采用大鼠大脑中动脉缺血2 h/再灌注22 h模型，神经病学评分，脑梗塞范围及脑组织水含量变化，观察脑康灵胶囊抗脑缺血/再灌注损伤的效应；通过测定大鼠脑组织中一氧化氮合酶（nitric oxide synthase，NOS），超氧化物歧化酶（superoxide disumtase，SOD）活性及丙二醛（malondialdehyde，MDA）含量的变化以探讨药物作用的机制。

方法采用大鼠大脑中动脉缺血2 h/再灌注22 h模型，神经病学评分，脑梗塞范围及脑组织水含量变化，观察脑康灵胶囊抗脑缺血/再灌注损伤的效应；通过测定大鼠脑组织中一氧化氮合酶（nitric oxide synthase，NOS），超氧化物歧化酶（superoxide disumtase，SOD）活性及丙二醛（malondialdehyde，MDA）含量的变化以探讨药物作用的机制。

The results of this study are summarized below.1 A variety of additives such as proteins,glyoxal / glycerol,alcohols,metal salts,as well as ethylene monomer were used in this study and all of them could improve the leaching resistance of the three kinds of borate(boric acid,borax,disodium octaborate tetrahydrate) in different extents through variety forms.

本研究的结果归纳如下：(1)本研究中所使用的多种助剂如蛋白质、乙二醛/丙三醇、醇类化合物、金属盐以及乙烯单体等都能通过各种形式，不同程度的提高三种硼酸盐(硼酸、硼砂、四水合八硼酸二钠)的抗流失性，但效果均不是特别明显。

Methods 1.Preparation of 5-Fu Model and TSA treatment Two weeks" Wistar rats provided by the animal center of the China Medical University. The animals were sacrificed by 10% Chloral Hydrate 0.4ml/100g. Then the tracheas were excised aseptically. Then they were douched with PBS and cultured in 1:1 mix of Dulbecco"s modified Eagle"s medium and Ham"s F-12 medium (DMEM/F12) containing 120 mg/ml 5-Fu and 10% fetal bovine serum for 12h at 37°C. The tissues were then refreshed by culturing in DMEM/F12 containing 10% FBS and 200nM TSA.

1、制备气管损伤模型及剥离上皮取约200克左右的Wistar大鼠，雌雄不限(中国医科大学实验动物中心提供)，腹腔注射10%水合氯醛0.4ml/100g，无菌条件下取出气管，无菌PBS反复冲洗，洗净血液和粘液，置于DMEM/F12培养液中(含10%胎牛血清)，于培养液中加入终浓度为100mg/ml的5-Fu，37℃，5%CO2孵育12小时，弃去上述培养液，换成新鲜DMEM/F12液(含10%胎牛血清)，同时加入终浓度为200nM的TSA继续培养，于换液后3、6、12、24、48小时分别10%中性福尔马林固定，石蜡包埋，制成4μm厚的组织切片。

采用左冠状动脉前降支结扎开放建立心肌缺血／再灌注模型，健康SD大鼠24只，随机分为心包打开假手术组(A组，n=6)，缺血／再灌注对照组(B组，n=6)，5 mg·kg~(-1)氯胺酮+缺血／再灌注组C组，n=6，缺血30min后右腹股静脉注入5 mg·kg~(-1氯胺酮，10mg·kg~(-1)KTM+I／R组D组，n=6，缺血30min后右腹股静脉注入10 mg·kg~(-1氯胺酮。10％水合氯醛40 mg·kg~(-1)腹腔注射麻醉，气管切开，连接多功能监护仪记录心电图；小动物呼吸机人工呼吸，呼吸频率60次／分，潮气量2ml／100g，于左胸第四肋间打开胸腔暴露心脏，在左心耳下1mm左冠状动脉处，用丝线，眼科外用不锈钢小圆针穿过心肌浅层，稳定10min后将U型含有铜丝的胶管置于冠状动脉表面一起结扎(A组不结扎，B、C、D组结扎)；结扎开始左心室心尖部即由红色变暗，30 min后呈暗红色，心电图中出现S-T段抬高，说明缺血形成。
