Cloning of the Schwanniomyces occidentalis α-amylase and high expression in S.cerevisiae.The E.coli / yeast shuttle plasmid YCEpl partial library of Schwanniomyces occidentalis DNA was constructed and α-amylase gene fragments were screened in Saccharomyces cerevisiae by amylolytic activity.Several transformants with amylolysis were obtained and one of the fusion plasmids has about 5.0 kb inserted DNA fragment.It contains the upstream and downstream sequences of α-amylase gene from S.occidentalis .

以E.coli/yeast穿梭质粒YCEp1为载体构建西方许旺酵母部分基因组文库，在大肠杆菌中扩增后提取混合质粒DNA，经电转化非缺陷标志酒精酵母 AS.2.1364，在YPDS平板(含1%葡萄糖和1%可溶性淀粉)上用淀粉水解活力筛选含水解淀粉的阳性转化子，从阳性转化子中分离重组质粒证实含5.0kb的插入片段，用α-淀粉酶基因两端序列设计的引物PCR扩增及扩增片段序列分析证实该片段中含有α-淀粉酶全部编码序列。

Results: All the 14 cases showed hypoechoic masses at the right of pancreas head or inferior to pancreas head. Thirteen cases were diagnosed as duodenal tumor by ultrasonography, another one was diagnosed as enlarged lymphnode. Of all the 14 cases, 13 cases were duodenal tumor demonstrated by the surgical and histological examination, one case was diagnosed as duodenitis by duodenoscopy.

结果：超声检查14例均表现为胰头附近的低回声肿块，13例超声诊断为十二指肠肿瘤，1例诊断为腹膜后肿大淋巴结。14例中，13例经手术病理证实为十二指肠肿瘤，1例经十二指肠镜诊断为十二指肠炎。

In our experiment, the GADes gene from rat brain was cloned and expressed in E.coli, so as to make the recombinant GADes produced by gene technology for clinical application and further study on the relation between GAD and type 1 diabetes mellitus.Firstly, with the pUClS/GADes recombinant plasmid as template, the full-length encoding sequence of GADes was amplified by PCR, and cloned into pGEM-T vector. Secondly, the target gene was cut by EcoRI and inserted into restriction sites of pET42a vector for expression.

本研究首先用PCR方法，以pUC18/GAD_(65)为模板，扩增目的基因，克隆到pGEM-T载体中，构建pGEM-T/GAD_(65)重组质粒，然后用EcoRI酶切，切下的目的基因片段插入pET42a融合型表达载体中，经酶切鉴定和测序，证实插入方向、读码框架正确；重组表达质粒转化大肠杆菌BL21(DE_3)中诱导表达，表达产物经亲和层析纯化后获得了融合蛋白GST-GAD_(65)。

objective the purpose of this study was to summarize our experience of the application of internal rigid fixation with miniplate to treatment of mandibular fractures.methods a retrospective analyisis of clinical data of 67 cases,who had experienced the anatomic reduction and rigid fixation by using miniplate.with the help of maxillomandibular distraction pre operation and post operation and temporary maxillomandibular fixation during operation,the fracture bones were smoothly reduced and fixed.results clinical and radiographic examination showed that all cases gained good clinical results and precise anatomic alignment of fracture postoperatively without infection,malocclusion,bone malunion or other complications.conclusion mocortical miniplate fixation of the mandible is a reliable and effective techinique providing rigid fixation and an ideal modality for the treatment of mandibular fractures.combined with maxillomandibular fixation.

对67例下颌骨骨折患者的临床资料进行回顾总结，所有患者应用小型接骨板行坚固内固定治疗。其中7例颏部骨折，12例颏旁骨折，16例颏孔区骨折，9例体部多发性骨折，5例体部粉碎性骨折；18例合并其他部位骨折。所有患者术前经临床检查和影像学检查明确骨折的位置和骨折线的数目后，分段牙弓夹板结扎、颌间弹性牵引，使移位的骨折段逐渐复位；术中经口内切开复位、细钢丝颌间结扎、小型接骨板坚固内固定；术后行临床检查和影像学复查，必要时颌间弹性牵引7～10 d。结果所有患者手术切口ⅰ期愈合，治疗效果满意；临床检查所有患者牙合关系良好、面部对称；影像学复查证实骨折线对位良好。结论经口内切开小型接骨板坚固内固定辅以颌间固定治疗下颌骨骨折效果可靠，术后并发症少，是一种理想的手术方法。
