The acidophile s composition and community structure in the sediment was studied with 16S rDNA clone library and denaturing gradient gel electrophoresis.

采集了酸性矿山废水区域酸性沉积物样品，运用16S rDNA克隆文库技术和变性梯度凝胶电泳技术，研究了沉积物中的嗜酸菌类群以及它们的群落结构。

The results show that this strain was a typical acidophile with optimal growth pH being 2.0 for hypha and pH 3.0-4.0 for the spore germination. Peptone was the most suitable nitrogen source which helped produce xylanase induced by xylan during stationary phase. The resultant xylanase reacted best at 50℃-55℃ and pH 3.5, and had activities 70% after 60 min maintenance under optimum reaction conditions.

结果表明：实验菌株嗜酸，菌丝生长最适pH为2.0，孢子萌发生长适宜pH为3.0-4.0；木聚糖诱导菌体在生长稳定期大量产生木聚糖酶，蛋白胨是菌体产酶的适宜氮源；菌株所产木聚糖酶属于酸性木聚糖酶，反应最适pH3.5、最适温度50℃-55℃，在最适反应条件下保温60min，残余酶活仍接近70%，适用于较强酸性的高温加工环境。

This nonmutagenic amino compound 4 (via the investigation into Salmonella/mammalian microsome assay) was also employed as diazo moiety coupling with acet-J-acid to form a diazo acid dye 5. The new dye 5 can color wool at 2% or silk at 3% o. w. f. into black one. The investigation of the uptake of 5 and its color appearance showed that it was assigned to an acid bluish black dye and its blackness was more excellent than that of Acid Black 10B.

Its high-capacity and strong-acid-resistance were proved. In the acid condition, the adsorption efficiency to acide green 6B and direct fast yellow RL reached above 90%. Through the adsorption rate curve on adsorption behavior of the three dyes, they were all according with pseudo-secondary adsorption rate curve. The rate constant of adsorption increased with increasing temperature. Through the adsorption isotherm of the three dyes, acide green 6B and direct fast yellow RL were according with Langmuir equation, while disperse blue 56 was according with Freundlich equation. The adsorption behavior was an endothermic process. Entropy was increasing during the adsorption. So raising temperature was in favor of adsorption.

结果表明，甲醛环氧氯丙烷交联壳聚糖对染料具有较强的吸附性能和耐酸性能，在酸性条件下，对酸性绿6B和直接耐晒黄RL的吸附效率达90％以上；拟合甲醛环氧氯丙烷交联壳聚糖对三种染料的吸附速率曲线，发现该吸附行为符合拟二级吸附速率曲线，其吸附速率常数随温度的升高而增大；通过测定不同温度下甲醛环氧氯丙烷交联壳聚糖吸附三种染料的吸附等温线表明，甲醛环氧氯丙烷交联壳聚糖对酸性绿6B和直接耐晒黄RL的吸附符合Langmuir方程，而对分散蓝56的吸附符合Freundlich方程，甲醛环氧氯丙烷交联壳聚糖对染料的吸附是一个吸热过程，吸附后体系熵增加，升温有利于吸附。
