In order to diagnose disease caused by muscovy duck parvovirus rapidly and accurately, and at same time, further study the taxonomic relationship between muscovy duck parivovirus and goose parvovirus, two pairs of PCR primers (LHMP1/LHMP2 and LHGP1/LHGP2) were designed respectively base on complete nucleotide sequences of Muscovy duck and Goose parvoviruses registered in the GenBank, The LHMP1 for the upper one and the LHMP2 for the lower one, corresponding to the specific nucleotides sequence 454-472 and 1173-1155 respectively in MDPV, and the LHGP1 for the the upper one and the LHGP2 for the lower one, corresponding to the specific nucleotides sequence 454-472 and 1173-1155 respectively in GPV.

为了能快速、准确地对番鸭细小病毒病进行诊断，同时为了能进一步弄清番鸭细小病毒与鹅细小病毒在分类学上是一种怎样的关系，比较GenBank中MDPV和GPV的基因序列，针对这两种病毒左阅读框编码非结构蛋白基因序列的相同区段，分别构建两对PCR引物。第一对引物LHMP1／LHMP2是根据MDPV的基因序列设计的，上游引物针对MDPV基因序列的454～472；下游引物针对基因序列的1173～1155，为19个碱基，扩增跨幅为719bp。

In order to compare the homology of the gene encoding VP2 between MDPV-Q and MDPV which was registered in the GenBank, also in order to find out changes of the VP2 gene and the immuno-genicity between the wild-strain MDPV-Q and the attenuated strain MDPV-26 which derived by continued passage of virulent wild-type in muscovy duck embryo, a pair of primer (LHMP7/LHMP8) was designed. The upper one LHMP7 and the lower one LHMP8 corresponded to the MDPV-specific nucleotides sequence 2885-2900 and 4618-4604 respectively, according to the MDPV nucleotide sequence databases. The length of the sequence embraced by the primers was 1734bp.

为了获取这一基因片段与国外分离株进行同源性比较，同时也为了了解MDPV强、弱毒株VP2基因之间的异同关系，及其免疫原性的变化规律，通过DNA重组技术，设计了一对引物LHMP7／LHMP8，该对引物选取位于2885～2900及4618～4604的两段序列，跨幅为1734bp，并在这两条引物中分别加入两种限制性核酸内切酶SacⅡ和KpnⅠ的酶切位点，分别对MDPV-Q和由该株病毒经人工致弱后得到的MDPV-26株进行PCR扩增，并将PCR产物克隆到pMD18-T载体上，分别得到二个重组子：pMD18-T—M-Q VP2和pMD18-T-M-26 VP2。

The main research work and its results are: 1Empirical results are analysed for the relations of fault displacement and earthquake magnitude, and difference between permanent ground deformation and maxium dynamic relative displacement on the two sides of fault; 2An analytic method is proposed for resopnse of a buried pipeline due to earthquake fault movement; 3A new shell model with an equivalent boundary and relted finite element analysis method are proposed for estimating response of a buried pipeline under large fault movement; 4By the proposed method, damage performance of water supply steel pipelines with large diameter is simulated to the real cases in Kocaeli Earthquake and Ji-Ji Earthquake, and simulating results show the real failure mode of pipes is revealed; 5Effects of overlying soil and soft/hard interlayers are analyzed on ground rupture mode and degree due to earthquake fault, and some earthquake rupture phenomena in soil layers are theoretically described for the first time; 6Pipeline response characters are discussed for the case of considering effects of soil layers on ground rupture mode; 7Research results and proposed method are applied in seismic analysis of pipline acrossing fault in the Gas Tansportation Project from West to East in China.

主要工作和成果包括：1）分析了不同断层位错形式下地表破裂位错量与震级之间的经验关系，并基于集集地震近断层记录分析了断层两侧永久地面变形和地震动相对位移幅值之间的差别，为合理地考虑地震断层位错地面变形动态影响提供了依据；2）发展了断层位错地面大变形下管线反应分析模型并给出物理概念明确、简单实用的解析分析方法；3）建立了断层位错下埋地管线反应分析的等效边界壳模型有限元方法，方法中引入了以非线性弹簧形式模拟离断层较远处管段影响的等效边界，简化了计算模型并突出了近断层处管体反应；4）利用等效边界壳模型方法，对土耳其地震和集集地震中大口径埋地管线震害进行了模拟，结果表明所提方法可以更清地解释震害所表现的管体破坏特征；5）研究了覆盖土层和软硬夹层的存在对断层位错地面破裂形式和程度的影响，首次从理论上解释了某些地震地表破裂现象；6）探讨了考虑土层对断层位错地面变形影响的管体反应特征；7）本项目成果已在西气东输工程的管道跨断层抗震问题分析中得到了应用。
