Twenty-seven differential proteins were tested by MALDI-TOF-MS, and 16 were identified. Of the 16 proteins, 13 were up-regulated in patients with HCC, including heat-shock protein 27, member RAS oncogene family (RAB14), actin, alpha 1 antitrypsin, RNA-binding protein regulatory subunit and so on. By contrast, the levels of proteins such as PDX6, HSPA8 and MnSOD were significantly reduced in HCC patients.

对27个差异蛋白质点进行了质谱分析，鉴定出16个蛋白质，在HBV相关性HCC患者PBMC中表达明显增强的有热休克蛋白质27、ras癌基因家族(RAB14)、肌动蛋白、α1-抗胰蛋白及RNA结合蛋白调节亚基等13个蛋白点；而PDX6、HSPA8及锰超氧化物歧化酶在HCC患者PBMC中低表达。

RESULTS: A well-resolved and reproducible 2-DE pattern of PBMC was obtained from patients with HBV associated HCC or LC. For HCC, the mean number of protein spots was 1186±43, with an average matching rate of 91.6%, and for LC, the mean number of protein spots was 1013±41, with an average matching rate of 90.2%. Twenty-seven differential proteins were tested by MALDI-TOFMS, and 16 were identified. Of the 16 proteins, 13 were up-regulated in patients with HCC, including heat-shock protein 27, member RAS oncogene family (RAB14), actin, alpha 1 antitrypsin, RNAbinding protein regulatory subunit and so on. 结果：得到了分辨率较高、重复性较好的HBV相关性HCC及LC患者PBMC双向凝胶电泳图谱，HBV相关性HCC及LC患者PBMC凝胶的平均蛋白质点数分别为1186±43及1013±41；组内的平均匹配率分别为91.6%，90.20%对27个差异蛋白质点进行了质谱分析，鉴定出16个蛋白质，在HBV相关性HCC患者PBMC中表达明显增强的有热休克蛋白质27、ras癌基因家族(RAB14)、肌动蛋白、α1-抗胰蛋白及RNA结合蛋白调节亚基等13个蛋白点；而PDX6、HSPA8及锰超氧化物歧化酶在HCC患者PBMC中低表达。

The Cry1Ac5 domainⅢhas a distant relationship with other Cry1 proteins and its longβ18-β19 loop possesses a unique tryptophanat residue 544 and a unique asparagineat the loop apex(residue 546),as well as two consecutive serinesfrom residue 547 to 548.Alanine scanning mutations within the loop were initially generated and all mutants could produce bipyramidal crystals and express 130 kDa protoxins.

利用Swiss-Model服务器和Swiss-pdb Viewer软件建立了Cry1Ac5蛋白的三维结构模型，并利用Clustal W和DS ViewerPro软件对比分析了其与其它Cry蛋白之间的差异，发现Cry1Ac5的结构域Ⅲ与其它Cry1蛋白的亲缘关系较远，其结构域Ⅲ上的β18-β19 loop比Cry1Aa、Cry2Aa、Cry3Aa和Cry4Aa蛋白对应的loop长，其组成的残基中色氨酸(W544)、连续的丝氨酸(S547、S548)和loop顶点的天冬酰氨(N546)在Cry蛋白中都是独特的残基分子成分。

Results showed:(1) The airway responsiveness of smoke control group was higher than that of normal control group, the airway responsiveness of smoke + exercise group was lower than that of smoke control group;(2) Plasma level of cortisol determined immediately after exercise was higher than that determined before exercise, no significant difference occurs between that determined next day morning after all exercise finished in contrast to that determined before exercise;(3) HE staining showed, there was severe chronic pulmonary inflammatory response in smoke control group, which was reduced in the smoke + exercise group;(4) chronic cigarette smoke downregulated the protein and mRNA expression of BKca in smooth muscles of bronchi and bronchioli, exercise increased the mRNA expression of BKca;(5) chronic cigarette smoke downregulated the protein and mRNA expression of Kv1. 5 in smooth muscles of bronchi and bronchioli, exercise increased both the protein and mRNA expression of Kv1. 5 in contrast to smoke control group in bronchioli and did not effect the expression of Kv1. 5 in bronchi;(6) chronic cigarette smoke induced upregulation of FIZZ1/RELMα in bronchial smooth muscle cells, exercise decreased these effect.

结果：(1)吸烟对照组的气道反应性明显增高，吸烟加运动应激可明显降低大鼠的气道反应性；(2)吸烟运动组运动后血浆皮质醇浓度明显高于运动前，但全部运动结束后次日晨测定的血浆皮质醇浓度与运动前无明显差异；(3)HE染色显示，吸烟对照组肺组织出现明显的慢性炎症反应，吸烟加运动应激组比吸烟对照组炎症反应轻；(4)慢性吸烟可降低大鼠大气道和小气道BKca mRNA和蛋白表达，而吸烟加运动应激可使BKca mRNA表达上调，但对小气道的蛋白表达无影响；(5)慢性吸烟可降低大鼠大气道和小气道Kv1.5蛋白和mRNA表达，在小气道吸烟加运动应激可减轻这种作用，大气道则无作用；(6)吸烟使支气管平滑肌的FIZZ1/RELMα蛋白表达明显增强，而吸烟加运动组的RELMα蛋白及mRNA均明显减弱。

The mass of added glycerin is 20-50% of the total mass of propylol lignin, soy segregation protein and glycerin.

羟丙基木质素改性大豆蛋白塑料，其特征在于它主要由羟丙基木质素、大豆分离蛋白和甘油原料制得，羟丙基木质素、大豆分离蛋白各原料所占重量百分比为：羟丙基木质素1-25、大豆分离蛋白75-99，甘油的加入量为羟丙基木质素、大豆分离蛋白以及甘油总重量的20-50％。

The main conclusions are as follows:The vitellin of Chrysopa septempunctata was extracted with 0.4mol/L NaCl and then precipitated in cold distilled water. Ion exchange chromatography and proteic gel recovery were used to purify it further. SDS-PAGE analysis shows that the vitellin is composed of two subunits, the large one 165kDa and the small one 46kDa.Then automatic amino acid analyzer was used to analyse amino acid composition of vitellin.

主要结论如下：用蒸馏水沉淀法对大草蛉的卵黄蛋白进行了初步纯化，而后又采用离子交换层析和蛋白胶回收的方法对卵黄蛋白进行了进一步的纯化；通过SDS-PAGE分析表明，大草蛉的卵黄蛋白由大165kDa和小46kDa两个亚基组成；用氨基酸自动分析仪对纯化后的卵黄蛋白进行了氨基酸组分分析。

Expression of Caspase-3 and bcl-2 protein In E20 cerebral cortex, the expression of cleaved Caspase-3 in the encephalocoele group are higher than that in control group, and the expression of bcl-2 in the encephalocoele group are lower than that in control group. These findings were consistent with the results of the western blot analysis in E14 Brain.

To investigate the presence of specific adhesion activity, the mrkD adhesin gene of the established type 3 fimbriae expression system was replaced respectively by each of the fimbrial adhesins. Transmission electron microscopy and western blot analysis revealed that most of the recombinant type 3 fimbriae expressed and assembled properly.
