In the present study, we examined the large-scale gene expression profiles with six sets of matched clinical and experimental fluconazole susceptible and resistant C. albicans strains by microarray analysis. We observed that azole resistances developed in multiple circumstances are all associated with increased expression of CDR mRNA. We also identified some common differential expressed genes and some interesting function-linked gene groups involved in drug-resistance.

利用所构建的基因芯片我们研究了共六对氟康唑敏感和耐药白念珠菌配对菌株的大规模基因表达谱，证实不同环境下产生的白念珠菌耐药性均与多药耐药基因CDR的mRNA表达增加有关，并从中鉴定了一些与药物耐受性相关的共同差异表达基因和一些有趣的功能连锁基因群。

There are three copies of R fragment in the S mt-genome, and each copy contains two closely adjacent open reading frames: orf355 and orf77. Among these three copies two BamHI fragments were cloned, and the sequence analysis showed that cox1 and cox2 gene lies upstream of the R region in each fragment, respectively. However, they locate in different strands and head to head with R. In this study, we found that:(1) The expression of cox1 and cox2 is not affected by the transcription of R region;(2) The two copies of R region transcribe in large amount in the microspores, and produce transcripts of 1.6kb and 2.8kb, respectively, in the sterile microspores; However, the abundance of these two transcripts is reduced in the fertility restored microspores, realized through the mRNA decay, and this reduction occurs at the uninucleate microspore stage;(3) In the sterile microspores, the 5 terminus of the 1.6kb transcript contains a palindrome region that can fold into a stem-loop.

本研究发现：(1) cox1和cox2的转录没有受到R区转录的影响；(2)两个拷贝的R区在在不育花粉中高丰度表达，其转录本大小分别为1.6kb和2.8kb；而在育性恢复的花粉中，这两个转录本的丰度被大大降低，进一步分析表明这一结果是通过mRNA分子的降解途径来实现的，此降解过程发生在单核花粉期；(3)在不育花粉中，1.6kb转录本的5′末端具有一段可形成茎环结构的迴文对称序列，而在可育花粉中，其5′末端短缺了9个碱基；(4)在不育花粉和育性恢复的花粉中，R区转录本的加尾位点都集中位于一个3′茎环结构之后；(5)不管是在不育花粉还是育性恢复的花粉中，orf77区域内均在第52位和100位核苷酸处发生了不同频率的C向U的编辑，从而形成UGA终止密码；(6)不管是orf55-orf77的成熟转录本还是转录前体分子，都已被加上了Poly尾巴，表明已进入降解途径；在以上实验结果的基础上，本研究对S型CMS花粉育性的恢复机理进行了如下推测：在不育花粉和育性恢复的花粉中，orf77区域内均发生了终止编辑，即由RNA编辑产生了提前终止密码子。

To reseach the mechanisms of ED wtih DM from the acpects of endocrinology, morphology and molecular biology, using rats from NDM and STZ groups as control, the concentration of testosterone, LH and FSH in sera, the microstructure of testicular tissue, bulbocavernous and ischiocavernous muscle tissue, and penile cavernous tissue, the ultrastructure of penile cavernous tissue, and the expression of nNOS mRNA, the content of nNOS and eNOS, the activity of NOS and the concentration of cGMP in the pathway of LARG-NO-cGMP in penile cavernous tissue from ED rats with DM were reseached or examinated in Group A, B and C.

本实验利用第一部分所建立的DM性ED模型，采用NDM及STZ大鼠作对照，通过对A组、B组及C组DM性ED大鼠血清中睾酮、LH及FSH浓度的测定，睾丸组织形态结构的研究从内分泌方面探讨DM性ED的发病机理；通过对阴茎海绵体组织显微及超微结构和球海绵体肌及坐骨耻骨肌显微结构的研究，首次从形态学方面探讨DM性ED的发病机理；通过对阴茎海绵体组织L-ARG-NO-cGMP通路中nNOS mRNA表达、nNOS及eNOS含量、NOS活性及cGMP浓度的检测，从分子生物学方面探讨DM性ED的发病机理。

