釉质细胞

PURPOSE: To study the apoptosis and expression of proliferating cell nuclear antigen, tissue non-specific alkaline phosphatase of stratum intermedium cell in postnatal stage of BALB/c mouse.

目的：观察小鼠釉质发育过程中成釉器中间层细胞的增殖、凋亡和其组织非特异性碱性磷酸酶(tissue non-specific alkaline phosphatase， TNSALP)的表达，探讨中间层在釉质形成过程中的可能作用。

It is suggested that the stratum intermedium participate in the differentiation of ameloblast and formation of enamel.

中间层细胞的增殖和凋亡及其TNSAP的表达与成釉细胞具有相关性，提示中间层细胞有可能参与成釉细胞的分化及釉质形成。

After 16 weeks growth, continuous and intact ameloblast, enamel, dentin, odontoblast was found.

生长16周的人牙胚，已经形成连续完整的成釉质细胞，牙釉质、牙本质和成牙本质细胞层。

Results showed that β-catenin was positive expression in stellate reticulum,inner dental epithelium,and dental sac at cup stage,in inner dental epithelium,dental papilla cell,and cell nucleus at early bell stage,in cervical loop and cusp cell membrane at late bell stage,and in ameloblast and odontoblast cell at amelogenesis stage.

结果表明，帽状期在星网层、内釉上皮、牙囊均为细胞核阳性表达；钟状早期在内釉上皮、牙乳头细胞核阳性表达；钟状晚期在颈环、牙尖细胞膜阳性表达；成釉期早期在成釉质细胞、成牙本质细胞阳性表达；成釉期晚期均阴性表达。

Histochemical study indicated that most of amelogenin stayed in enamel matrix and othersdiffused to dentin, dental tube and odontoblast layer.

组织化学研究结果显示在釉质形成期，釉质蛋白大部分留在釉基质层，也有一部分向牙本质内扩散到牙本质、牙本质小管以及造牙本质细胞层内。

Simmer. Results: Immunohistochemistry showed that the temporal and spatial expression patterns of AMELX, AMBN and ENAM in ameloblasts and enamel matrix were different. With the mineralization of enamel matrix, expression of AMELX decreased, but it weakly expressed in adjacent bone.

结果：免疫组化结果显示，三种蛋白在三种小鼠牙胚的成釉细胞、釉质、牙本质中的表达存在时空顺序差异，随着牙胚发育和釉质成熟，AMELX呈递减表达，而AMBN和ENAM则较恒定表达。

The ameloblasts are the key cells during odontogenesis, and they have been already cultured in vitro. The amelogenin is the extracellular matrix synthesized and secreted by the ameloblasts, and it plays a crucial role during amelogenesis.

成釉细胞是牙器官形成的关键细胞，国内外已成功离体培养出成釉细胞，成釉细胞合成和分泌的细胞外基质－釉原蛋白在釉质的形成中起着关键作用。

After 8 weeks growth, the polarization became more obvious in the pre-ameloblast and pre-odontoblast. Cells became high stylolitic shape and small amounts of dentin formation were found in the future of dental cusp region.

On the termination date, the cultured explants were all examined by Western blot, HE and transmission electron microscope. Our results showed that after 12-days in culture, the cultivation treated with AS-ODN reduced the synthesis of AMBN and had a deformed dental cusp with thinner enamel matrix. Ultrastructure analyses showed that there was hardly any cisternae of the rough endoplasmic reticulum in the ameloblasts at the tip of the cusp of AS-ODN treatedexplants. However, on average the enamel matrix was thinner compared with that in the control group. Furthermore, the collagen fibers in extracellular matrix were found disorganized. These findings seemed to provide a direct experimental evidence that tended to indicate that the arrested AMBN translation in cultured tooth germs might result in the delay of the tooth development.

经用Western蛋白印迹检测表明，所设计的反义核酸对AMBN InRNA具有良好的封闭效果并成功阻断了牙胚对AMBN的表达；在缺乏AMBN情况下，与对照组相比，实验组牙胚在体外可以继续生长发育至钟状晚期，出现成釉细胞和成牙本质细胞的分化，成釉细胞可以分化成为分泌期型成釉细胞，胞浆中缺少合成蛋白质所必需的粗面内质网和高尔基氏体，缺乏溶酶体，表明对蛋白合成和脚的能力降低；实验组牙胚有牙尖形成和基质分泌，但牙尖形态异常，基质形成减少，牙尖周围基质最厚处为O.6卜m，明显薄于对照组的5.spin，基质中胶原纤维粗细不等，排列稀疏， 3 第四军医大学硕士学位论文未见钙化现象，充分证明了AMBN在牙胚发育中参与釉质基质形成和矿化过程，影响胶原纤维和牙本质基质的合成，促进成釉细胞对蛋白质的合成和釉质基质蛋白降解。
