胶素

Functional properties of silk fibroin were measured. The results showed that its emulsion capacity and emulsion stability were good at wide pH range from 3 to 10.Sodium chloride decreased EC and ES to some extent. EC and ES improved as the concentration of silk fibroin increased, until reaching its critical micelle concentration 0.5%. Foam capacity of fibroin was lower than that of caseinate, while its foam stability was markedly better than that of caseinate.

对丝素的功能性质进行了研究，结果表明，丝素在pH3～10范围内具有较好的乳化能力和优良的乳化稳定性；氯化钠的添加使丝素乳化能力和乳化稳定性略微下降；随着丝素浓度增加，丝素乳化能力和乳化稳定性均提高，当丝素浓度高于丝素临界胶束浓度0.5％时，乳化能力增幅变缓。

Based on the previous literatures, the following major innovation works were carried out in this dissertation:(1) The derivatization reaction conditions of ephedrine and pseudoephedrine with sensitive derivation reagent in aqueous system were studied and two new methods were developed for the assay of them by microemulsion electrokinetic chromatography and micellar electrokinetic chromatography with LIF detection. The sensitivity and analysis times were greatly improved compared with the previous reports;(2) A new method of CZE with indirect LIF detection was developed for the simultaneous determination of six coumarin compounds (esculin, esculetin, isofraxidin, genistein, naringin and sophoricoside) with fluorescein as the probe. The proposed method enlarges the application range of LIF detector, and provides new approach for the analysis of certain compounds difficult to derivatize;(3) Based on the above research, an MEKC with indirect LIF detection method for the simultaneous determination of adenine and guanine in DNA extracts from fungus, maize and soybean was established;(4) A new method for the investigation of the complexes formed between human serum albumin and ampicillin sodium under the simulated physiology conditions using laser light scattering technique was developed.

该论文在综述前人工作的基础上，开展了如下未见文献报道的创新性的研究工作：(1)使用灵敏的衍生试剂对麻黄碱和伪麻黄碱在水体系中的衍生反应条件进行了系统研究，建立了微乳电动色谱-激光诱导荧光检测法和胶束电动色谱—激光诱导荧光检测法测定麻黄碱和伪麻黄碱的灵敏分析新方法，与以前的报道相比，灵敏度和分析时间均有很大改善；(2)使用荧光素钠作为背景荧光试剂，建立了同时分析测定六种黄酮类化合物(秦皮甲素、秦皮乙素、异秦皮定、染料木素、柚皮苷和槐角苷)的毛细管区带电泳—间接激光诱导荧光检测新方法，扩大了激光诱导荧光检测的应用范围，对难衍生化合物的分离分析提供了一种新思路和新途径；(3)以荧光素钠作为背景荧光试剂，建立了一种用于同时测定食品提取DNA中的腺嘌呤和鸟嘌呤含量的胶束电动色谱—间接激光诱导荧光检测新方法；(4)用动态和静态激光散射法研究了模拟生理条件下人血清白蛋白与氨比西林钠盐相互作用所形成的复合物，为药物分子与蛋白质的相互作用、药代动力学等的研究提供了一种新思路。

The experimental results showed that pinocembrin, chrysin and galangin were the main flavonoids in Chinese propolis.

实验结果表明：乔松素、白杨素和高良姜素是中国蜂胶的主要黄酮类化合物；我国不同产地的蜂胶主要化学成分基本相同。

HPLC-DAD methods for analyzing of the nine compounds in Chinese propolis, samples, including caffeic acid, ferulic acid, p-cinnamic acid, kaempferol, apigenin, pinocembrin, chrysin and galangin and for the chromatographic fingerprint of propolis in different regions of China were established.

建立了高效液相色谱法测定中国蜂胶中9个酚类化合物(咖啡酸、p-香豆酸、阿魏酸、山奈酚、芹菜素、白杨素、高良姜素、乔松素、松鼠素)含量的方法；建立了中国蜂胶的高效液相色谱指纹图谱。

From the Gel Chemistry we know that the homogeneousness of gels' structures had effect on its' transparence.

