血清疫苗

Rabbits were inoculated with formaldehyde-inactivated bacteria suspensions of Actinobacillus pleuropneumoniaestrains of 12 serotypes with bee glue as an adjuvant.to prepare a set of monospecific antisera against each serotypes of APR.

将猪传染性胸膜肺炎放线杆菌(Actinobacillus pleuropneumoniae APP)全部12个血清型的国际参考菌株接种培养基大量培养，回收菌体、甲醛灭活后加入蜂胶佐剂制成疫苗，分别免疫健康的试验用白兔，获得了针对APP菌单一血清型的抗血清。

The results showed that the sera samples got from the pigs immunized with Hog cholera haplized virus using bursin as adjuvant have higher antibody titers than those without using adjuvant according to the criterion established here, which is conform to the previous study. This study lay on foundation for standarding the I-ELISA assay established here.

以此方法建立的间接ELISA方法对收集的约120头份血清样品进行检测，结果显示：使用囊素与疫苗共同免疫猪的血清抗体水平明显高于未加囊素组猪的血清抗体水平，与以往的报道一致，为以CSFV重组蛋白作为抗原建立的间接ELISA方法的标准化奠定基础。

Part Ⅱ Objective: To study the immunoprotective and the immunotherapic effects of the new nucleic acid vaccine of the hepatitis B: pVAX1-C3d-S2S; respectively in normal and HBV transgenic mice after being inoculated intra- muscularly with this Eukaryotic Expression Plasmid, such as the in situ production of the expressed protein in muscle tissue section and specific humoral immune response in normal mice、the variation of specific antigen、antibody and HBV DNA, especialy the changes of specific antigen in liver.

第二部分新型乙肝核酸疫苗pVAX1 C3d-S〓S免疫预防效应和治疗作用的研究目的：研究含有新型分子佐剂补体C3d的HBV adw血清型的乙肝核酸疫苗重组真核表达质粒pVAX1-C3d-S〓S接种正常BALB／c小鼠和HBV转基因小鼠后，其免疫预防作用（接种局部肌肉中特异性抗原蛋白的表达和所诱生的体液免疫应答规律）以及免疫治疗作用（血清中特异性抗原的表达、特异性抗体的产生、HBV DNA的消长和肝组织中抗原表达的变化）。

To evaluate the effect of PRRSV on humoral and cellular immune responses,threeweek old SPF piglets and conventional piglets with antibody to PRRSV were inoculated nasally with PRRSV BJ 4 strain,and immunized intramuscularly with hog cholera vaccine 48 hours postinoculation.

对 2 0日龄 SPF猪和 2 0日龄猪繁殖与呼吸综合征血清阳性猪，人工感染猪繁殖与呼吸综合征病毒北京分离株( BJ-4 )后 4 8h接种猪瘟疫苗，利用 ELISA方法检测仔猪针对 PRRSV和猪瘟疫苗的体液免疫，利用 MTS法检测仔猪外周血单核细胞对有丝分裂原 Con A的刺激反应。

We used Aeromonas hydrophila which we separated from the diseased Paralichthys olivaceus to made a inactivated whole cell bacterin .We injected this vaccine in the mice and the Paralichthys olivaceus .Both the mice and the Paralichthys olivaceus gained a good protection of 80% and 70%.The fish with dipping immersion gained a good protection of 85%,and the incidence of disease was 6%.We detected the antibody level in the antiserum by agglutination reaction and indirect ELISA which seemed to be immunogenic crucially.We anatomized the mice then we found that all the purtenance of the immunzied mice are healty.All of this proved that the vaccine is effective.

本实验将所分离到的嗜水气单胞菌用终浓度为0.2%~0.5%的甲醛对菌体进行灭活，制备了灭活疫苗，并优化灭活疫苗的灭活条件；将制备的灭活疫苗分别注射小鼠和牙鲆，均取得了较为理想的效果，其中小鼠的免疫保护率达到80%，牙鲆的免疫保护率达到70%；采用渗透压差法大规模浸泡免疫牙鲆也取得了很好的效果，浸泡免疫过的牙鲆发病率仅为6%，攻毒后免疫保护率为85%；通过应用间接ELISA方法检测受免动物的抗血清，并将牙鲆的抗血清注射新西兰白兔，从而获得兔抗鱼血清，更便于检测，结果都表明动物体内产生了较高的抗体水平；剖检攻毒后的动物，发现免疫过的动物内脏健康无异样，表明该疫苗安全有效，从而为防治牙鲆腹水症开辟了一条新途径，也为进一步制备其基因工程疫苗奠定了基础。

objective to study the efficacy of the tr/patoc 1 slide agglutination assay for vaccination new pentavalent whole cell vaccine for leptospirosis.methods using tr/patoc 1 slide agglutination assay to detect the searial of serum specimens of 100 healthy people after vaccination for leptospirosis and comparing with the microsopic agglutination test.results both basic immunizaton and 20 days after the enhanced immunization,their positive seroconversion rates were higher than 90% in tr/patoc 1 slide agglutination assay,they were not signficantly different between slide agglutination assay and mat,the antibody titer was greatly lower in 90 days after basic immunization.conclusion the tr/patoc 1 slide agglutionation assay was one of useful assays in evaluation the lately immune efficency of leptospira vaccination.

目的 采用tr/patoc 1玻片凝集试验评价钩端螺旋体疫苗免疫效果及应有价值。方法采用tr/patoc 1玻片凝集试验检测100名健康人钩端螺旋体疫苗免疫后的血清抗体变化，并与显微镜凝集试验比较。结果无论是基础免疫还是加强免疫20？d后，玻凝试验抗体阳性率高达90%以上，与标准的显微镜凝集试验比较差异无统计学意义，玻片凝集试验检测的抗体持续时间较短，90？d后阳性率已大幅度降低，表明sat检测的抗体是早期抗体。结论 tr/patoc 1玻片凝集试验操作简易，可以作为基层卫生机构监测钩端螺旋体疫苗近期免疫效果的方法之一。

BALB/c mice were vaccinated with this DNA vaccines.The mice injected withBCG were positive control and the mice injected with pCDNA3.1 and PBS were negative control.The mice were immunized 3 times with 2-wk intervals. The animals in group BCG were only inoculatedsubcutaneously with 1×10~6 CFU BCG at initial vaccination. The serum IgG titers and IgG isotype weredetermined using iELISA coated with M. bovis PPD and rMAE protein expressed and depurated inprokaryotic expression system every week.
