Terminal oxidative inhibitors, such as KCN, Salicylhydroxamic acid , antimycin A and NaN〓, can cut off electron transport chains of cytochrome pathway and alternative pathway, marked decreased BADH protein levels. Oxidative phosphorylation uncoupler 2, 4-dinitrophenol had a similar negative effect on BADH gene expression, and exogenous ATP and ADP increased the BADH levels. These results, in conjunction with the terminal oxidation results, indicated that ATP could be a critical component in BADH gene expression.

2.2 末端氧化抑制剂KCN、SHAM、抗霉素A和NaN〓能打断呼吸链，阻断电子传递，使BADH蛋白的表达量明显降低，说明末端氧化同样参与BADH基因表达的调节：末端氧化影响基因表达的实质就是ATP的影响，线粒体磷酸化解偶联剂DNP明显降低BADH蛋白的含量，而外源的ATP和ADP对BADH基因表达的正效应作用最大。

The mc2155 carrying pMind-glmU-AS was induced by different concentration of tetracycline, and GlmU protein was detected in the mc2155 carrying pMind-glmU-AS by Western blotting. The results showed that tetracycline of 20ng/ml has obviously inhibited the growth rate of mc2155 carrying pMind-glmU-AS, but the amount of GlmU protein did not change compared to that of uninduced mc2155 carrying pMind-glmU-AS.

glmU反义RNA的诱导表达与GlmU蛋白的检测分别用5 ng/ml、10 ng/ml和20 ng/ml的四环素诱导携带pMind-glmU-AS表达载体的mc~2155菌株表达反义RNA，绘制细菌生长曲线，结果表明20 ng/ml浓度的四环素可抑制mc~2155/pMind-glmU-AS的生长，但Western Blotting分析结果显示经四环素诱导和未诱导的mc~2155/pMind-glmU-AS中，GlmU蛋白表达量无明显的变化。

Combined with the test of All〓wden reaction and observation with SEM, the data have proved that protease SZ can digest the protein of epicuticle and then hydrolyzed the cuticle of wool, while the other proteases have no this distinctive effects, although they can confer wool bigger weight loss too. As protease SZ can digest the epicuticle and cuticle, the wool can be confered excellent antifelting property with low tenacity and elongation loss.

The present study is involved in the expression of vitronectin , a ligand of integrin, on human spermatozoa and its role in fertilization; the expression of integrin α〓、α〓、β〓 sbunits on human and mouse oocytes and their relationship with maturation of oocytes; the reorganization of cytoskeletal protein, actin, of mouse oocytes mediated with RGD peptide binding with integrins; the roles and mechanisms of integrins in activation of mouse oocytes as trasmembrane signaling receptors, using the methos of cell culture, immunohistochemistry, flow cytometer, laser confocal microscope.

Our results revealed that TGF-β1 at 1 ng/ml induced collagen deposition of HSC-T6 cells (145 ± 5 % of controls), and pre-exposure of cells to curcumin (1.25~10 μM) inhibited the stimulatory effect of TGF-β1 in a concentration-dependent manner. Curcumin (10 μM) significantly reduced the TGF-β1-induced collagen deposition (97 ± 7 % of controls) without cytotoxicity in the MTT assay .

我们的结果显示，1 ng/ml 浓度的TGF-β1可以有效地增加星状细胞对胶原蛋白的制造(达控制组的145 ± 5 %)，而在预先投予浓度范围为1.25~10 μM的姜黄素后可对TGF-β1引起的刺激反应产生浓度效应的抑制作用；姜黄素在浓度10 μM下可明显地抑制TGF-β1引起的胶原蛋白沉积(达控制组的97 ± 7 %)而不致产生细胞毒性。

Main works listed below as:1、Preparation and identification of polysaccharide antigen of L. pneumophila Bacteria of LP1 ~ 7,9 and 10 were cultured on yeast agar buffer activated carbon for three to five days at 37℃, 5%CO2. Protein-free polysaccharide antigens were obtained after harvest in cells, extraction, deproteinization, dialysis and other steps. Their immunogenicities were verified by ultraviolet spectrophotometer full-wavelength scanning and Western blotting.2、Preparation and identification of rabbit anti-LP1 antibodies and rabbit anti-LP5 antibody Rabbit anti-LP1 and anti-LP5 antibodies were purified after rabbits were immuned with antigens isolated as described above. The purities of both antibodies were above 80% and the titer of blood serum 1:32 tested by double antibody sandwich assay.3、Development of colloidal gold immunochromatographic assay kit The size of colloidal gold particles in the kit was 25nm. The optimal concentrations for antibodies were 30μg/ml and the sensitized concentrations of NC membrane were 5 mg/ml.

主要研究工作从以下几个方面进行：1、LP1~7、9和10型多糖抗原的制备与鉴定将LP1~7、9和10型菌株分别接种在缓冲活性炭酵母琼脂培养基上，37℃、5%CO2的条件下培养，3~5天后洗下菌苔，经抽提、除蛋白、透析等步骤后得到基本无蛋白的LP多糖抗原，经紫外分光光度仪全波长扫描及Western blotting验证其抗原良好。2、兔抗LP1抗体和兔抗LP5抗体的制备与鉴定分别用LP1、LP5型多糖抗原免疫家兔，采用琼脂糖双向扩散试验检测，两种抗体血清效价均为1：32；饱和硫酸铵法提取抗体，SDS-PAGE检定其抗体纯度均达到80%以上。3、胶体金免疫层析检测试剂的初步研制采用柠檬酸三钠还原法制备约25nm大小的胶体金颗粒；分别制备兔抗LP1抗体、兔抗LP5抗体的金标探针，两种抗体的最适标记量均为30μg/ml；选择适当孔径的微孔滤膜为载体包被两种抗体，NC膜包被浓度均为5mg/ml。
