The genome of infectious hepatitis B virus is a partially double stranded circular DNA, and that of replicative HBV is a covalently closed circular supercoiled DNA. The patients with chronic HBV infection manifest from non-active virus carrier state to severe active hepatitis, and 15%~25% of which will evolve decompensate cirrhosis and hepatocellular carcinoma.

感染型的乙型肝炎病毒基因组为部分双链环状DNA，复制型的HBV基因组共价闭合环状DNA慢性HBV感染的进展过程多变，从非活动性病毒携带到严重的活动性肝炎；其中约15%~25%的病人最终导致失代偿性肝硬化、肝细胞癌。

Methods Peritoneal macrophages of guinea pigs were infected in vitro by three different Leptospira strains, the virulent Leptospira interrogans serovar Lai type strain Lai, the avirulent L. interrogans serovar Lai type strain IPAV, and the nonpathogenic L. biflexa serovar Patoc type strain PatocⅠ, respectively, and heat inactivated Staphylococcus epidermidis was added 0.5, 1.5, 3 and 6 h after infection and incubated for 30 min. The effect of Leptospira on the phagocytosis of macrophage was evaluated by the inactivated Staphylococcus epidermidis phagocytosis rate and phagocytosis index. Phagocytosis and ultrastructure of peritoneal macrophages were observed by transmission electron microscopy 3 h after infection, and changes of cytoskeleton of the macrophages were observed by laser scanning confocal microscopy.

It was confirmed through passive immunization of IgY that healthy nonimmunized rabbits whose weight varied during 2Kg and 2.5Kg were intramuscularinjected with IgY 5.0ml (HI=13log2),they could resist the attact of 1.0ml（HA=9log2） RHDV-RC.

The results showed that: the explants of strawberry leaves were rather sensitive to kanamycin, and the corresponding concentration of 20 mg/L could substantially inhibit the growth of non-transformed tisse and be used to screen the leaves of strawberry after transformation. Timentin at the concentration of 300 mg/L could be used to eliminate Agrobacterium tumefaciens and had minor impact on the growth of the explants. The preculture time of 2-4 days was beneficial to improve transformation frequency. Co-culture for 2-3 days not only had higher gus transient expression but also avoid the over-growth of agrobacterium. The bacterial concentration of OD(subscript 600nm) 0.4 was the best inoculum density and 10 minutes was the best inoculation time.

结果表明：草毒叶片外植体对卡那霉素较为敏感，适宜的筛选浓度为20mg/L，可明显抑制非转化组织的生长；300mg/L的羧吩青霉素可有效抑菌，对外植体生长影响也较小；预培养2-4d有利于转化；共培养2-3d有利于提高转化频率并避免了农杆菌的过度生长；OD(下标 600nm)值为0.4的菌液侵染10min效果最佳。
