血清疫苗

The dynamic variety of specific antibodies in sera and saliva were checked by ELISA.

Results ①DNA sequencing showed that pBX1 contained a 477bp inserted gene fragment,and when it was compared with the published sequence of the specific region of the gene of the 83kd antigen protein from Borrelia burgdorferi B31 strain,only one amino acid codon was different.

结论该研究成功地对莱姆病螺旋体83kd抗原蛋白特异性区段进行了基因重组和表达，为进一步研究其在莱姆病血清学诊断和基因工程亚单位疫苗研制中的应用奠定了基础。

Methods The SpaA was purified by electroelution from NaOH-extracted antigen of Erysipelothrix rhusiopathiae strain C43311. The rSpaA-N was expressed in E. coli BL21 as a soluble protein by IPTG inducing,and purified with GST affinity chromatography. Mice of each group were subcutaneously immunized three times with 50μg or 100 μg of native SpaA, rSpaA-N or NaOH-extracted antigen with in complete or incomplete Freund adjuvant at 2-week intervals.

将猪丹毒丝菌C43311株SpaA-N以可溶形式表达在大肠杆菌BL21中，用GST Bind Resin纯化试剂盒纯化rSpaA-N，采用电洗脱法从猪丹毒丝菌C43311株NaOH提取抗原中纯化天然SpaA，将rSpaA-N、天然SpaA和NaOH提取抗原制成亚单位疫苗，同时设GST及生理盐水对照组，间隔2周分3次皮下免疫小鼠，第3次免疫后2周用100LD50猪丹毒丝菌C43065株进行腹腔攻毒，采用间接ELISA方法检测免疫组小鼠血清的抗体动态变化。

Nucleoprotein is also a major immunogenetic antigen for SARS; however, several tests have shown that N protein had the reactivity with other coronavirus positive sera. It is therefore an advantage of S protein as recombinant target antigen for SARS serum diagnosis compared with recombinant N protein. Moreover, S protein has been shown to be the major antigenic determinant that induces neutralizing antibodies and protective immunity to SARS-CoV, so it is also important in designing SARS genetic engineering vaccine.

SARS-CoV S 蛋白由1255 个氨基酸组成，体外表达如此大的蛋白很困难，而且表达蛋白的可溶性不高；其次，实验已经证实，全长S 蛋白含有大量无关的表位，致使表达的全长S蛋白对SARS 阳性血清的检出率低于S 蛋白片段；另外作为疫苗研究，S 蛋白上大量无关表位作为异体蛋白进入体内，会引发机体的过度免疫反应，这也是SARS-CoV 可能的致病机理之一。

Considering, it is limited the cross-reactivity displayed by a single HVR peptide. We designed to ligate several high reactive HVR1 genes, and the recombinant fused protein containing several HVRls would have the ability to cross-react with most of HCV infected sera, and could be an effective immunogen of HCV vaccine.

Methods104 selected volunteers aged from 10 to 65 year old were vaccinated with the domestic product and the import rabies vaccine.Serum antibody levels of three blood samples of the pre-vaccination,the 14th and the 45th days after vaccination were determined with mice neutrodyne test.

方法使用国产和进口无佐剂人用狂犬病疫苗，全程免疫104名10～65岁入选志愿者，在免前、免疫后d 14和d 45抽血，采用小鼠中和试验检测血清抗体水平。
