Research Direction Our research try to block the activation of transmembrane signal transduction related to inflammation by blocking the receptor of cellular membrane.

研究方向细胞内信号转导通路的激活要通过细胞膜受体的介导，本研究拟阻断细胞膜受体的激活，从而阻断肺细胞内各种炎症相关性信号转导通路的激活。

In the whole rats, the following tissues or organs were extracted and fixed in the most appropriated fixator (Bouin liquid or Formalin at 10%) then included in paraffin blocks : all the organs showing significant macroscopic lesions, encephalon (brain hemisphere, cerebellum and annular protuberance), spinal cord, sciatic nerve, salivary glands, esophagus, stomach, duodenum, jejunum, ileon, caecum and colon, rectum, liver, spleen, bone marrow, lymph node close to and at distance, thymus, heart, aorta, trachea, lungs, thyroid/parathyroid, surrenals, pancreas, hypophysis, kidney, bladder, gonad, uterus, mammary glands, epididymis, prostate, eyes, lachrymal glands, bones, skin, thigh muscle and implantation sites (2 per animal).

从整体大鼠内，下列组织或器官提取后用最适宜的固化剂（ Bouin 液或10%的甲醛溶液）进行固定，然后包入石蜡块内：所有显示有明显肉眼可见损害的器官、脑髓、脊髓、坐骨神经、唾液腺、食道、胃、十二指肠、空肠、回肠、盲肠、大肠、直肠、肝脏、脾、骨髓、近端和远端淋巴结、胸腺、心脏、主动脉、气管、肺、甲状腺/甲状旁腺、肾上腺、胰腺、垂体、肾、膀胱、性腺、子宫、乳腺、附睾、前列腺、眼睛、泪腺、骨骼、皮肤、股肌以及假体植入部位（每只动物2处）。

During the first two weeks after birth, the growth performances were not significant between the mutant mice and the normal ones, but after that, the mutant mice grew much slower. The differences were significant in the indexes of eyes, lungs, thymuses, and adrenal glands between the genders. At the same time, the differences were significant in the organ indexes, such as lungs, adrenal glands, uterine and testicles between the normal phenotype mutant mice and the abnormal ones. And these two kinds of mice showed different amount of hemoglobin and red blood cell.

结果：B6-Co小鼠不育率22%、平均产仔数5.62±2.55只、离乳成活率56.59±41.22显著低于正常的B6小鼠，而胎间隔29.50±10.50天比正常B6小鼠短。B6-Co小鼠出生后2周内生长发育与正常B6小鼠基本一致，2周后，其生长发育慢于正常B6小鼠。B6-Co小鼠雌雄间的眼球、肺、胸腺、肾上腺的脏器指数差异有统计学意义。B6-Co表型异常小鼠与B6-Co表型正常小鼠相比，肺、肾上腺、子宫、睾丸的脏器系数差异有统计学意义。B6-Co小鼠表型异常小鼠在血红蛋白和红细胞数量上异于B6-Co表型正常小鼠。

Results: B6-Co mice showed higher sterile rate, and significant lower average litter size and survival rate at weaning than the normal B6 mice, but the time interval between deliveries was shorter in the mutant population. This survival time was not significantly altered by bilateral denervation of cervical vagi (the major sensory system for detecting changes in pulmonary conditions), by bilateral denervation of peripheral arterial chemoreceptors (the major sensory system for detecting changes in arterial blood gases and pH), by pretreatment with dimethylthiourea, or by pretreatment with milrinone (a phosphodiesterase Ⅲ inhibitor that may avoid respiratory muscle fatigue).

而截断双侧颈部迷走神经（负责侦测肺内部情况的最主要感觉神经）、摘除双侧的周边化学接受器（负责侦测动脉血液气体及酸碱值的最主要化学接受器）、或投予Milrinone（一种phosphodiesteraseⅢ抑制剂，藉抑制cAMP被分解，以增加细胞内cAMP的量，可以使疲乏的呼吸肌内再增加收缩力）、Dimethylthiourea等药物的动物，都无法延长其存活时间。

气管内注入博莱霉素（bleomycin，BLM）复制大鼠肺纤维化模型，并分离培养肺成纤维细胞，用阿托伐他汀对其进行干预，分为空白组、TGF-β1诱导组和不同剂量阿托伐他汀阻断组，用倒置纤维镜观察细胞的形态结构，MTT计数法观察各组细胞的增殖情况，免疫组化染色法定性观察α-平滑肌肌动蛋白(α-smooth muscleactin ，α-SMA)的表达情况。

用地高辛随机引物法标记exJSRV特异的env片段，制备探针，原位杂交检测SPA肺组织中的RNA及前病毒DNA，结果表明SPA患羊肺组织内有JSRVenv基因mRNA的表达，同时也检测到了前病毒DNA，而相应的阴性对照却无阳性信号，证实外源性病毒特异的DNA探针在致瘤性前病毒的整合位点和整合的外源性前病毒的检测中具有可信度。

To confirm the hypothesis that the pathogenic mechanism of air infusion in the circulatory system is not bubble blockade of blood vessels but the consequence of anoxia of the vital organs,different amounts of air were injected into the great sapenous vein of 22 dogs,and then heart rate,cardiac rhythm and bubble sound were monitored with a Doppler probe at the precordial region.

为探讨循环系统内注入空气致病机理，向22只狗大隐静脉内注入不等量空气并在心前区用Doppler监测心率、心律和气泡音。结果发现，维持心前区Ⅳ级气泡音的17只狗，依次出现呼吸和心率加快、心律紊乱、心率缓慢、腹式呼吸加强，其中12只狗继续缓慢注入空气至呼吸停止，任选10只狗胸外按压，8只狗复苏后行为无异常，2只狗未能复活。另2只狗呼吸停止后立即尸检，可见右心房、室、腔静脉和肺血管中有大量气泡。
