Get the neonate Sprague-Dawley rats medulla spinalis, through the conventional primary cell culture procedure, we use the method of difference adherence time and get the motor neurons of anterior spinal cord. When the neuron is in maturity, we make the mechanical injury model in vitro. All the models were divided into four groups: group A is control group; group B is 100μM ATP group; group C is 100μM ATP+20μg/ml suramin group and group D is 100μM ATP+10μM ouabain+10μg/ml Thapsigargin group. Culture the four groups neurons, after one day, we count the motor neuron, observe the survival and activity of neurons through MTT shade selection experiment, use flow cytometry to analyze the percentage of apoptosis of motor neurons of anterior spinal cord and detect the expression of protein p-GSK-3β(ser9) through Western-Blot technology.

本实验主要由以下三部分组成:一、n-HA/PA66 复合材料在动物体内、体外的生物相容性及生物安全性评价二、n-HA/PA66 复合材料植入动物体内的生物活性及近期对机体钙、磷代谢影响的实验研究三、网孔 n-HA/PA66 复合材料作为支架修复兔桡骨节段缺损的动物实验研究主要研究目标及方法如下:参照 GB/T16886.5-1997-ISO 10993-5:1992《医疗器械生物学评价细胞毒性试验体外法》之评价标准和要求,采用规定的 L929 细胞(小鼠结缔组织成纤维细胞),分别经直接接触和材料浸提液与细胞共培养等方式对 n-HA/PA66 复合材料进行细胞毒性测试,采用细胞形态观察法观察两种细胞各组在 24h、48h、72h、5 天后各时相点的细胞形态学变化,并在显微镜下照相,从而对细胞与材料的生物相容性进行定性评价;同时采用细胞生长抑制法,以酶标仪定量测定评价各组 1,2,4,7 天 L929 细胞的相对增殖率,以定量测定并判别材料对细胞的毒性程度。

Pristine Li-Al LDHs are synthesized by hydrothermal process in different reaction conditions by varying the aging time for 1 and 24 hours. Both the Li-Al layered double hydroxides (abbreviated as Li-Al LDH1 and Li-Al LDH24 for aging time 1 hour and 24 hours, respectively) were modified by using sulphanilic acid sodium salt hydrate, modified agent to form modified Li-Al LDH1-SAS and Li-Al LDH24-SAS. The morphology of the pristine layered double hydroxides are investigated by using wide angle X-rays diffraction, scanning electron microscopy and particle size analyzer which indicates that the crystallinity and aspect ratio is increased with increasing the aging time from 1 hour to 24 hours. The particle size of Li-Al LDH24 and Li-Al LDH1 are found to be ~1000 nm and ~250 nm, respectively. Both the pristine and modified LDHs are characterized by XRD, FTIR spectra and thermogravimetric analysis.

本研究目的在於改变长晶时间的长短,合成出同径大小的Li-Al LDHs,再经由改质剂sulphanilic acid sodium salt hydrate将Li-Al LDHs无机层材表面有机官能化制备出改质型Li-Al LDHs-SAS,於是进一步藉由扫描式电子显微镜、射径仪和穿透式电子显微镜等仪器分别观察LDHs改质前和改质后其主层结构型态上的变化;从中可以发现Li-Al LDHs随著长晶时间的增加其径有随之增大的趋势,但经有机化改质后其径会由於改质环境的影响明显低许多,针对此现象本实验将未改质和改质后之Li-Al人工无机层材制备成复材进一步探讨其在热性质和难燃特性上的为表现。
