A pair of primers were designed based on M1 gene sequence of known H5N1 Avian influenza virus.M1 gene was cloned from total RNA,extracted from tissue of H5N1 subtype virus inoculated embryo by reverse transcriptase-polymerase chain reaction using high proofreading polymerase(Pyobest~TM DNA Polymerase),and expressed using Invitrogen champion~TM pET directional TOPO expression system.Recombinant protein containing polyhistidine(6xHis) tag in N-terminal about 29.8kDa in size,wac obtained and purified.

根据已发表的禽流感病毒M1基因序列设计合成PCR克隆引物，自接种H5N1亚型病毒的鸡胚组织中提取RNA，反转录后采用高可信度DNA聚合酶经PCR扩增M1基因，采用Invitrogen定向表达系统(ChampionTMpET directional TOPO expression system)进行克隆表达，纯化获得N末端携带多聚组氨酸标签的重组蛋白，分子量约29.8 kDa。

Secondly, the genomic DNA is extracted from Zostera marina L. and then amplified by PCR with two oligonucleotide primers synthesized to obtain the partial nucleotide sequences of the Na~+/H~+ antiporter gene. The Na~+/H~+ antiporter gene clone was obtained after a series of manipulation: purificatioin of PCR product, ligation and transformation of recombinant plasmids, blue-white selection and so on. DNA sequence analysis shows that its DNA sequence is highly homologous with the genomic DNA sequence of E.

其次，通过提取大叶藻基因组DNA，根据设计的简并性引物进行PCR扩增，纯化PCR产物，连接，重组质粒转化，蓝白斑筛选等操作，并测序，提交测定序列到NCBI进行BLASTN，结果表明：该序列与大肠杆菌的基因组序列相似性较高，而与其它物种的Na~+/H~+逆向转运蛋白基因没有相似性。

Methods The total RNA was acquired from young rabbit particular cartilage, the BMP7 gene was inserted into pcDNA-3.1 to construct eukaryotic expression vector plasmid of pcDNA3.1-BMP7; the target gene-BMP7 was identified respectively by PCR analysis, restriction enounces analysis and nucleotide sequencing; the plasmid was transfected into adult rabbit knee articular chondrocytes, then the expression of BMP7 was detected by in situ hybridization, PCR and Western blotting.
