血清酶

A sandwich ELISA was established taking the antiserum as the primary antibody. The sensitivity of the ELISA was 0.412 ng GH/ml of sample. Inter-assay and intro-assay coefficients of variation were 2. 58％(n=6) and 5. 69％(n=6) respectively. Serial dilutions of serum and pituitary homogenate from orange-spotted grouper yielded dose response curves that were parallel to the standard curve. And the GH of Acanthopagrus butcheri was also parallel to the standard curve. Prolactin of goldfish and recombinant growth hormone of common carp showed no cross-reactivity with the antiserum of orange-spotted grouper GH. It showed that the established RIA was specific and sensitive.

以斜带石斑鱼重组GH抗血清为第一抗体，羊抗兔抗体为第二抗体建立了双抗体夹心酶联免疫测定法，该系统的灵敏度为0.412ng GH／ml(n=6)，组内变异系数为2.58％(n=6)，组间变异系数为5.69％(n=6)，斜带石斑鱼血清和脑垂体稀释液曲线与斜带石斑鱼重组GH蛋白的标准曲线相平行、黑棘鲷GH的系列稀释曲线与标准曲线相平行，金鱼催乳素、重组鲤鱼GH的系列稀释曲线与标准曲线不平行，表明建立的斜带石斑鱼GH ELISA具有良好的特异性和重复性，灵敏度达到了测试血清样品中GH的水平。

Methods Blood samples were collected from the precaval vein of 6 female Tibet mini-pigs to test the concentration of sex hormones including T,P4,E2,FSH,LH,PRL and blood biochemical indexes including Ca, Mg, P, ALP, Cre, CHO, TP, and Alb during the estrous cycle.

对6头雌性西藏小型猪在发情周期内的发情当天、第5天、第10天、第15天、再次发情时分别采血5 mL并分离血清，应用放射免疫法测定血清睾酮、孕酮(P4)、雌二醇(E2)、促卵泡生成激素、促黄体生成激素、泌乳素；用自动生化仪测定血清钙、镁、磷、碱性磷酸酶、肌苷、总胆固醇、总蛋白、白蛋白。

Results scrub typhus complicated with hepatic lesions occurred to 73.5%(36/49) of patients, gastrointestinnal manifestations including nausea and vomit occurre to 33.3% of the patients (12/36,) and abdominal distention(21/36,58.3%).elevated serum alanine aminotransferase and aspartate aminotransferase were observed in allthe 36 cases. of which 9 cases(25.0%) had elevated serum total bilirubin,16 cases (44.4%) had elevated alkaline phosphatase and 14 cases (38.9%) had decreased serum albumin.

结果 36例恙虫病合并肝损害患者占同期恙虫病患者的73.5%，其中胃肠道症状有恶心呕吐12例，腹胀21例；36例患者血清alt及ast均升高，其中血清tbil升高9例，血清碱性磷酸酶升高16例，alb降低14例；b型超声检查肝脏肿大13例，脾脏肿大16例，有腹水者9例。

Results It showed that the serum active enzymatic of 5′-NT in hepatic and gall diseases,osteal diseases levels were increase, especially the more significant increase of the serum active enzymatic of 5′-NT in level appeared in the patients with hepatic and gall diseases,osteal diseases .

目的 检测各型肝胆疾病、骨骼疾病患者血清5′-核苷酸酶(5′-NT)，谷丙转氨酶和γ-谷氨酰转肽酶，碱性磷酸酶酶活性的变化，观察在肝胆疾病和骨骼疾病中的临床意义，同时与正常健康者作对照。

In this research, B2 gene of HCV was highly expressed as a fusion protein (Trx-B2) by cloning into pThioHisA, and induced specific anti-HCV antibody in higher titer than unmodified B2 protein after immunize mice or rabbit.The results showed antigen Trx-B2 has obvious immunogenicity and induced specific antibody,which might be able to serve as diagnosis HCV. Consequently, this PcAb of rabbit was used to coated Immulon plate, and a method of both antibodys sandwiching antigens was developed to detect 32 HCV positive serum and 32 nomal serum.Results show that ratio of positive are 87.5%and 43.75% respectively.

但缺点是该抗原所诱导的抗体滴度较低，可能是由于其分子量较小，所以本课题将人工合成的HCV复合多表位抗原基因B_2克隆到表达载体pThioHisA中，利用硫氧还蛋白融合方式对该抗原进行修饰，并在大肠杆菌中表达融合蛋白(Trx-B_2)，纯化该蛋白后免疫小鼠及家兔，结果表明用硫氧还蛋白修饰后的融合蛋白免疫小鼠和家兔后，得到的抗体滴度均高于未经修饰过的复合多表位抗原合成肽所诱导的抗体滴度，并从免疫后的兔血清中提取IgG包被酶联板，利用双抗夹心法分别检测了32份HCV阳性血清和32份正常人的血清，结果阳性率分别为87.5%和43.75%，符合率为71.88%。
