Methods Three-dimensional gels of type I collagen containing rabbit keratocytes were incubated in the presence of different concentrations of exotoxin A (0.1,1.0,10 mg·L-1),cultivated for 24 h at 37℃, the change of keratocytes in morphology was observed under the light microscope,and the amount of lactate dehydrogenase was measured by enzyme linked immunosorbent assay.

含兔角膜基质细胞（1×105·mL-1）三维胶原凝胶表面添加提纯的不同浓度绿脓杆菌外毒素A（0.1、1.0和10 mg·L-1）后，放入无血清MEM培养液中37℃培养24 h，采用光学显微镜观察培养的兔角膜基质细胞形态学变化，采用酶联免疫吸附法检测损伤的兔角膜基质细胞释放的乳酸脱氢酶含量。

Methods The cells were divided into 5 treatment groups（10,25,50,75 and 100 μmol·L-1 QUE）, blank control and menstruum control group. The rat C6 cells were cultivated to 1×106·mL-1 in the RPMI 1640 medium, then added into 96 holes board with various doses of QUE by 3 holes per group,and MTT assay was used to observe the proliferation of the cells treated for 24,48 and 72 h. The change of cell cycle was also observed by flow cytometry after the cells were treated with 50 and 100 μmol·L-1 QUE for 48 h. The changes of the protein P53 and Bcl-2 of C6 cells treated with 50 μmol·L-1 QUE for 48 h were detected by immunocytochemical methods.

按QUE浓度分成10、25、50、75及100 μmol·L-15个处理组和空白对照组及溶剂对照组，大鼠脑胶质瘤C6细胞在RPMI 1640培养基中生长达1×106·mL-1后，在96孔板中分别加入上述浓度的QUE继续培养，每组设3复孔，作用24、48及72 h，采用MTT比色法检测QUE对大鼠脑胶质瘤C6细胞的增殖抑制情况，流式细胞术对50及100 μmol·L-1的QUE作用48 h的大鼠脑胶质瘤C6细胞进行周期分析，免疫组化法检测50 μmol·L-1的QUE作用48 h 的p53和bcl-2基因产物。

He type of ossein in group B was mainly type Ⅰ collagen while that in group A type Ⅱ and it exhibited as a process of fiber formation in group C. Inaddition,we extract total RNA from cultural U251 cell of humanglioma,make coding gene extron amplification of TfR through ReverseTranscription-Polymerase Chain Reaction,construct expressionplasmid of pCDNA3.1-TfR after digestion of enzyme andconjunction,do transfection to U251 cell by liposome afteridentification,cause excess expression,detect the efficiency of transientexpression of TfR protein by pEGFPN1-TfR, sieve positive cell ofpCDNA3.1-TfR through G418, choose positive cell clone, detect theexpression of TfR in anteroposterior period through Western blot,FCM,cell immunochemistry and mRNA through RT-PCR, identify the effect of transfection.

另外，培养人脑胶质瘤U251细胞，进行细胞总RNA抽提，运用RT-PCR方法进行TfR基因的完整编码区外显子的扩增，经过酶切、连接后构建pCDNA3.1-TfR的表达质粒，鉴定正确后按脂质体转染方法转染人胶质瘤U251细胞，使其过量表达，通过pEGFPN1-TfR检测其瞬时转染的效率；用G418对稳定表达pCDNA3.1-TfR的细胞进行筛选，挑选阳性细胞克隆，运用Western blot、流式细胞检测技术和细胞免疫化学以及mRNA半定量的方法检测TfR在转染前后的表达水平，鉴定转染的效果。

研究结果显示：未妊娠时，泛角蛋白在子宫内膜腺上皮细胞、腔上皮细胞内表达，波形蛋白在子宫内膜基质细胞内表达，平滑肌肌动蛋白在子宫平滑肌和血管平滑肌内表达，牦牛子宫任何部位均不表达角蛋白7；妊娠30天左右时，泛角蛋白在子宫内膜腺上皮细胞、子宫内膜腔上皮细胞、滋养层细胞、内胚层细胞和尿囊细胞内表达，波形蛋白在子宫内膜基质细胞和内胚层细胞内表达，平滑肌肌动蛋白在子宫平滑肌和血管平滑肌内表达，角蛋白7在尿囊细胞内表达，偶尔在腔上皮细胞的细胞核边缘表达；消化法进行原代培养时，组织经胶原酶消化并通过100目和400目筛网组合可以有效地分离原代子宫内膜基质细胞和子宫内膜腺上皮细胞；分离得到的子宫内膜基质细胞活率达90%以上，并可在体外传代7次以上；分离得到的子宫内膜腺上皮细胞活率可达85%以上，并可在体外传代5次以上；RPMI1640培养基最适合子宫内膜基质细胞和子宫内膜腺上皮细胞的生长，维持子宫内膜基质细胞正常生长的FBS添加量为20%，维持子宫内膜腺上皮细胞正常生长的胎牛血清添加量为30%。

At histopathologic analysis, neuronal tumors are usually classified as pure neuronal cell tumors (gangliocytoma, Lhermitte-Duclos disease , central neurocytoma) and mixed neuronal-glial tumors (ganglioglioma, desmoplastic infantile ganglioglioma, dysembryoplastic neuroepithelial tumor, ganglioneuroma).

Results: The findings on CT and MRI were as follows: 57 cases of Oligodendroglioma, 15 cases of Ganglioglioma , 5 cases of Dysembryoplastic neuroepithelial tumor , 3 cases of Pleomorphic xanthoastrocytoma .

Many factors may be involve in the course. To investigate the regulation activity of mesenchymal cells to differentiation of epithelial cells from hair follicle and to study its differentiation property, mesenchymal cells gel was made by nubby dermal papilla cells, free dermal papilla cells, skin fibroblasts. Skin keratinocytes and epithelial cells from hair follicle were inoculated on the gel surface and cultured in air-liquid interface. Three-dimensional model of DPC using to induce epithelial cells differentiation is built in vitro.
