Polymarase Chain Reaction may be the better method of detecting M.tuberculosis than other methods and lead us to carry out a series of research on this problem. This thesis is composed of five parts:(1)systematic evaluation for the first time of the ability of five defferent PCR methods to detect M. tuberculosis;(2) using the endonuclease and southern blot by a radioactive and a nonradioactive labeled probe, to confirm the specificity of the PCR products and to analyse the sensitivity of detection by southern blot method using digoxigenin-labeled probe;(3) evaluation of acid-fast staining, bacterial culture and PCR for the detection of M. tuberculosis in sputum, pleural and peritoneal effusion samples;(4) The feasibility of PCR for detection of M tuberculosis on formalin-fixed paraffin embedded tissues;(5) 26 cases of sarcoidosis were retrospectively studiedwith PCR for M. tuberculosis and nontuberculosis Mycobacterium and the problems on histopatholoical diagnosis of sarcoidosis are discussed.

Results (1)CIMT and FIMT in CHD group were significantly higer than those in non-CHD group(p<0.05), The detection rate of plaques in CHD group was significantly higer than those in non-CHD group(p<0.05);(2) The sensitivity of carotid and femoral atherosclerosis plaques in young CHD patients was 62% and 57%. The specificity was 90% and 86%. The positive predictive value of carotid and femoral atherosclerosis plaques in young CHD patients was 92% and 88%. The negative predictive value was 58% and 53%. The predictive value of combinant measurement of carotid and femoral atherosclerosis plaques was not improved.

Then transplantation of the sciatic nerve from donor rats to the recipient rats in two groups were progressed.7 days after nerve transplantation,the contents of cd4 and cd8 in peripheral plasma of recipient rats were detected by flow cytometry.8 weeks after nerve transplantation,transplanted nerve were observed under light microscope.results:compared with nonirradiated group,the percentage of cd4 and the ratio of cd4 to cd8 in uvb irradiated group were significantly decreased(p<0.05,p<0.01),cd8 percentage was significantly increased(p<0.01).conclusion:uvb-irradiated donor splenocytes were injected in the recipient rats through caudal veil,which could induce immunological tolerance to allogeneic transplantation.

EMB-resistance in 18 isolates was caused by the mutation at coden 306 in embB gene with positive rate of 62.1%. The PCR result was negative in 20 specimen from pulmonary diseases without active TB and 5 negative control strains of no MTB. The data in this study showed high specificity of 100%. Only one specimen in 7 specimen with negative result in acid-fast stain and cultivation was shown alteration in coden 306 in embB gene.
结果:130例活动性肺结核患者痰标本抗酸染色阳性38例,阳性率为29.2%(38/130);罗氏培养阳性为79例,阳性率为60.8%(79/130)。79例培养阳性痰标本中耐乙胺丁醇29例,其中有18例在embB基因met306ATG突变,基因突变率为62.1%。20例非结核肺部疾病患者痰标本和5株非结核分枝杆菌菌株经套式PCR扩增全部为阴性,特异性为100%。7例涂阴培阴PCR扩增阳性的序列中有1例在embB基因306位密码了发生突变。
