Methods Three-dimensional gels of type I collagen containing rabbit keratocytes were incubated in the presence of different concentrations of exotoxin A (0.1,1.0,10 mg·L-1),cultivated for 24 h at 37℃, the change of keratocytes in morphology was observed under the light microscope,and the amount of lactate dehydrogenase was measured by enzyme linked immunosorbent assay.

含兔角膜基质细胞（1×105·mL-1）三维胶原凝胶表面添加提纯的不同浓度绿脓杆菌外毒素A（0.1、1.0和10 mg·L-1）后，放入无血清MEM培养液中37℃培养24 h，采用光学显微镜观察培养的兔角膜基质细胞形态学变化，采用酶联免疫吸附法检测损伤的兔角膜基质细胞释放的乳酸脱氢酶含量。

RESULTS We successfully obtained human caspase-3 gene, the genes and expression vectors of reversed caspase-3 and its fusion protein with part of the translocation domain of Pseudomonas exotoxin A. Compared with the control group, the expression of both genes caused in a similar degree the decrease of living cell numbers and massive occurrence of apoptosis.

结果成功获得了caspase-3基因，构建了重构型caspase-3基因和重构型caspase-3与绿脓杆菌外毒素转膜结构域部分肽段的融合蛋白基因及其表达载体，与对照组相比，在转染后1～4 d内，两种基因的表达均导致HeLa细胞存活数减少，大量细胞发生凋亡，且二者活性相当。

During the surgery, the lesion was found to be a solid tumor. A surgical wedge biopsy was done and purulent dark yellow fluid was drained from the center of the mass. Immunohistochemical staining was positive for vimentin, a mesenchymal marker; the reticulin staining was consistent with carcinoma.

PTX at tenuates the ALI induced by LPS or sepsis in rat. The therapeutic effects of PTX include:improve oxygenation, at tenuate pulmonary edema formation, enhance animal survival in sepsis-induced ALI. The mechanisms may related with the effects of PTX inhibiting the chemotaxis, migration, activation of inflammatory leukocytes, diminishing the production of active oxygen, and inhibiting inflammatory cytokine release partly.

4PTX对大鼠内毒素性ALI及脓毒症性ALI均有一定的防治作用，主要表现在改善动脉血氧合，减轻肺水肿形成，提高脓毒症性ALI大鼠的存活率等方面，其机理可能与抑制白细胞趋化与激活，减少肺内白细胞渗出及活性氧产生，一定程度上抑制炎性细胞因子生成等有关。
