Polymarase Chain Reaction may be the better method of detecting M.tuberculosis than other methods and lead us to carry out a series of research on this problem. This thesis is composed of five parts:(1)systematic evaluation for the first time of the ability of five defferent PCR methods to detect M. tuberculosis;(2) using the endonuclease and southern blot by a radioactive and a nonradioactive labeled probe, to confirm the specificity of the PCR products and to analyse the sensitivity of detection by southern blot method using digoxigenin-labeled probe;(3) evaluation of acid-fast staining, bacterial culture and PCR for the detection of M. tuberculosis in sputum, pleural and peritoneal effusion samples;(4) The feasibility of PCR for detection of M tuberculosis on formalin-fixed paraffin embedded tissues;(5) 26 cases of sarcoidosis were retrospectively studiedwith PCR for M. tuberculosis and nontuberculosis Mycobacterium and the problems on histopatholoical diagnosis of sarcoidosis are discussed.

论文共分为五个部分，(1)首次系统地比较了五种不同引物序列的PCR检测结核杆菌方法的特异性和敏感性等情况；(2)利用限制性内切酶酶切分析和同位素及非同位素标记DNA探针进行Southern杂交的办法，分析PCR产物的序列与原设计是否相符及杂交检测PCR产物对提高检测结核杆菌敏感性的情况；(3)并对PCR用于快速检测临床标本中的结核杆菌进行了初步评价；(4)探讨了PCR用于检测石蜡包埋组织中结核杆菌的作用；(5)并利用PCR检测结核杆菌和非结核分枝杆菌的方法对26例原病理诊断为结节病的病例进行了PCR检测及组织病理学诊断的探讨。

The content of the parental rights and minor children's rights of privacy has all aimed to the private life of minor children's study and entertainment, which has created the condition of the conflicts between the two parties. Neither the law of our country has detailed stipulated for the content of parental rights, nor stipulated restrictedly to minor children's rights of privacy. The legitimate right is the basis to realize the right and ensure the right is legal. Because of the unperfected law, both two parties haven't got a clear confine in using their rights. So it also becomes one important reason of the conflict. In the past, parents use their guardianship as the hegemony which including some long-history autocratic tradition. It is the cultural factor of disregarding and excluding the individual privacy. With the development of democracy and legal system, citizen including minor children have realized their rights to privacy which has also become one reason to raise the right conflict.

Subsequently, the optimized technology was successfully scaled up to 〓 pilot bioreactor scale by using the parameter simulation method. Restrictive changes of TCR Vp repertoire and clonal expanded T cells could be found in peripheral blood T cells from patients with T-ALL. However, the clonal expanded T cells' property (clonal expansion leukemic cells or leukemia antigen-specific expansion T cells) should be further characterized. It may play a certain role on detection of minimal residual disease and design of anti-leukemia idiotypic vaccine. The dominant utilization of TCR VjJ repertoire could be found in peripheral blood T cells from patients with B-ALL, meanwhile clonal expansive T cells were existed. It may be a feature of the host immune response for leukemia-associated antigen. The clonal expansive tendency of T cells in V021 and VP23 subfamilies was rather obvious, which may be correlated with certain malignant B-cell clone. The CDR3 sequences of monoclonal expansive T cell in T cell strains were different, which was the base of developing DNA vaccine.

T-ALL患者外周血T细胞的TCR Vβ谱系出现限制性改变，均可检测到克隆性增殖T细胞，尚需进一步鉴定其性质(肿瘤性或抗原特异性增殖)，对于研究微小残留病变检测和设计抗白血病独特型疫苗均有一定的意义。B-ALL病人外周血T细胞的TCR Vβ谱系呈现优势利用的特点，并存在克隆性增殖的T细胞，这可能是机体对白血病相关抗原产生的特异性免疫反应；Vβ21和Vβ23亚家族T细胞发生克隆增殖改变的趋向性比较明显，可能与B-ALL相关抗原刺激有关。T细胞株中检测到的肿瘤克隆其CDR3序列具有高度的特异性，是构建DNA疫苗的基础。
