Bacterial strains were isolated from humus mud by employing the dilution methods. Seven cellulose-degradation Strains marked as B-37, B-35,B-31, B-25, B-17, Z-a, and Z-b were selected according to the hyaloid rings on the medium of carboxymethylcellulose sodium Congo red and their efficacy of decomposing filter paper. Z-b had the strongest ability in decomposing filter paper among the seven strains and 16S rDNA analyzed indecated that it was Stenotroplaomnonas maltophilia owning to 99.8% homology. Comparing clony size of the seven strains in media with different carbohydrates and inorganic salt, we found that the potato extract and inorganic salt which contained CaCl2 0.20, MgSO4 1.25, NaCl5.00,(NH4) SO4 1.30, KH2 PO4 1.35, FeSO47H2O 0.015, Na-EDTA 0.02 were suitable for the bacteria growth.

Seven major bacterial isolates were obtained and purified. Their 16S rRNA genes were amplified by PCR for sequencing and their identities were determined with homology comparisons. Five of the seven isolates belong to Actinomycetales including Kocuria KD139, Rhodococcus KD140, Rhodococcus KD142, Arthrobacter KD230, and Arthrobacter KD232; one is classified as Bacillus d KD178; and another one as Stenotrophomonas KD237. The phylogenetic tree was also constructed based on the analysis of 16S rRNA gene sequences.
