MAIN OUTCOME MEASURES: The contents of monoamine neurotranmitters of norepinephrine, adrenalin, dopamine and 5-serotonin (5-HA) and its metabolites of DHPR, 5-hydroxyindoleacetic acid (5-HIAA) and homovanillic acid in the striatum and marginal area of brain in rats were observed.

主要观察指标：大鼠纹状体和脑边缘区单胺类递质去甲肾上腺素、肾上腺素、多巴胺和5-羟色胺及其代谢产物3，4-二羟苯乙二醇、5-羟吲哚乙酸、高香草酸的含量。

The dopamine release in vivo was monitored by fast cyclic voltammetry and the concentrations of DA and its metabolites dihydroxy-phenylacetic acid and homovanillic acid were determined by high performance liquid chromatography electrochemical detection in the process study. In addition, the effects of some traditional Chinese herbs and related marine drug on behavioral changes were also evaluated in order to provide some suggestive clue for rational drug design of anti-Parkinon's disease .

许多因素都参与PD的DA能神经元的死亡，所以，本研究在制备PD动物模型的基础上，应用国内率先引进的快速周期伏安法检测设备在体动态监测DA的释放，同时应用另一种电化学检测技术—高效液相色谱电化学检测法离体检测DA及其代谢产物二羟苯乙酸和高香草酸的含量，结合行为学测定及其它方法，研究了雌激素和铁在PD发病中的作用，以及一些药物，如吲哚醌、牛磺酸、铁离子螯合剂、胆囊收缩素（CCK-8）、乌鸡白凤丸有效成份、大豆异黄酮及海洋酸性寡糖HSH971，对DA能神经元的影响，从DA能神经递质角度对PD的发病机理提供进一步的实验依据，并为PD的预防和治疗提供新的干预措施。

In the pretreatment group, Bak Foong Pill extracts were given for 14 days prior to MPTP. Immunostaining of tyrosine hydroxythase was used to observe the damage of dopamine neurons, and of Bax to detect the protein levels of Bax. High performance liquid chromatography-electrochemical detection was used to investigate the contents of DA and its metabolites dihydroxy-phenylacetic acid and homovanillic in the Str.

给予BFP预处理后，用酪氨酸羟化酶免疫组织化学法观察黑质神经元的变化；同时应用免疫组织化学法检测Bax蛋白表达；应用高效液相-电化学方法检测纹状体巴胺及其代谢产物二羟基苯乙酸和高香草酸的含量变化。

The plasticity of NSCs was detected by the differentiating test. HESCs were induced into dopaminergic neurons by means of adding Sonic Hedgehog Peptide and fibroblast growth factor eight (FGF8) early under the condition of mocked microenvironment and different development stage of dopaminergic neurons in vivo ,meanwhile made comparison with traditional method . The marker of dopaminergic neurons was tested by immunocytochemistry and RT-PCR, the content of dopamine and its derivation homovanillic acid were measured by HPLC-ECD.

Methods The effects of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine and 7-nitroindazole (7-NI) on nitric oxide synthase activity, levels of dopamine、3,4-dihydroxyphenylacetic acid、homovanillic acid, and TH-immunostained postive nerve fibres in the striatum of mice were determined by using colorimetic analysis, HPLC with electrochemical detection, and immunohistochemical analysis.

Hours later,these mice were decapitated,and the striatums were isolated from thebrains. The levels of dopamine, serotonin ( 5-HT ), homovanillic acid, 5-hydroxyindoleacetic acid ( 5-HIAA ) andnorepinephrinein the striatum were measured by high-performance liquid chromatography with electrochemical detection.

Immunohistochemistry technology was used to evaluated tyrosine hydroxylase of DA synthesis.
