In autumn,there were GTH1 cells and GTH2 cells in GTH cells. A large number of secretory granules in GTH1 cells were circumnuclear; In GTH; cells cytoplast,there were two types of secretory granules,small circle and large circle, in small number.Structure of STH cells had no obvious changes.

秋季，在GTH细胞中发现GTH_1和GTH_2两种细胞，GTH_1细胞具有大量的分泌颗粒，并呈围核分布，GTH_2细胞胞质中具有数量较少的小圆形分泌颗粒和大圆形颗粒；STH细胞的结构无明显变化。

Results:①The amount of human colon carcinoma cell line SW480 treated by quercetin decreased. The morphology of partial SW480 cells was shrunk volume, integrated cell membrane, condensed cytoplasm, pyknotic chromatin, nuclear fragmentation. Apoptotic Corpuscles were found by electron microscope.②MTT colorimetric assay showed quercetin inhibited the growth of human colon carcinoma cell line SW480 in a time- and dose-dependent manner when the concentration of quercetin was 30、60、90μmol/L.③Flow cytometry analysis showed the cell cycle of SW480 cell was restricted in G1/S. G0/G1 phase rate increased and S phase rate decreased with increasing concentration of quercetin and time lasting.④ Zymogram analysis assay showed the secretion of matrix metalloproteinases in human colon carcinoma cell line SW480 treated by quercetin decreased. With increasing concentration of quercetin, the secretion of MMP-2 and MMP-9 decreased.⑤Immunohistochemistry method demonstrated the position expression of Cathepsin-D in SW480 cell was suppressed by quercetin in a time- and dose-dependent manner.

研究结果：经槲皮素处理的人结肠癌SW480细胞数量减少，部分细胞体积缩小，细胞膜完整，胞浆浓缩，核染色质固缩，细胞核碎裂，形成凋亡小体；MTT法检测显示当作用浓度为30μmol/L~90μmol/L时，槲皮素对人结肠癌SW480细胞的生长有抑制作用，其抑制作用随着作用浓度的增加和作用时间的延长而增强；流式细胞学发现槲皮素主要作用于人结肠癌SW480细胞周期的G1/S期，大部分细胞被阻断于S期，随药物浓度的升高和作用时间的延长，G0/G1期细胞比例逐渐增加，S期细胞比例逐渐减少；酶谱分析法检测显示不同浓度的槲皮素能够抑制人结肠癌SW480细胞分泌MMP-2及MMP-9，随浓度的升高，MMP-2及MMP-9的分泌量减少；免疫组织化学法显示不同浓度的槲皮素处理人结肠癌SW480细胞后，Cathepsin-D的表达随药物浓度的升高和作用时间的延长而降低。

Under inverted microscope, stereognosis of neurons died down, there were blank bulb in the neurons, dark particle were seen in the neuritis; the result of MTT test showed that the survival rate of neurons was obviously decreased. Res 40μmol/L increased the survival rate of neurons. Aβ(subscript 25~35) dose-dependently increased the level of MDA, decreased the level of GSH. Res dose-dependently increased the level of GSH, decreased the level of MDA.

结果：Aβ(下标 25~35)在终浓度为2μmol/L时神经元形态有明显的变化，细胞立体感减弱，胞体内空泡形成，细胞突起内可见黑色颗粒状物；细胞的存活能力明显下降，与对照组相比较有显著性差异(P.05)；Res在终浓度40μmol/L时，能明显提高细胞的存活能力。Aβ(下标 25~35)可以明显升高细胞培养上清液中MDA含量，降低GSH的含量；Res可使细胞培养上清液中MDA含量下降，GSH含量增加。

Flow cytometric analysis showed that after mock infection, Ad5null infection, and Ad5hSeFT infection, the mean fluorescence intensity values for the binding of Ulex europaeus Ⅰ lectin to NIH3T3 cells were 2. 3±0.6, 2. 1±1. 0, and 36. 5±5. 9, respectively; MFI values for the binding of Griffonia simplicifolia isolectin B4 (GS-IB4) lectin to NIH3T3 cells were 167±23, 170±19, and 100±14, respectively; MFI values for the binding of human natural IgG and IgM antibodies to NIH3T3 cells were 31±3, 32±4, and 22 ±4, respectively.

流式细胞分析结果表明，小鼠NIH3T3细胞及分别被Ad5null和Ad5hSeFT转导的NIH3T3细胞，UEA-1凝集素结合细胞的平均荧光强度值分别为2.3±0.6、2.1±1.0和36.5±5.9；GS-〓凝集素结合细胞的MIF值分别为167±23、170±19和100±14：人天然IgG和IgM抗体结合细胞的MFI值分别为31±3、32±4和22±4。

The frequency value in S phase is 4. 83 times of M phase, 2. 9 times of G1 phase, and 3. 41 times of unsynchronized cells. These results suggested that gene correction events were highly related to S phase. Then we compared the targeting efficacy for two groups of cells in S phase. One group was contact with thymidine and the other one went smoothly through S phase without replication blockage.

细胞水平检测实验分5组（K562组、K562/A02组、K562+ADM组、K562/A02+ADM组和K562/A02+TTD+ADM组），采用MTT法检测TTD对K562和K562/A02细胞的非细胞毒性剂量，流式细胞术检测细胞内阿霉素的浓度，基因、酶学、蛋白水平检测实验分3组（K562组、K562/A02组和K562/A02+TTD组），采用RT-PCR法检测mdr1 mRNA的表达，免疫细胞化学方法检测谷胱甘肽S转移酶π和拓扑异构酶Ⅱ的表达水平，Western-blotting法检测P-糖蛋白和bcl-2表达。

Total RNAs from KAx-3 cells and AK127 cells(developed for 14h) were isolated. After the reverse transcription and PCR reaction, two distinct differential fragments were acquired., fragment A was from KAx-3 cells and fragment C was from AK127 cells. After retriving and reamplifying the differentially expressed fragments, white-blue plaqueselection, the fragments were purified. Northern blot proved that fragment A was from KAx-3 cells and fragment C was from AK127 cells. The results of sequencing and researching for NCBI database have been showed: part sequence of fragment A shows 91% similarity to the gene encoding DhkA, 92% similarity to the gene encoding DhkF, 91% similarity to the gene encoding STATc, 97% similarity to the homoeobox gene encoding protein. These genes play important part in controlling cell differentiation and cell proportion in Dictyostelium discoideum.

本研究通过提取盘基网柄菌发育14小时的野生型KAx-3细胞和突变型AK127细胞的总RNA，运用mRNA差异显示法分离出了两条明显的差异表达片段，其中片段A来自野生型KAX-3细胞，片段C来自突变型Ak127细胞；并通过凝胶回收差异片段、对差异片段进行再次PCR、蓝白斑筛选克隆、提取质粒、酶切电泳纯化差异片段；接着进行Northern杂交的结果表明，片段A只与野生型KAx-3细胞的总RNA有杂交信号，片段C只与突变型AK127细胞的总RNA有杂交信号，这就排除了差异片段假阳性的可能；最后通过测序，搜索NCBI BLAST数据库发现：片段A的小部分序列与编码组氨酸激酶DhkA基因中一段序列的相似性高达91%，与编码组氨酸激酶DhkF基因中的一段序列相似性高达92%，与编码STATc蛋白基因的一段序列相似性达91%，以及与编码同源框蛋白的基因中的一段序列相似性达97%，这些基因在盘基网柄菌细胞分化和细胞比例调控过程中起着相当重要的作用，这些数据进一步说明了突变细胞不能完成发育的原因。
