN, TIME AND SETTING: In vitro cytology experiment was performed at the laboratory of Department of Pediatrics, First Affiliated Hospital of Guangzhou Medical College from December 2007 to August 2008. MATERIALS: MSCs were provided by 30 leukemia children admitted to First Affiliated Hospital of Guangzhou Medical College, including 22 acute lymphoblastic leukemia and 8 acute myeloblastic leukemia.

材料：骨髓间充质干细胞来源于广州医学院第一附属医院住院的30例白血病患儿，其中急性淋巴细胞白血病患儿22例，急性粒细胞白血病8例，患儿家属对实验均签署知情同意书。K562/AO2细胞株由天津血液病研究所提供。

材料和方法使用来源于人类骨髓基质细胞的细胞株HS-5作为滋养层细胞进行急性淋巴细胞性白血病细胞的体外培养，通过细胞生物学和免疫学方法评估培养体系并鉴定出难治性白血病细胞克隆；以ELISA和RT-PCR方法检测急性白血病细胞株和患儿白血病细胞VEGF及其受体的表达，了解不同治疗阶段VEGF及其受体的表达状况，并结合临床指标进行分析，明确VEGF及其受体在白血病发生过程中的作用；流式细胞仪和免疫荧光染色法对正常健康儿童、初发白血病患儿、复发白血病患儿及缓解后患儿进行凋亡因子检测和分析，初步阐明难治性白血病抗凋亡形成的原因；蛋白印记分析检测PI3K/AKT信号传导通路在健康儿童、初发白血病和复发白血病患儿的表达，初步了解难治性白血病形成的分子生物学机制。

The inoculation of MDV in general indued to decrease the production of IL-2、 IFN-γ-Th〓 cytokines in vitro, and there are differences between spleen and Thymus lymphocyte on the production of IL-2 in vitro, only thymus lymphocyte IL-2 prodution increased transient in early time. MDV接种总体上降低IL-2和IFN-γ-Th〓细胞因子体外诱生，其中IL-2体外诱生在胸腺细胞和脾淋巴细胞有差别，前者有早期短暂增强作用；而对TNF在上述两种细胞和骨髓巨噬细胞的诱生作用呈现类似规律，即呈现&V&形曲线，在接种后早期（1~5天）和25天升高，而在10天开始下降，至第20天降至低谷，MDV感染导致雏鸡细胞免疫水平下降，促进MD发病。

本文报告的结果表明7β-羟基胆固醇双琥珀酸单酯钠(RS 024)在有效抗肿瘤剂量下(45mg/kg/day，×7ip)对荷瘤小鼠骨髓细胞DNA、RNA和蛋白质合成有促进作用，对肝、脾的DNA、RNA合成无显著影响，但能促进肝，脾中及血清蛋白质合成。按400mg/kg/day连续给药30天，对大鼠红细胞、白细胞数及淋巴细胞百分率无明显影响。提示该药在有效抗癌剂量下毒性较小，并在一定程度上对免疫功能有增强作用。
