Based on electro-characteristic and double electric layers models, directional mechanisms of soy protein molecular are analyzed for the first time on DC heating condition. After the main factors which effect soy protein gels transparency and strength being studied on DC heating condition, the different affects on soy protein gels transparency and strength by DC heating and water bath heating are analyzed and compared, and the transparent mechanisms of soy protein gels are further explored. In addition to temperature action as water bath heating, DC heating is added as electric field directional action. As a results, heating rate is increased. Directional range degrees of soy protein molecules are raised. These advantage the increase of soy protein gels transparency and strength. Microstructures of soy protein gels forming on DC heating condition are observed by SEM and the results showed that ordered structure of transparent soy protein gels on DC heating condition is more ordered than that of transparent soy protein gels on water bath heating condition.
首次从大豆蛋白的电特性和双电层模型上分析了直流电加热条件下,大豆蛋白分子定向的机理;通过对直流电加热条件下影响大豆蛋白凝胶透明性和强度主要因素的研究,分析和比较了直流电加热与水浴加热对大豆蛋白凝胶透明性和凝胶强度的不同影响,又进一步探讨了大豆蛋白凝胶透明的机理;直流电加热除了水浴加热中的温度作用以外,又增加了直流电场的定向作用,提高了加热速率,提高了大豆蛋白分子定向排列的程度,有利于蛋白质凝胶透明性和强度的提高;利用SEM对电场下大豆蛋白凝胶微观结构的观察,进一步证明了直流电加热比水浴加热形成的透明大豆蛋白凝胶微观结构具有更高的有序性。

Mainly using protein - film voltammetric technique, we have achieved direct electrochemical response of lots of proteins such as hemoglobin, myoglobin, cytocrome c, cytocrome b5, horseradish peroxiase, GOD ,Vitreoscillia sp. haemoglobin, etc., at a series of protein membrane modified electrodes, for instance, protein/kieselgubr, protein/polyethylene glycol, protein/polyethyleneimine, protein/phosphatidylcholine, protein/DNA, protein/montmorillonite/polyvinyl alcohol, protein/silver nanoparticle/ cysteamine membrane modified electrode, etc., which are prepared by casting, incorperating or layer by layer self-assembly methods. Electrochemical behavior of these proteins in the films have been studied, and their structural and conformational changes have also been examined with spectrometric techniques.

In this article, we used automotive immunohistochemical stainer to check the expressions of HBsAg and HCV antigens in 100 cases of HCC and their adjacent tissues, mean while, we checked their histological activity index and hepatic fibrosis degree. We also used radioimmunoassay to detect serum levels of 4 serum fibrosis markers and serum AFP levels in these cases. Immunofluorescence labeling and flow cytometry were used to check the T-cell subpopulations and the activity of NK cells of these 100 cases. Then the correlations among these factors were studied.
本文采用全自动免疫组化仪对100例肝癌组织及癌旁组织中的HBsAg、HCV抗原表达进行了标记,同时对其进行肝组织活动指数、纤维化分期;同时采用放射免疫测定方法检测同期血清透明质酸、Ⅲ型前胶原蛋白、层粘连蛋白和Ⅳ型胶原蛋白等四项血清肝纤维化标志物和血清甲胎蛋白水平;采用免疫荧光标记技术和流式细胞仪对T细胞亚群及NK细胞活性测定,对各项指标的相关性进行系列对比研究。

Results Same as hyaline cartilage, the calcified cartilage was stained light flavo-green and weak double refraction by the Picrosirius-polarization method. In the immunohistochemistry analysis of collagen Ⅱ protein, the calcified cartilage was stained strong green fluorescence and the collagen protein parallelled in joint surface near the tidemark. The content of collagen Ⅱ protein was (20.16%±0.96%) of tissue dry weight by amino acid analysis, and that of the hydroxyapatite was (65.09%±2.31%) of the inorganic composition in calcified cartilage.
