Results: In contrast to negative expression in normal oral mucosa, survivin was expressed in 42 of 50 (84%) cases of oral squamous cell carcinoma samples. There was no significant correlation between survivin expression and age and sex. In squamous cell carcinoma tissues, the expression of Survivin gene stepped up following malignant degree, but the positive rate of Survivin was not statistically significant.

结果：Survivin蛋白在10例正常口腔粘膜组织中呈阴性，而在50例口腔鳞癌组织中有42例阳性，占84%，差异有极显著性(P.01)；COX-2蛋白在10例正常口腔粘膜组织中呈阴性，但在口腔鳞癌组织中的阳性率为86%(43/50)，差异有极显著性(P.01)；Survivin蛋白阳性率与年龄和性别无相关性，而与COX-2蛋白表达阳性呈正相关(P.05)。

The SMMC-7721 hepatoma cell line of experimental groups and control groups were cultured with the addition of sodium selenite, potassium iodide or triiodothyronine (T3). The cell quantity was checked by the MTT assay , and the cell cycle was observed by flow cytometry and levels of AFP and ALB in the culture solution were determined by RIA.The SD rats were divided into positive control group, negative control group, T3 group and mixture group (the mixture of sodium selenite, KI and T3). After two weeks of the administration of sodium selenite、 KI and T3,Walker256 cells were transplanted into the abdominal cavity. The administration was kept on for another two weeks, then observed the morbility and mortality of the rats with ascites carcinoma. At the end, the ascites were for the cells counting and the cell cycles analysising.

方 法：在体外，应用细胞培养的方法，培养SMMC－7721肝癌细胞系，分为试验组和对照组，试验组加入不同浓度Se、I或T3干预，培养一段时间后，通过噻唑蓝试验、流式细胞仪，观察细胞数量及细胞周期的变化，测定细胞培养上清液中甲胎蛋白、白蛋白的水平，观察细胞分化情况；在体内，将试验用SD大鼠分成阴性对照组、阳性对照组、T3组和混合组（为Se、I和T3混合物），首先预防性用药2周，然后给试验组大鼠和阳性对照组大鼠腹腔注射Walker256细胞，制造腹水癌大鼠模型；继续用药2周，观察各组大鼠发病率及死亡率，测量不同时期大鼠血清FT3含量，通过流式细胞仪测量大鼠腹水细胞数及细胞周期变化。

Results Analysis of the expression of proteins of JAK2 in leukemic blasts, there was no detectable constitutive expression of phosphorylated JAK2 in all the AML samples analyzed; Furthermore, we could detect a constitutive activation of STAT 3 by immunoprecipitation of the targeted protein in primary leukemic blasts from 3 of 11 AML patients without any cytokine treatment; Pretreatment clinical characteristics, and treatment regimens did not differ significantly between patients with and without constitutive STAT3 activity, there was not significant relation between the constitutive activation of STAT3 and the FAB subvariety and chromosome karyotype; However, the 3 patients with constitutive STAT3 activity seemed to have adverse treatment outcome, one did not show remission, the other two patients relapsed shortly after remission; Consistent with the above-mentioned result, leukemic blasts from the 3 samples with constitutive expression of STAT3 showed higher percentage of spontaneous apoptosis and S-phase cells.

在实验研究基础上，本研究进一步运用免疫沉淀和Western Blot印迹方法，检测AML患者原代细胞JAK2和STAT3活化情况，并分析了STAT3活化与AML患者预后有关指标改变的关系。结果我们的初步研究发现，11例AML患者中，有3例呈现STAT3组成型活化（STAT3阳性，下同），无一例呈JAK2组成型活化。上述3例STAT3组成型活化与FAB分型和染色体核型改变未见明显相关性；在此3例患者中，1例未取得缓解，另外2例虽然取得短暂缓解，但均在二个月内复发。上述3例STAT3阳性AML患者S期白血病细胞百分率数值较组高，而凋亡率数值较低。
