Six weeks after implantation, small amount of oval lacunae appeared. PGA fibers began to be absorbed. The amount of collagen increased and Safranin O stained positively. Eight weeks after implantation, cartilage with single or occasionally double chondrocytes in round lacunae was found.

体内埋植4周后可见PGA纤维间被不成熟的软骨细胞充填。6周时出现椭圆形软骨陷窝，PGA纤维开始吸收，胶原增多，番红花—"O"有阳性染色。8周时软骨陷窝呈圆形，单个，双个对称或多个排列。

Sections revealed a tumor of mild to moderate cellularity composed of bland uni- and binucleated cells lying within lacunae amid a chondromyxoid stroma, with foci of calcification and ossification (Figure 2, A ).

病理切片显示肿瘤由位于陷窝内的单核和双核细胞组成，陷窝位于软骨粘液样间质之中，并伴有灶状钙化和骨化(见图2，A)。

Methods Marrow cells from NH mouse were harvested and cultured in α-MEM with 10% fetal bovine serum.The appearance of tartrate-resistant acid phosphatase-staining multinuclear cells and formation of bone resorption pits were measured after 3,6,9 and 12 day exposure to various concentrations of 1,25-2D3 in culture. Results 1,25-2D3 (concentration higher than 10-9mol/L) could induce recruitment of OLCs and their bone resorption activity in a dose-dependent manner.

收集NH小鼠骨髓细胞于含10%胎牛血清的α-MEM培养基中行体外培养，设置不同的1，25-2D3浓度组和给药时间组，并于培养第3、6、9、12天观察记录抗酒石酸酸性磷酸酶(tartrate-resistant acid phosphatase，TRAP)阳性多核巨细胞［或破骨细胞样细胞(osteroblast-like cell，OLC)］以及骨磨片上骨吸收陷窝的数目。

Results: At the concentrations of 0.1, 1 and 10 μmol/L, berberine dose-dependently suppressed the formation of TRAP-positive multinucleated cells, the TRAP activity and the osteoclastic bone resorption. The strongest inhibitory effect was exhibited at the concentration of 10 μmol/L, with the inhibiting rate of 60.45%, 42.12% and 72.69% respectively.

结果：小檗碱在0.1~10μmol/L范围内，浓度依赖性地抑制TRAP阳性多核破骨细胞的形成和TRAP活性，减少破骨性骨吸收陷窝的面积；在10μmol/L浓度下，对破骨细胞的抑制作用最强，对TRAP阳性多核破骨细胞的形成和TRAP活性的抑制率分别达到了60.45%和42.12%，骨吸收陷窝面积减少72.69%。

Histopathological observation: 4 weeks after operation, diffuse presence of empty lacunae and pyknotic nuclei of osteocytes were found in the trabeculae, and few newborn micrangium could been seen in group A; lots of empty lacunae and a small quantity of newborn micrangium could been found in group B; and large amounts of osteoblats and newborn micrangium were found around the necrosis regions in groups C and D.

组织病理学观察，术后4周A组见空骨陷窝，无新生毛细血管；B组空骨陷窝较多，有少量新生毛细血管：C、D组空骨陷窝减少，坏死区有大量增生活跃的成骨细胞及新生毛细血管。D组空骨陷窝阳性数及微血管密度分别为19.30 1.52和7.08 1.09，与其他组比较差异有统计学意义（P〈0.05）。

Moreover, the border of resorptive lacuna became clouding.

与对照组相比，培养第三、五、七天10~(-5)mol／L、10~(-4)mol／L、10~(-3)mol／L NaF组骨片上骨吸收陷窝的数量和面积均减少，差异有统计学意义(P＜0.05)，而且陷窝周缘也变得模糊。

Results ICD (0.5,5 and 50 μmol/L) caused dose-dependent reduction in the number of osteoclastic bone resorbing lacunae. The inhibitory effect ICD (5 μmol/L) was more remarkable than that of aminopropanediphosphonate. Both ICD and APD reduced the surface area of osteoclastic bone resorbing lacunae.

结果 ICD(0.5，5与50 μmol/L)使体外破骨细胞性骨吸收陷窝数目呈剂量依赖性减少，中等浓度(5 μmol/L)的抑制作用明显高于阳性对照药帕屈磷酸钠，两药均可使吸收陷窝表面积明显降低，而且ICD作用更明显。

At the end of the culture, PBMCs cultured on coverslips were fixed and stained for tartrate resistant acid phosphatase, a marker of osteoclast.

对培养终末细胞作组织化学染色，检测破骨细胞特征性标志物抗酒石酸磷酸酶的表达；并以象牙磨片上虫蚀样骨吸收陷窝的形成为指标检测其生物学活性。

RESULTS: Light microscope showed that the collagen fibers arranged regularly in AECM with blankness of bone lacunas by HE, Mallory-Heidenhain rapid one-step dyeing and that the region around bone lacunas was stained different degrees of blue-green by Alcian blue dyeing.

结果 HE、Mallory- Heidenhain氏快速一步法染色后镜下可见AECM胶原纤维排列规则，骨陷窝空虚；阿利新蓝染色可见 AECM骨陷窝周边染色呈不同程度蓝绿色；免疫组织化学染色可见 FN和 L N的抗体染色阳性部位在细胞外基质。
