The sizes of the self-assembly micelles of the amphiphilic block copolymers and the effects werestudied by dynamic light scattering and UV-visible spectrophotometer. The micelle sizes werelargely influenced bythe hydrophobic chain content in the copolymer, the properties of the solvent and the copolymer concentration in the organic phase. The morphology of the micelles was investigated by transmission electron microscopy. Theresults showed that the micelles appeared spheres with inner core and outer shell. The critical association concentrations were determined by pyrene monomer fluorescence probe technology. The degradability of copolymer was studied by the loss of the intrinsic viscosity in the degradation process. The results indicated that the degradation rate of PECL was slower thanthat of PELLA and PEDLLA. The stability of the copolymer micelle dispersion was examined by measuring the critical flocculation concentration, which gradually decreased with increasing the content of thehydrophobic chain in the copolymer. The rheological results showed thatthe viscosity ofthe micelle dispersion with higher content of PEG segments was firstly decreased and then increased with the increasingthe temperature. Otherwise, the variation of viscosity was irregular. The viscosity of the micelle dispersion increased with the increase of the electrolyte (Na 2SO 4) concentrations. Polymer micelles paclitaxel was prepared by self-emulsification/solvent evaporation method and solid dispersion technique with amphiphilic block polymers as the carrier material.
运用动态光散射和紫外分光光度计研究了两亲性嵌段共聚物自组装胶束的粒径及其影响因素,结果表明,自组装胶束的形成机理受制备方式的控制,胶束的粒径随着共聚物相对分子质量或疏水嵌段相对分子质量和有机相中共聚物浓度的增大而增大,随着有机溶剂的水溶性的增强而减小;采用透射电镜观察胶束的形态结构,发现胶束呈具有核壳结构的均匀球形;采用芘荧光探针法测两亲性嵌段共聚物的临界聚集浓度;通过降解过程中共聚物特性粘度的变化研究两亲性共聚物的降解性能,发现与PELLA和PEDLLA的相比,PECL 降解速率较慢;随着共聚物中疏水嵌段含量的增大,纳米分散液的稳定性逐渐下降;PEG含量高的PEDLLA纳米分散液随着温度的升高,纳米分散液的粘度先下降然后增大,反之,没有规律性,且随着电解质Na 2SO 4浓度的增大,体系粘度增大。

In Part 1 of our research, BGSCs were sorted through immunomagnetic beads marking by CD133 and cultured in vitro, and character as a stem cell was identified by stem cell markers (CD133 and Nestin) and differentiated cell markers [ microtubule-associated protein 2(MAP2), glial acidic fibrillary protein and myelin basic protein] , ultrastructure observing with electron microscopeand engrafting to severe combined immunodeficiency mice for tumorigenesis test. The results were as following: Only a small subset of CD133+ glioma cells in glioma cell lines and fresh specimens from various pathologic grade could express stem cell markers CD133 and Nestin, view ultrastructure of a stem cell and be capacity of serial passage in culture. These CD133+ cells possese a marked capacity for multipotent differentiation and could differentiate into tumor cells expressing MAP2,β-TubulinⅢ, GFAP and MBP; When engrafted into SCID mice, they can generate and form tumors that phenotypically resembl the tumor from the patient.
在本课题中,在第一部分实验中采用以CD133为标志的免疫磁珠法从人脑胶质瘤组织和细胞株中分离脑胶质瘤干细胞并进行体外培养,通过免疫荧光技术检测干细胞标志物CD133、Nestin,诱导分化后检查分化细胞标志物MAP2、GFAP、MBP以及电镜超微结构观察和移植SCID鼠致瘤实验,对其干细胞特性加以鉴定,得到如下结果:不同病理分级的新鲜胶质瘤标本和胶质瘤细胞株中存在一小部分CD133+的胶质瘤细胞,能表达干细胞的标志物CD133和Nestin,符合干细胞的超微结构特点,体外培养能连续传代;具有多向分化潜能:诱导分化后能产生MAP2、β-TubulinⅢ、GFAP、MBP染色阳性的细胞;移植SCID鼠后能形成与亲本肿瘤表型一致的移植瘤。

The rubber seal issue under operating system has been studied by the theory of dynamic sealing. The deformation and stress distribution of rubber material were calculated under the well in-pressure and sealing out-pressure using then-linear finite element. Through the experimental study of the rubber strength and fatigue, the intensity limit and the fatigue life were obtained. According to the study results, a new type of sealing equipment was designed by adding the reinforcing steel bar into rubber core to reduce the maximum shear stress, so as to solve the problems of dynamic seal under hyperbaric pressure.