Early in 1995, in his "Desire" series where red and blue lines come in strong contrast with each other, Ma Yiping instinctively used his personal experience to drop a hint of the transcendence of life. From 1996 to 2001, his "Images" series established the development direction of his art, owing to its artistic expression_r_r of overloaded psychological priori images. In his works before or after 2003, dream-like symbolic images presented perplexed feeling of human beings about the universe and the physical world. In his "images" series in 2006, uncertain images stood on black cube-shaped objects and ambiguous edges pointed to various uncertain directions with the accompaniment of silver light and shadow. At this point, the intuitive perception of the essence of nature made his artistic style gradually take shape. In his later "fetish" series, in which he gradually became mature in spiritual and symbolic expression_r_rs, Ma Yiping fully grasped the eternal sense of the spiritual world.

在早期，1995年的《欲望》马一平用对比强烈的红色、蓝色层层涂盖、本能地运用自身感受以暗示的手法表达对生命的超越。1996-2001年以物象为主题的系列作品对超载心理先验物象的表达方式确定了他艺术作品的发展方向，2003年前后的作品中，梦一般的暗示性物体程现了生命本体对宇宙物理世界的茫然，在2006年的《物象》系列中，黑色的方体上不确定的物状、模糊的边缘在泛着银色光影的衬托下，泛生着各种不确定的指向感，此时对本质直观的感知性的把握使作品艺术风格基本形成，到后来的《神物》已经充分地把握了精神世界里的永恒感，画面中所能达到的精神性和神密暗示意义的表现手法已近成熟。

By using electroporation transfects,the TGF〓expression vector were introduced into HL-60 cells.Thecells were allowed to grow for 1 day before beingsubjected to selection for the ability to grow in mediumcontaining 600ug/ml geneticin.Stable cell clones resistantto G418 sulfate were obtained after 2-3 WK growth withdrug-containing medium.Southern blot hybridization wascarried out.RNA slot have higher expression of TGF〓mRNA in HL-60 transfected.All results followed suggestedthat TGF〓 have suppressed the maliganant phenotype of HL-60 cells.

利用电击法将〓表达载体导入HL-60细胞中，经G418筛选培养20天后再行Southern杂交检测外源〓整合情况，利用RNA狭线杂交法证明了在转录水平上表达有所提高，进一步进行了流式细胞仪测定DNA含量，使细胞在〓期产生生长阻滞作用，细胞生长速度，在软琼脂中成集落数、在组织中的浸润性及在裸鼠中的成瘤性均发生变化，我们的实验结果表明TGF〓对HL-60细胞具有抑制恶性表型的作用。

Results （1）The positive rates or the scorings of MMP-1,MMP-9and MVD were significantly higher in human gallbladder neoplasm than that in human chronic cholecystitis（P.01）,But the positive rates of MMP-2in human gallbladder neoplasm were significantly lower than that in human chronic gallbladder cholecystitis（P.01）.（2）The closely positive correlations were found among the expressive scorings ofMMP-1,MMP-9,TIMP-2,MVD and the clinicopathological type,differentiated degree,with or without metastasic tumor.

结果 （1）31例胆囊癌组织中MMP-1，MMP-9的阳性表达率及其评分和MVD计数均值（以CD34阳性计数）分别为16例（51.6%）和20例（64.5%），明显高于10例慢性胆囊炎组织1例（10.0%）和2例（20.0%），差异有非常显著性（P.01）；胆囊癌组织中TIMP-2的阳性表达13例41.9%）则低于慢性胆囊炎组织9例（90.0%），差异有非常显著性（P.01）。

Results Total bilirubin, Direct bilirubin increased significantly. In BDL rat liver, the membrane protein MRP2 expression was significant down-regulation and no nuclear receptor FXR was expressed.

结果 梗阻性黄疸大鼠总胆红素、直接胆红素显著升高；与假手术对照组相比，梗阻性黄疸组大鼠肝细胞MRP2蛋白表达明显降低(P=0.041)，FXR蛋白无表达，差异均有统计学意义。
