Abortus, amplify BP26 gene by PCR and clone to pMD18-T simple vector. Identify the constructed recombinant plasmid pMDBP26 by sequencing, then subclone to vector pET-28a. Transform the constructed recombinant plasmid pETBP26 to E. coli BL21(DE3) for expression under induction of IPTG. Purify the expressed product by histidine-binding resin column chromatography and identify by Western blot.

提取布鲁氏菌基因组DNA，用PCR扩增出BP26基因，并克隆到pMD18-T载体上，测序正确后，再亚克隆至pET-28a载体上，转化到大肠杆菌BL21(DE3)中，用IPTG诱导表达，经组氨酸结合树脂柱纯化，并对纯化后的表达产物进行Western blot鉴定。

This paper deals with making stop bleeding sponge with dried small shrimps. Firstly breaking away with inorganic salt and proteir in the dried small shrimps with acid and NaOH, secondly, taking off vinyl-in the chitin with 50% NaOH, temperature is 110 ℃. the last, reacting with chitosamine and PVA-124 make stop bleeding sponge were produced, catalyst is PdCl 2. The UV and IR spectroscopic studies of the monomer films showed that the photoreaction ratio of monomer (about 50%) was higher than the corresponding photosensitive polymer (about 10%, PVCi). The polarized IR spectra proved that obvious anisotropic absorption was displayed in the irradiated photosensitive film. With polarizing microscopy method, we found that these kinds of photoalignment films exhibited good alignment ability, and the stability of the photoalignment film could reach about 100℃. 2 A photosensitive self-assembled monolayer was fabricated that contained photo-cycloadditable groups, such as, coumarin and cinnamate. The reaction process were detected by contact angle method because the IR absorption of monolayer was very weak. The photosensitive self-assembled monolayer promote uniform alignment of nematic liquid crystals by LPUV, and it thermal stability could reach about 130℃. 3 A cationic bolaform amphiphiles was synthesized that had two photosensitive double bonds, and can be polymerized by ultraviolet. A layer-by-layer self-assembled deposition film was prepared in aqueous solution by using this material and poly (sodium 4-styrenesulfonate). The UV-Vis. spectra proved that the multilayers film was uniform and layer-by-layer. When LPUV irradiated the film, the photopolymerizable double bond underwent [2+2] cycloaddition, and the production of the reaction distributed along the polarized direction of incident LPUV. Polarized UV-Vis. spectra indicated that the irradiated self-assembled multiplayer film was anisotropic, and the dichromic ration is about 0.09, it is higher than the spin-coating method of photosensitive polymer or monomer. We used this anisotropic film as alignment layer of nematic liquid crystals, and obtained a uniform and stability alignment result, the static contrast ratio is about 100, which is very close to application of liquid crystal device. This thesis is an instance that the photochemical material was used in the physics region of liquid crystals, the route that was from photosensitive monomer, self-assembled monolayer film to self-assembled multiplayer film was undergone, by this logical research method photoailgnment of liquid crystal was extended and consummated.

围绕上述思路开展了如下工作：1合成了三种可进行光二聚反应的光敏小分子单体，LPUV 辐照制作了光控取向膜；用紫外-可见光谱法和红外光谱法分析了薄膜中的光化学反应，发现小分子的光反应程度可达到50%以上，至少比光敏高分子高出5倍；薄膜的偏振红外光谱证实，经过LPUV 辐照后薄膜出现了明显的各向异性；将小分子光控取向膜制成液晶器件，在偏光显微镜下观察看到均匀一致的液晶取向；并且热稳定性达到100℃左右，接近实用化要求。2制备了含肉桂酸、香豆素基团的自组装单层膜；采用接触角的方法监测自组装反应的动力学过程；紫外-可见光谱证实了自组装单层膜在石英基板表面形成；自组装单层膜经LPUV 辐照后能诱导液晶均匀取向，而且热稳定性可达130℃，热稳定性进一步提高。3合成了含有光敏双键的双季铵盐，在水溶液状态下将它和聚乙烯基苯磺酸钠沉积生成layer-by-layer 自组装多层膜，紫外-可见光谱证实这是一个逐层均匀的沉积；在LPUV 照射下，自组装膜中的光敏双键按照偏振方向发生[2+2]环加成反应，形成取向膜，用二向色性值评价的分子取向度高于高分子及小分子光控取向膜的水平；实验证明用自组装多层膜制成的液晶器件亮暗态均匀，静态对比度约为100，达到实用化器件的水平。

The polysaccharose of mycelium of Grifola umbellate (PPS1) and the polysaccha rose of sclerotium of Grifola umbellate (PPS2) both were extracted by hot water leaching way, which were purified dispoing by Sevage way, determined the polysac charose content by 3,5-dinitro-salicylate way, and comparablly analysed infrared spectrum by Fourier transforming infrared spectrophotometer,which suggest that two kinds of polysaccharose were the similar infrared spectrum.

本试验通过人工液体浅层培养方式成功培养出猪苓菌丝体；经热水浸提法分别提取猪苓菌丝体多糖(PPS1)和猪苓菌核多糖(PPS2)；用Sevage法进行纯化处理；用3，5-二硝基水杨酸法测定其糖含量；用傅里叶变换红外分光光度计对两种多糖进行红外光谱比对分析，结果表明两者红外光谱相似。

The process of three-step countercurrentleaching is presented, in the first leaching step Fe as leading impurity inthe matte is leached priorly, Co and Ni are enriched in the residue of thefirst step and then are leached in the second leaching step with a lowconcentration acid, the residue from the second step containing hardleaching Fe, Co and Ni are leached with a high concentration sulfuricacid in the third step, and Cu stays in the final resude, the leachate of thethird leaching step is diluted and divided into two parts as the leachantsto the first and second leaching step.

在此基础上，提出了三段逆流选择性浸出技术：即第一段用低酸优先浸出钴冰铜中杂质铁，将钴镍富集到第一段的浸出渣中，第二段再用低酸浸出渣中部分钴镍，第三段用高酸浸出第二段渣中难浸的钴镍铁，将铜留在最终的浸出渣中，第三段浸出液经稀释，分流，分别返回至第一段，第二段作低酸浸出介质。

METHODS: Bone marrow was sterilely separated from human. After heparinization, human BMSCs were harvested using density gradient centrifugation and adherence method. At the fifth passage, BMSCs at 1×108/L were incubated in the 6-well plate and divided into 2 groups. BMSCs in the edaravone group were 50% confluent and incubated in L-DMEM containing basic fibroblast growth factor and fetal bovine serum for 24 hours. After washing in PBS, these BMSCs were incubated in serum-free L-DMEM containing 20 mg/L edaravone for 24 hours. BMSCs in the blank control group were incubated in L-DMEM, supplemented with 10% fetal bovine serum.

无菌抽取的骨髓经肝素化后，采用密度梯度离心法及贴壁筛选法分离获得人骨髓间充质干细胞，传至第5代按1× 108 L-1接种于6孔板内，设立2组，依达拉奉组细胞达50%融合时用含碱性成纤维生长因子、胎牛血清的L-DMEM预诱导24 h，PBS洗涤后再用20 mg/L依达拉奉无血清L-DMEM诱导24 h；空白对照组始终用含体积分数为10%胎牛血清的L-DMEM培养，不加任何预诱导剂和诱导剂。
