To produce the scientific evidence for developing and manufacturing new antitumor drugs.Methods: 1 The inhibitory effect on cell growth of Hela was measured by MTT assay in treated or untreated groups (3.125, 6.25, 12.5, 25, 50μg/ml TAM and control) for three different treatment times (24h, 48h and 72h).2 Apoptosis and cell cycle were measured by FCM in four experimental groups (0, 4, 16, 40μg/ml TAM) for 48h.3 Adopting Wright and Giemse's staining to observe the morphology of Hela cells which treated with 40μg/ml TAM.4 Using invasion experiment to detect the Hela cells'invasive abilities which treated with 40μg/ml TAM.5 The protein expressional levels of P-ERK, ERK, C-myc and Cyclin D1 in Hela cells untreated or treated with 4, 16, 40μg/ml TAM for 24h were measured by Western blot.6 Expression of anti-apoptotic gene bcl-2, apoptotic gene bax and MMP-9 in Hela cells of four experimental groups (0, 4, 16, 40μg/ml TAM for 24h), were observed by revers transcription PCR.7 The protein expression of P-ERK, ERK, Bcl-2 and Bax in Hela cells treated with 40μg/ml TAM for 24h observed by laser scanning microscopes.

1采用四甲基偶氮唑蓝法检测不同浓度北豆根总碱(3.125、6.25、12.5、25、50μg/ml)处理不同时间(24、48和72小时)对Hela细胞增殖反应的抑制作用。2采用流式细胞技术(flow cytometry，FCM)检测不同浓度北豆根总碱(0、4、16、40μg/ml)作用48小时，对Hela细胞凋亡及周期变化的影响。3瑞氏-姬姆萨染色后显微镜观察北豆根总碱(0、40μg/ml)作用24小时后Hela细胞形态学变化。4采用Transwell小室法检测北豆根总碱(0、40μg/ml)作用24小时后对Hela细胞侵袭性的影响。5采用免疫印迹方法检测不同浓度北豆根总碱(0、4、16、40μg/ml)作用24小时后，Hela细胞中磷酸化ERK、ERK、C-myc、CyclinD1的表达变化。6采用逆转录-聚合酶链反应(revers transcription PCR，RT-PCR)半定量检测北豆根总碱(0、4、16、40μg/ml)作用24小时，Hela细胞抗凋亡基因bcl-2、促凋亡基因bax、基质金属蛋白酶-9(MMP-9)的表达变化。7应用激光共聚焦显微镜(laser scanning microscope，LSM)观察北豆根总碱(0、40μg/ml)作用24小时后，Hela细胞内P-ERK、ERK、Bcl-2、Bax蛋白的表达变化。

The expression of RACK1 was conserved in various human organs and did not display a robust and stable pattern of rhythmicity in mouse brain. Up and down regulating of Per1 had no significant effects on the RACK1 expression in cell tests.

结果 人下丘脑区域RACK1蛋白与Per1存在相互作用；RACK1在多器官组织保守表达，但其表达未呈现明显节律性；上调及下调Per1表达，RACK1的RNA水平无显著变化。

The 5＇coding region of its eDNA was optimized according to the codon bias of Pichia pastoris to make it secretively express in Pichia pastoris with higher performance. Two-step DNA synthsis was used to synthesize the eDNA sequence being optimized of ADTZ. OPT-ADTZ was connected with constitutive plasmid pGAPZaA to construct the reecombinant plasmid pNOA. pNOA was linearized and then transformed into Pichia pastoris GS115. Then codonoptimized ADTZ was constitutively and secretively expressed in Pichia pastoris.

为了使重组蛋白rADTZ能在毕氏酵母中高效分泌表达，根据毕氏酵母密码子的偏好性对rADTZ的5'末端的编码区域进行了优化，利用two-stepDNAsynthsis技术合成出ADTZ优化的基因序列OPT-ADTZ，并与组成型表达载体pGAPZaA连接，构建重组质粒pNOA，线性化pNOA后转化至毕氏酵母GS115中，实现了密码子优化的rADTZ组成型分泌表达。

With western blot analysis and DNA polymerase chain reaction-single strand conformation polymorphism assay,protein product of Rb gene and mutation in exon 5～8 of P_(53) gene were examined in 17 cases of human testicular seminoma.

