In order to establish a sensitive immuno-PCR assay for detecting serai anti-P53antibodies, investigate the relation of anti-P53 antibodies in patients with breast carcinoma to P53 protein expressions in tissue and clinical significances and afford a practical tool in diagnosis of breast carcinoma clinically. SaHIgG was coupled to PNCa DNA by chemical cross-linker PEI and SaHIgG-DNA probe was constructed.

本研究建立了血清抗P53蛋白抗体免疫聚合酶链反应(Immuno-polymerase chain reaction，免疫PCR)检测方法，探讨了乳腺癌患者血清中抗P53蛋白抗体与组织中P53蛋白表达之间的关系及临床意义，旨在为临床乳腺癌P53蛋白检测提供实用的替代工具。

The serum samples were detected by Indirect ELISA and Indirect hemagglutination test as reference method to assess the value of the recombinant protein in serodiagnosis.

结论1、大量表达并纯化了重组蛋白SjRPS4，并证实其具有良好的免疫反应性，能与日本血吸虫病阳性血清特异性结合，可应用于日本血吸虫感染血清学诊断。2、成功建立了以重组蛋白SjRPS4为包被抗原的间接ELISA法，在日本血吸虫病的免疫诊断中具有良好的应用价值。

Methods Fifty-three patients diagnosed as chronic hapatitis of C were treated by IFN and Ribavirin for at least 24 weeks. Serum HCV-RNA was detected by fluorescent quantitative PCR. Genotype was determined by Simmonds. Results There was no difference of HCV-RNA levels in different genotypes, as that of ApoB.

临床确诊为慢性丙型肝炎的53例患者，采用干扰素联合利巴韦林抗病毒治疗至少24周，Simmonds酶切分型方法进行HCV基因分型，荧光定量聚合酶链反应法定量检测HCV-RNA，全自动生化分析仪检测血清载脂蛋白B，对不同基因型患者血清HCV-RNA水平和载脂蛋白B的关系进行研究分析。

Objective:in order to compare the curative effects of using lamirudine and thymosin together with using thymosin only to the paients of chronic hepatitis b.methods:we had chosen 27 medical cases of chronic hepatitis b,which were divided into two groups:15 cases asone group,and 12 cases as the other group.let group a take 100 mglamirudine orally,qd,with half a year as a period of treatment.at the same time givethe patients injections of 100 mg thymosin,qd,for a month.to group b was only takenlamirudine.alt,tbil,the indicators of the virosis of hepatitis b (hbsag,hbeag,hbv-dna)were observed as quatos.results: at the end of the treatment, the rates that groupa and group b made hbsag transform into negative were respectively 26.67% and 16.67%(p＞0.05);hbeag:33.33%,8.33%;hbv-dna:53.33%,33.33%(p＞0.05).conclusion:combining thymic peptide with 3tc to treat chronic type b hepatitis, thymic peptide will not only promote b-cell to mature and transformation, but also to keep the subpopulation of t-cell in immune system in balance, which can enhance the reaction of matured t-cell to the antigen or other stimulus.

目的：比较拉米夫定（3tc）与胸腺肽联用和单用拉米夫定治疗慢性乙型肝炎的疗效。方法：选择慢性乙型肝炎患者27例，分为联合组15例和拉米夫定组12例；联合组用拉米夫定100 mg，口服，qd，疗程半年，合并用胸腺肽100 mg，静滴，qd，疗程1个月。拉米夫定组单用拉米夫定。以谷丙转氨酶、血清胆红素、乙肝病毒标志物(hbsag，hbeag，hbv-dna)为观察指标，比较不同治疗方案对指标的影响。结果：治疗结束时联合组与拉米夫定组对慢性乙肝患者血清hbsag阴转率分别是26.67％和16.67％(p＞0.05)；hbeag阴转率分别是33.33％和8.33％(p＞0.05)；hbv-dna阴转率分别是53.33％和33.33％(p＞0.05)。结论：胸腺肽联合3tc治疗慢乙肝，胸腺肽具有促进t淋巴细胞的成熟和转化，并维持免疫系统各t淋巴细胞亚群间的平衡作用，能增强成熟t淋巴细胞对抗原或其他刺激的反应。

After thecomplete genome extraction of the strain was performed, the genomic DNA was partiallydigested by restriction enzyme Sau3AⅠ, the DNA fragments from 1 to 5Kb was clonedinto prokaryote expression vector pET-28a-c, and transformed host bacteria. The resultsshowed that we succeeded in constructing the gene expression library of haemophilusparasuis serovar 5, which is fundamental for the study of advanced gene screening. Inaddition, primer design was performed based on haemophilus influenzae in this study. In addition, PCR was performed by using genomic DNA of haemophilus parasuisserovar 5 as the template. The results demonstrated that we obtained two neo-gene:23SrRNA gene(conserved gene belonging to the large-subunit of ribosome) and adenylatecyclase gene(encodes adenylate cyclase and participates in converting adenyl nucleosidetriphosphate to cyclic adenosine3",5"-monophosphate). Furthermore, the phylogeneticanalyses between the species was performed, and neighbor-joining tree was constructedbased on comparison of 23S rRNA gene sequences, so it was illuminated betweenHaemophilus parasuis and other species in molecular evolution relationship.

选择我国流行优势菌株副猪嗜血杆菌血清5型地方株为研究对象，提取细菌基因组DNA，用限制性内切酶Sau3AⅠ对基因组DNA进行部分酶切，回收大小为1～5Kb的DNA片段，将其连接入原核表达载体pET-28a-c，最后转化宿主菌，结果成功地构建了基因表达文库，为后续的基因筛选工作奠定基础；另外，本研究选择嗜血杆菌属的流感嗜血杆菌为参考对象进行引物的设计，以副猪嗜血杆菌血清5型地方菌株的基因组DNA为模板，进行PCR扩增反应，结果表明成功地获得两个新基因：23S rRNA基因(存在于核糖体大亚基中的保守性基因)和腺苷酸环化酶基因(负责将腺嘌呤核苷三磷酸转变为环腺苷酸)，并进一步做了不同物种之间的分子系统发育分析，构建了基于23S rRNA基因的邻接法系统发育树，阐明了副猪嗜血杆菌与其它菌种的分子进化关系。

Result150of patients and infected sheep,Cattle,Swine serum positive rates of DAgS -ELISA were 91.4%,74.6%,and 66.7%re spective-ly. In the main, DAgS -ELISA gain adva ntage over of SAT and RBPT.

结果上述3种试验对健康人、羊、牛、猪共计140份血清检查均为阴性反应，用DAgS-ELISA对布氏菌感染人、羊、牛、猪共计150份血清检查，其阳性率分别为91.4%、74.6%、66.7%及66.7%，从总体而言，检查的特异性及敏感性，以DAgS-ELISA优于SAT及RBPT。
