Objective: To modify the technique of surface preparation of cochlea and staining for hair cell examination.

目的 ：探讨一种观察耳蜗听毛细胞病理改变的耳蜗铺片与酶组织化学检查技术。

Objective To introduce a method of chronic infusion of drugs into the cochlea using a osmotic pump.

目的 介绍微型渗透压泵进行耳蜗灌注的方法，达到耳蜗慢性给药。

But the cochlear lesions could been occurred in hydropic ears because the ototoxic sensitivity was increased in both cochlear and vestibular sensory cells of hydropic ears which should be paid more attention to clinical application.

然而积水状态下耳蜗和前庭感觉细胞的耳毒敏感性均增加，临床应用时可能导致耳蜗损害，听阈升高，应引起注意。

METHODS: Healthy, pigmented, adult guinea pigs of both sexes with intact Preyer's reflex were divided into 3 groups (cochlea group,brain group and lung group).

成年健康杂色豚鼠，耳廓反射灵敏，分别取耳蜗、脑、肺三组组织，免疫组织化学法及Western Blot研究紧密连接蛋白occludin，ZO-1在耳蜗外侧壁血管纹中的表达，以脑组、肺组为阳性对照。

The major HRCT findings were presented with soft tissue mass in 89 middle ears(100%),the destruction of temporal bone including enlargment of the tympanic cavity 89 ears(100%),tympano scutum 55 ears(61.80%),ossicular chain 82 ears(92.13%),facial nerve cannal 54 ears(60.67%),semicircular canal 18 ears(20.22%),external auditory canal 10 ears(11.24%),tympanic tegmen 15 ears(16.85%),vestibular 1 ear(1.12%),cochlea 1 ear(1.12%) and sigmoid sinus plate 7 ears(7.86%).

HRCT表现：中耳腔扩伴软组织团块89耳(100%)，盾板破坏55耳(61.80%)，听小骨破坏82耳(92.13%)，面神经骨管破坏54耳(60.67%)，半规管破坏18耳(20.22%)，外耳道骨质破坏10耳(11.24%)，鼓室盖破坏15耳(16.85%)，前庭及耳蜗破坏各1耳(1.12%)，乙状窦周围骨质破坏7耳(7.86%)。

In Dex-pretreated group, no changes in CoBF was found after the administration of salicylate sodium.

水杨酸钠可减少耳蜗血流量，地塞米松可减轻水杨酸盐对耳蜗血流量的影响，内耳花生四烯酸代谢异常可能介导了水杨酸盐耳毒性的发生。

Then the animals were sacrificed and the surface preparation and the section of cochleae stained by MyosinⅥwere observed.Other cochlea were observed with SEM.

分别于术后第3.5个月分别行听性脑干反应检查后取双侧耳蜗标本做基底膜铺片和耳蜗冰冻切片MyosinⅥ染色观察有无新生毛细胞样细胞出现，部分耳蜗作扫描电镜观察。

Results In three Hank's solution infused cochleas, the thresholds of IC potential were kept in the normal level and no hair cell loss was found 14 days after the beginning of the infusion, suggested that the pump implantation and Hank's solution infusion had no significant effects on the cochlear functions. In contrast, threshold shifts of IC potentials and hair cell loss were found in two BSO infused animals, suggested that the infused solution had distributed into the whole cochlea.

结果 应用渗透压泵耳蜗灌流Hank液后，下丘电位阈值维持正常，耳蜗毛细胞无损伤；灌流BSO液14天后，所测频率(1、 2、 4和8 kHz)的下丘电位阈值平均升高28 dB，内耳毛细胞平均缺失39.3%，缺失分布于耳蜗各回，表明渗透压泵工作良好。

Objective:to primarily find out the effect about luteolin rivalizing ototoxicity of gentamicin.methods: in this experimental study,gentamicin group was compared to the group of gentamicin with different dose of luteolin (10 mg,20 mg).the changes in hearing and vestibule function were discovered by measuring the threshold of abr and the frequency of eng.the method of cochlear preparation stained with sdh was used to observe morphological changes in cochleas.results: the auditory examination results showed that the thresholds of the group of gentamicin with different dose of luteolin were higher than the one of the gentamicin group,and the difference was significant in statistics.the morphology analysis result was consistent with auditory examination.

目的：初步了解木樨草素对庆大霉素耳毒性的影响。方法：设庆大霉素组与庆大霉素加不同剂量木樨草素（10 mg、20 mg）两组，并进行组间比较，观察听性脑干反应阈值、眼震电图的频率，了解其听功能及前庭功能的变化。结合耳蜗基底膜铺片琥珀酸脱氢酶染色方法观察耳蜗形态学变化。结果：听功能检查结果显示，庆大霉素加不同剂量木樨草素组的abr阈值明显低于庆大霉素组，经统计学处理，差异有显著性。形态学检查结果与听功能检查结果一致。
