The expression of AT2R gene can be regulated efficiently in vivo by local delivery of MSC with dual-stable expression of AT2R gene, thus reduces the formation of intimal hyperplasia.④AT2R gene can influence the expression of ECM and significantly promote the cellular apoptosis in the injury rat carotid artery.⑤These data demonstrate the clinical potential of AT2R regulatable expression to prevent restenosis after PCI.

研究结论：(1)与VSMC直接接触可以诱导MSC向血管成分细胞分化；(2)构建的Dox-on可调控的MSC细胞株诱导活性可靠，使外源AT2R基因的表达处于有效的主动控制下；(3)局部导入MSC不影响球囊损伤后大鼠颈动脉新生内膜增生；(4)由MSC介导的AT2R可调控表达受到Dox的良好调控，可有效抑制球囊损伤后大鼠颈动脉新生内膜增生；(5)AT2R影响血管损伤后修复时ECM成分的表达，并促进细胞凋亡的发生，可能是AT2R介导抑制新生内膜形成的机制。

①Rat MSC and VSMC were cultured and identified, respectively. MSC were labeled with DAPI firstly, and then co-cultivated with VSMC. The changes of morphology and ultrastructure of co-cultured cells were observed. Immunfluorescence analysis was performed by using monoclonal antibodies against specific antigen.②We established the regulatable system in two steps: a stable MSC line expressing rtTA has been constructed and characterized firstly by transfected with pUHD 17-1hyg and then selected by hygromycin B; in a second step, this line was used for trandfer the AT2R gene to MSC to get the well establishing double stable MSC lines;③The expression of AT2R regulated by doxycycline was evaluated by western blot;④The MSCs were transduced into rat carotid arteries with regulatable AT2R gene after the establishment of rat carotid balloon injury restenosis model. The intimal/medial area ratio were measured by digital analysis system.

研究方法：(1)密度梯度离心法及胶原酶消化法分别培养原代大鼠MSC及VSMC，细胞共培养并行免疫荧光化学染色和透射电镜观察超微结构；(2)组成受Dox调控的哺乳动物表达系统的四种成分的转化、扩增及提纯并酶切鉴定；(3)采用常规分子生物学方法连续两个回合转染体外培养的MSC，并分别采用发光计检测不同细胞克隆萤光素酶活性改变以及RT-PCR方法检测AT2R目的基因mRNA表达情况，根据各个细胞克隆受Dox调控表达的程度，选择低背景、高诱导表达AT2R的细胞系，作为双重稳定MSC细胞系；蛋白免疫印迹法观察该细胞系在Dox调控下AT2R表达的时相性、持续性及在不同浓度Dox调控下的表达情况；(4)建立大鼠颈动脉球囊损伤动物模型，将双重稳定MSC在术中导入血管，分别于14 d、28 d进行病理切片，检测可调控AT2R对新生内膜增生的影响；采用RT-PCR免疫组织化学免疫荧光等技术观察AT2R基因在新生内膜中的表达以及细胞外基质成分表达的改变，TUNEL法检测血管组织中细胞凋亡的变化情况。

Shed blood reinfusion alone did not restore the decreased vascular reactivity. ICI174,864 improved the decreased vascular reactivity in dose-dependent manner.

单纯回输失血不能纠正血管的低反应性，ICI174，864对休克血管的低反应性有不同程度的恢复作用，且呈一定的剂量依赖关系。

Results Following hemorrhagic shock, the systemic and local vascular responsiveness was significantly decreased significantly and it was time dependent. Shed blood reinfusion alone did not restore the decreased vascular reactivity.

结果失血性休克后全身（血压对 NE的升压反应）和局部（SMA对NE的收缩反应）血管反应性显著降低，且呈一定的程度和时间依赖性，即休克程度越重，时间越长，血管反应性降低越多。

Results 45 infarct related arteries among 40 cases acquired coronary artery repatency with TIMI 3 degree flow, a success rate of 95.2%(40/42), the residual stenosis in repatency coronary arteries was from 0 to 20%. The clinic symptom of the 5 cases with left ventricular dysfunction and 2 of the 3 cases with cardiogenic shock were obviously improved, 1 case(2.5%) with cardiogenic shock was dead in hospital.

结果 40例患者中的45支梗塞相关血管获得再通，成功率达95.2%(40/42)，均达到TIMI-3级血流灌注，再通血管残余狭窄0%～20%。5例合并左心功能不全(Killip分级Ⅱ～Ⅲ级)及3例合并心源性休克患者术后症状明显改善，住院期间1例死亡(病死率2.5%)。

To the clinical medicine, it is a great development that the acute myocar- dial infarct patients have been cured by coronary artery repatency, thromboclasis, precutaneous transluminal coronary angioplasty and other heart surgery in the last years. The myocardial tissue receives blood reperfusion again due to cardiovascular repatency or reconstructive operation, which can really obtain favorable therapeutic efficacy in multitudinous conditions.

方法随机选择160例近22年来应用较广的5种动脉跨区供血反流轴型皮瓣，即：颞浅血管为蒂的乳突区皮瓣、眶上血管为蒂的岛状耳廓复合组织瓣、颏下动脉蒂的对侧颏颈皮瓣、颈横动脉供血的项肩背皮瓣、会阴动脉供血的股内侧皮瓣，考查其完全成活率、部分成活率、完全坏死率、动脉缺血发生率、动脉缺血致皮瓣坏死率、静脉淤血发生率、静脉淤血致皮瓣坏死率等指标，并与我院同期内应用的常规顺流轴型皮瓣随机样本120例进行比较。

The trophoblast blood flow spectrum could be easily detected with the guide of CDE.The ultrasonic findings were characterized by broad arterial flow spectrum,high diastolic component of the low resistance trophoblastic flow,RI＝0.40～0.51 with single or biphasic spectrum.The salpingian ring with high blood flow indicated early and complete salpingian pregnancy.

结果：①异位妊娠包块血流信号丰富，血管连续性好，能较完整显示血管床、血管网及范围广的低流速血流信号；②脉冲多普勒取样为滋养层周围性血流频谱，其特征为：频谱增宽、单相或双相持续存在、阻力降低(RI＝0.44±0.04)、舒张期血流丰富的动脉频谱；③具有鲜明丰富的滋养层周围性彩色血流频谱特征的&输卵管环&是早期输卵管妊娠的特征性表现，并提示是输卵管完整存在的标志。
