To determine whether N6amt1 methylates adenine residues in DNA, we examined its activity by in vitro assay.

为了检测N6amt1是否能够甲基化DNA中的腺嘌呤碱基，我们运用体外酶活检测系统对它的活性进行了测定。

The results show that all the hmgr genes from pigs, cattle and chickens are astable enzyme proteins with the same gene sequence characteristics and encoding protein and physical and chemical properties; all the amino acid sequences have a high degree of homology, containing two conservative HMG-CoA binding motifs and two NADPH binding motifs; all the amino acid sequences, containing signal peptide, are secreted proteins; they are all hydrophobic transmembrane proteins with five transmembrane domains; random coil and α-helix are the main structural elements in the HMGR polypeptide chain; all the predictions of three-dimensional structure are V- shaped; there are more closer evolution relationship between the HMGR gene from pigs and cattle.

结果表明：猪、牛和鸡HMGR基因序列特点及编码蛋白质理化性质基本相同，均属不稳定类酶蛋白；保守结构域氨基酸序列具有高度的同源性，均含有保守的2个β-羟基-β-甲基戊二酸单酰辅酶A结合基序和2个还原性烟酰胺腺嘌呤二核苷酸结合基序；均有信号肽，属分泌型蛋白质；属于跨膜的疏水性蛋白，均有5个跨膜结构域；无规则卷曲和α-螺旋是HMGR多肽链中的主要结构元件；三维结构预测都呈"V"字形；猪和牛HMGR基因的进化关系最近。

RNA editase transforms or modifies the nucleotide of pre-RNA by deaminization, and hydrolytic deaminize C6 amino group of the adenosine at the specific RNA site, changing adenosine to inosine or changing cytosine to uracil,but inosine was recognized as guanine in mRNA.

After thecomplete genome extraction of the strain was performed, the genomic DNA was partiallydigested by restriction enzyme Sau3AⅠ, the DNA fragments from 1 to 5Kb was clonedinto prokaryote expression vector pET-28a-c, and transformed host bacteria. The resultsshowed that we succeeded in constructing the gene expression library of haemophilusparasuis serovar 5, which is fundamental for the study of advanced gene screening. Inaddition, primer design was performed based on haemophilus influenzae in this study. In addition, PCR was performed by using genomic DNA of haemophilus parasuisserovar 5 as the template. The results demonstrated that we obtained two neo-gene:23SrRNA gene(conserved gene belonging to the large-subunit of ribosome) and adenylatecyclase gene(encodes adenylate cyclase and participates in converting adenyl nucleosidetriphosphate to cyclic adenosine3",5"-monophosphate). Furthermore, the phylogeneticanalyses between the species was performed, and neighbor-joining tree was constructedbased on comparison of 23S rRNA gene sequences, so it was illuminated betweenHaemophilus parasuis and other species in molecular evolution relationship.

选择我国流行优势菌株副猪嗜血杆菌血清5型地方株为研究对象，提取细菌基因组DNA，用限制性内切酶Sau3AⅠ对基因组DNA进行部分酶切，回收大小为1～5Kb的DNA片段，将其连接入原核表达载体pET-28a-c，最后转化宿主菌，结果成功地构建了基因表达文库，为后续的基因筛选工作奠定基础；另外，本研究选择嗜血杆菌属的流感嗜血杆菌为参考对象进行引物的设计，以副猪嗜血杆菌血清5型地方菌株的基因组DNA为模板，进行PCR扩增反应，结果表明成功地获得两个新基因：23S rRNA基因(存在于核糖体大亚基中的保守性基因)和腺苷酸环化酶基因(负责将腺嘌呤核苷三磷酸转变为环腺苷酸)，并进一步做了不同物种之间的分子系统发育分析，构建了基于23S rRNA基因的邻接法系统发育树，阐明了副猪嗜血杆菌与其它菌种的分子进化关系。

MethodsOur study was to treat gastric ulcer by invigorating spleen and benefiting vital energy Chinese drugs through clinical observation and experimental research, and compared with mucosa protectant -gastropine and H2 receptor antagonist -famotidine about the mucosa mitochondrion 8-oxygen guanine DNA glycosylase, thymine glycol DNA glycosylase,3-methyl adenine DNA glycosylase, the effect of gastralgia, splenic asthenia and quality of ulcer recovery.

比较健中愈疡片、黏膜保护剂胃舒平以及H2受体拮抗剂法莫替丁对胃黏膜线粒体 8－氧鸟嘌呤DNA糖基化酶、胸腺嘧啶乙二醇DNA糖基化酶、3－甲基腺嘌呤DNA糖基化酶的影响，并进行健脾益气方药与黏膜保护剂胃舒平以及H2受体拮抗剂法莫替丁对胃溃疡愈合质量的比较和评价。

The results indicate that Ca(superscript 2+) and GA3, single or combined application treatments, promote cucumber seed germination treated with P. harmala alkaloid, increase germination rate, germination vigor and index; as well as α-amylase, protease, lipase, isocitratelyase, nicotinamide adenine dinucleotide kinase and Ca(superscript 2+)-ATPase activities; and GA and CaM content in cucumber seeds; but decrease abscisic acid content. The synergistic effects of combined Ca(superscript 2+) and GA3 on increasing GA and decreasing ABA are oberseved compared with Ca(superscript 2+) treatment.

结果表明，Ca(上标 2+)和GA3单一及复合处理可促进骆驼蓬生物碱处理的黄瓜种子的萌发，使黄瓜萌发率、发芽指数和活力指数提高，萌发种子的α-淀粉酶、蛋白酶、脂肪酶和异柠檬酸裂解酶活性及赤霉素和钙调素含量增加，烟酰胺腺嘌呤二核苷酸激酶和Ca(上标 2+)-ATP酶活性增强，脱落酸含量降低；且Ca(上标 2+)和GA3复合处理对GA含量的提高和ABA含量的降低与Ca(上标 2+)单一处理比较呈显著的加成作用。