采用Western印迹和DNA聚合酶链反应－单股构造多态性分析法对17例人睾丸精原细胞瘤组织标本的抗癌基因Rb表达的蛋白产物和P_（53）基因外显子5～8的突变进行检测，发现有3例标本的Rb基因表达的蛋白产物缺失，有4例标本的P_（53）基因有点突变，对照的正常人睾丸组织和视网膜组织均呈Rb基因蛋白产物表达阳性，对照的正常人睾丸组织中未发现P_（53）基因突变。

RT-PCR and Real-time PCR analysis revealed that mRNA levels of the FK506-binding protein 12 (FKBP12) and two unknown genes were higher in the diapausing pupae, whereas the ecdysteroid-regulated 16kDa protein (esr16), NADH dehydrogenase 1 alpha subcomplex subunit 6, and two unknown genes were higher in the pupae of diapause termination.

Results 73 differentially expressed protein peaks were found between SLE patients and normal controls (P＜0.05), in which, 44 peaks had very significant difference (P＜0.01). Detection model of protein mass spectrometry (10542 Da m/z and 2554 Da m/z) was set up by ZUCIPDAS software with the discriminate rate and accuracy rate as 100%, which could distinguish the sample from SLE and normal persons. 10542 Da protein expression peak missed in SLE might be the mixed protein fragments peak of HLA gene family, while the 2554 Da protein expression peak might be a SLE-specific unknown protein.

结果SLE患者治疗前与正常人比较，找出差异表达蛋白峰73个（P.05），其中44个峰有非常显著性差异（P.01），用ZUCIPDAS软件建立的蛋白质质谱检测模型（10542 Da m/z和2554 Da m/z）的判别率和正确率均为100%，可以通过以上两个峰将SLE与正常人区别开来，10542 Da SLE缺失表达的质荷峰可能是HLA基因家族相关的多个蛋白片段的混合峰，而2554 Da可能是SLE特异表达的未知蛋白峰。

The results suggest the expression of Nm23-〓 Protein is more often seen in the cases of adenosis with atypical hyperplasia than in the cases of carcinoma, the expression of Nm23-〓 might be a marker of adenosis with atypical hyperplasia in cases of FCD; Lower level of Nm23-〓 suggests in had a high metastatic potentiality in breast cancer.

在浸润性导管癌组中，有腋窝淋巴结转移组的Nm23-〓含量显著低于无液窝淋巴结转移组。结果提示：Nm23-〓蛋白在不典型增生上皮中的表达比癌组织更常见，Nm23-〓蛋白过度表达可能是乳腺上皮不典型增生的一个特征；在乳腺癌病例中，Nm23-〓蛋白表达与腋窝淋巴结状态有关。

Results: Neonatal rats in hypoxic-ischemicgroup behaved restlessness, cyanosis, deep and rapid breath, astasia, lethargy, irritation and spasm; There were local pyknosis, fragmentation, dissymmetry, blur or disappearance of nucleoli, raritas of ground substance in left cerebral cortex; but there was no statistic significance in chinese traditional medicine group and sham group. The expression of HIF-1α mRNA in brain tissue of experimental group and chinese traditional medicine group were higher than that in sham group, especially chinese traditional medicine group at 3d; HIF-1α positive cells were found in cerebral cortex and hippocampus. There were more HIF-1α positive cells in experimental group and chinese traditional medicine group and mainly in vascular endothelial cells.

结果 缺氧缺血后大鼠相继表现为烦躁不安、全身发给、呼吸加深加快、站立不稳、嗜睡、激惹或间断发作的痉挛和抽搐；HE染色显示实验组大鼠左侧大脑皮层出现局灶性神经元核固缩，核碎裂，核仁偏位，不清或消失，基质疏松；中药组大鼠脑组织结构基本接近假手术组，无明显水肿和坏死表现；实验组和中药组大鼠脑组织HIF-1α mRNA表达较假手术组明显增加，尤其是中药组实验第3日者；免疫组化显示实验组和中药组各时间点大鼠大脑皮层和海马区均可见不同程度的HIF-1α表达，明显高于假手术组，阳性表达主要在血管内皮细胞，海马和皮层的锥体细胞亦有HIF-1α阳性细胞分布。

Results: we estalished ideal copd models in stort time. there existed the expression of nos in both the normal lung tissue and the lung tissue of copd,mainly orienrared in the cytoplasm of cell.the immunohistochemical result indicated that inos increased significantly ,in comparison with the control group, in bronchiolar mycoderma epithelial and enos remakably decreased in pulmonary blood vessel.

结果：联合应用烟熏和ppe在较短的时间内建立了较理想的copd模型；一氧化氮合酶在正常肺组织与copd的肺组织均有表达，主要定位在细胞浆内；免疫组化结果表明模型组inos在支气管黏膜上皮呈强阳性表达，肺血管enos表达明显减弱，与健康对照组比较有显著性（p.01）。
