The experiment with 9 treatments including 3 growth stages: from wheat seedling to jointing, from wheat jointing to flowering and from wheat flowering to maturity and 3 water gradients: the capacity of water in field from 30% to 40%, from 50% to 60% and from 70% to 80% was designed and the content of protein and nucleic acid and the osmotic potential of winter wheat leaf were measured for the research on drought stress affecting them.

方法］将小麦生育期划分为出苗至拔节、拔节至开花、开花至成熟3个阶段，同时设3个水分梯度，即田间持水量的30%~40%、50%~60%、70%~80%，共9个处理，测定冬小麦叶片的蛋白质、核酸含量和叶片渗透势，分析干旱胁迫对不同生育期冬小麦叶片蛋白质和核酸含量的影响。

The metabolism of protein and nitrate nitrogen in tobacco leaf during curing was studied by using the electric-heated flue-curing barn designed and made by Henan Agricultural University in which the temperature and moisture were controlled automatically.The results indicated that the content of protein decreased gradually while the content of amino acid increased with the proceeding of curing.And either of them had a rapid-changing stage in the middle of yellowing period and color-fixing period.The activity of proteinase reached the peak respectively after curing 24 hours and 60 hours.

The total proteins from TGIF transfectant and vector control cells were separated by 2D electrophorosis. 760±41 and 680±38 spots were detected in TGIF transfectant and vector control cells respectively. Locus repeat analysis of protein spots showed that the mean deviation in IEF direction in TGIF transfectant and vector control cells were 0.864±0.123 mm and 0.843±0.115 mm respectively whereas the mean deviation in SDS-PAGE direction in these two cells were 0.812±0.109 mm and 1. 125±0.123 mm respectively. So the well-resolved and reproducible 2DE patterns from TGIF transfectant and vector control cells were established.

通过双向凝胶电泳分别分离TGIF转染细胞和PcDNA3.1空白质粒转染细胞的总蛋白质，测得两组细胞的蛋白质斑点数分别为760±41和680±38个；位置重复性分析表明TGIF转染细胞在等电聚焦方向上的平均偏差为0.864±0.123mm，在垂直板SDS-PAGE电泳方向上的平均位置偏差为0.812±0.109mm；PcDNA3.1空白质粒转染细胞在IEF方向上的平均偏差为0.843±0.115mm，在垂直板SDS-PAGE电泳方向上的平均位置偏差为1.125±0.123mm。

Results Well-resolved, reproducible 2-DE profiles of human colon carcinoma tissues and paired normal tumor-adjacent colon tissues were obtained. For colon carcinoma tissues, a total of (980±28) spots were detected; for paired normal tumor-adjacent colon tissues,(860±45) spots were detected. For normal tumor- adjacent colon tissues, the average deviation of spot position was (10.487±0.11) mm in IEF direction and (0.944±0.12) mm in SDS-PGE direction; for colon carcinoma tissues the average deviation of spot position was (0.654±0.11) mm in IEF direction and (1.20±0.22) mm in SDS-PGE direction. Number of differentially expressed proteins was 72.00±12.34 between colon carcinoma and paired normal tumor-adjacent colon tissues.

结果 建立了结肠癌及正常肠组织的双向凝胶电泳图谱，其中癌旁正常结肠组织和结肠癌组织电泳图谱中平均蛋白质点数分别为860±45和980±28；癌旁正常结肠组织和结肠癌组在IEF方向上的平均偏差分别为(0.487±0.11)mm和(0.654±11)mm，在SDS-PAGE方向上的平均偏差分别为(0.944±0.12)mm和(1.20±0.22)mm，结肠癌与癌旁正常结肠组织的差异表达蛋白质点数为72.00±12.34。
