Using immuno histochemical analysis was seen a positive staining in the basal cytoplasm of blastoderm embryos at 4 hours. This result confirms that the DLMO is a cytoplasmprotein, not like human LMO protein.

免疫组织化学的分析在0～4小时胚盘外周胞浆中有Dlmo基因的阳性染色信号，结果证实，Dlmo与人类LMO不同，其表达产物是一个胞浆蛋白。

The results showed:(1) the cellule morphous and the disposition in chick embryo of PGCs were coincident no matter stained by PAS or HE staining, and HE staining could disclose the morphologic characteristics more clearly, exactly and completely;(2) genesis of blood island could be observed at the boundary of light and dark region of the extraembryonic blastoderm at about 26 hours;(3) both the blood vessel endothelium cells and free cells of the blood island were differentiated from PGCs. The generating of genuine yolk sac was at about 44-48 hours.

结果表明：①用经典的识别原生殖细胞的PAS染色方法与HE染色所观察到的PGC在鸡胚的分布及细胞形态基本一致，而HE染色较PAS法能更有助于清晰、完整、准确地识别PGC的细胞内部结构特征；②在孵化26小时的鸡胚胚外胚盘的明暗交界处可见血岛的发生，血岛发生的时间比真正的卵黄囊产生时间（大约44-48小时）要早很多；③血岛周围的血管内皮细胞与管腔中游离细胞都来自PGC。

It is concluded that the initial ancotomic site of blood island genesis may be is mesoblast of extraembryonic blastoderm rather than the yolk sac; the blood vessel endothelium cells and the blood cells are generated parallel; the PGCs are the common ancestry of angioblast and HSC.

血岛最早发生的解剖位置可能不是卵黄囊而是胚外胚盘中胚层，血管内皮细胞与血细胞平行发生，PGC是成血管－造血干细胞的共同祖先。

GFP-labeled and non-labeled cells from X blastoderm were large and round with the same characters of morphology. The GFP protein was not expressed in CEF cell .Our results suggested that the CVH promoter regions have specific activity in stage X blastoderm.

In blastula embryos and 50% epiboly embryos, the zygotic pou2 transcript is also distributed in every blastomere cell. At the stage of 90% epiboly, expression of pou2 converges to the dorsal midline of embryos and locates in two waves of lateral neural plate and two transverse stripes in the brain region. In embryos at 100% epiboly, pou2 transcript is restricted in the neural plate and areas of presumptive midbrain amid himdbrain. From somite stage, gibel carp pou2 can not be detected any more, which is consistent with the RT-PCR result.

在高囊胚期和50%下包期，高度表达的合子转录本仍在所有的胚盘细胞中，但至90%下包期时，pou2的表达向胚胎背部的正中线汇聚，集中在神经板的两侧区域和脑部的两条横向条带在100%下包期时，pou2的表达集中在神经板的中间区域以及预期形成的中后脑区域至体节期时，转录本消失，这与RT-PCR结果高度一致。

Temporal and spatial expression pattern of gibel carp pou2 during embryogenesis was investigated by RT-PCR and whole-mount in situ hybridization. RT-PCR result indicates that there is maternal transcript of gibel carp pou2 in early embryos and the zygotic pou2 transcript is abundantly expressed from blastula stage. Large amount of pou2 transcript is detected at both 50% and 90% epiboly stages. At 100% epiboly stage, however, the pou2 transcript reduces rapidly and disappears at the later embryogenesis. Whole-mount in situ hybridization shows that the maternal transcript ubiquitously exists in all blastomere cells.

我们用RT-PCR和整体原位杂交的方法研究了银鲫pou2基因在胚胎发育过程中的时空表达图式RT-PCR结果显示，银鲫pou2基因有母源转录本，其合子基因在高囊胚期强烈表达，在50%下包期和90%下包期也有高量的转录本，但在100%下包期表达量急剧降低，至体节期时已经完全检测不到其转录本胚胎整体原位杂交结果显示其母源转录本在所有的胚盘细胞中。