纯凝胶和MCMB凝胶素坯的热分析结果显示，凝胶高聚物从233.3℃开始发生热分解，在444.7℃之前基本分解完毕，而MCMB则从392.3℃附近开始发生热分解，在678.8℃附近热分解基本结束。

Collagenase activity was defined as the radioactivity extractable by 50% dioxane. Clostridial collagenase was used as a standard. The lower limit of this assay method was 0.01 units of clostridial collagenase. Among 17 glaucoma eyes, 6 had trace collagenase activity (around 0.01 units) in aqueous humor; and for cataract eyes, 8 out of 27 had trace collagenase acitivty.

我们所采用之测量方法可侦测出约0.01单位以上的胶原酵素；一般前房液之胶原酵含量非常低，许多患者无法以我们之方法利得胶原酵素之存在：17位青光眼患者中，只有6位可於其前房液中到得约0.01单往之胶原酵素；27位白内障急者中，有8位可测得微量胶原酵素；二者在统计学上与太大差异存在。

Results The bletilla colloid was a homogenous viscous colloid whose relative viscosity was 2324.6 mm2/s. It was easily injected through 4-F catheter and hyperattenuating under fluoroscopy, meanwhile, with good histocompatibility and hemo-compatibility, without pyrogenetic response and toxicity. In vitro, the mixture of bletilla colloid and MMC did not produce separation and suspention phenomena but released 50%of MMC at 1.8h and 100%at 3.4h. The bletilla colloid mainly embolized peripheral arteries, maintaining occlusion for 5 weeks and without formation of collateral circulation. The injuries of normal hepatic tissues were slight, without hepatic cytonecrosis.

结果 (1)白芨胶为一种均质的胶状物，有一定粘滞性，相对粘度2 324.6 mm2/s，不透X线，易经4F导管注射；(2)白芨胶具有良好的血液相容性和组织相容性，无致热原性及毒性作用；(3)白芨胶-丝裂霉素C体外混合后，无分层及悬浮现象，释放MMC 50%约需1.8小时，100%需3.4小时；(4)白芨胶主要栓塞末梢小动脉，栓塞牢靠，维持时间可达5周，不易形成侧支循环；(5)白芨胶栓塞对正常肝组织损伤轻微，未出现肝细胞坏死。

目的：甘露聚糖结合凝集素(Mannose-binding lectin， MBL)是属于C型凝集素家庭胶原凝集素的一种血清蛋白，包括N末端区（N-terminal segment）、螺旋铰链区（helical coiled-coil hinge region）、胶原样区（collagen-like region，CRL）及球形C型凝集素区或糖识别区（carbohydrate recognition domain ，CRD）等4部分结构区域，四个亚单位的MBL形成有功能的多聚体。

objective to investigate staining methods for two-dimensional gel(2-de)electrophoresis in multidrug resistance of gastric cancer.methods cultured vincristine-resistant human gastric cancer cell line sgc7901/vcr and its parental cell line sgc7901.variant amount protein of those cells were separated by 2-de.gels were stained with silver nitrate or colloidal coomassie brilliant blue,and scanned by image scanner.results well-resolved,reproducible 2-de patterns of sgc7901/vcr and sgc7901 were established.silver staining was better when protein sample amount was low,overloaded protein will interfere resolution of the maps.gels stained with colloidal coomassie brilliant blue had more protein spots numbers and abundance without apparent trails when increased loading protein sample.conclusion two staining methods were influenced largely by the sum of protein samples,properly selection may be helpful for further study with proteomics in multidrug resistance of gastric cancer.

低甲氧基果胶的胶凝机理及防止预凝胶。。。他扎罗汀凝胶与克林霉素凝胶治疗痤疮。。。注射隆乳后经乳晕切口取出聚丙烯酰胺。。。目的分析利用蛋白质组学方法研究胃癌耐药相关蛋白质中双向电泳凝胶的染色显示。方法培养胃癌细胞sgc7901和长春新碱诱导的耐药胃癌细胞sgc7901/vcr，用双向凝胶电泳技术分离总蛋白，银染及胶体考马斯亮蓝染色，image scanner扫描仪扫描凝胶。结果获得了背景清晰、重复性好的双向凝胶电泳图谱，两种染色凝胶相比，硝酸银染色在样品少时显示更佳，过量则影响图像质量，而胶体考马斯亮蓝染色在上样量增加时的凝胶蛋白质点数目及丰度均增加，并无明显拖尾。结论两种显色方法受样品量影响较大，恰当选用有利于通过蛋白质组学研究胃癌耐药机制工作的进一步开展。
