Results Compared with that in the NP tissues, the number of α-SMA positive cells increased significantly in BPH tissue, and the number of vimentin positive cells increased moderately in the stroma and prominently surrounding the acinus and in the basal layer. In BPH tissue, the myosin and ERα staining signal was lost in the stromal cells surrounding the acinus, and the positive staining cells gathered into bunches in the stroma far away from the acinus, while the positive cells were sporadically distributed in the NP tissue. The PCNA positive cells increased moderately in the stroma and increased significantly in the basal layer.

与BEAS-2B相比较BEAS-2BNNK第1、2、5代细胞及胞核形态、细胞器形态及数量无明显差别；第10、15代细胞出现肿胀，细胞核逐渐变形，出现核畸形，核仁明显增大，边集，细胞器肿胀，数量明显增多，功能活跃具明显的转化细胞特征；第20、25代细胞及胞核明显畸变，出现明显的核碎裂及多个核仁，细胞器明显减少，具明显的肿瘤细胞特征。

Both testis and ovary are at stage IV in Nov. hi testis the germ cells are in spermiogenesis, and the mean GI is 0.93%. hi ovary the oocytes are mostly at phase 4, which are filled in the cytoplasm with vitellin granule, and the zona radiate in membrane begins to formation. Nucleus moves to one side of the oocyte gradually. The mean GI of ovary at this stage is 9.37%.

Methods Fourteen mutated vpr fragments were selected from patients with HIV. Both eukaryotic expression vector pcDNA3.1 and PCR products were purified, double-cut by Hind Ⅲ and BamH Ⅰ, and the cut products were legated and transformed into competent cells JM109. The 14 reconstructed plasmids were transfected into Hela cells. Cells with pcDNA vpr-wt, pcDNA vpr-Fs and pcDNA3.1 blank cells, and without pcDNA3.1 cell were established.

Total RNAs from KAx-3 cells and AK127 cells(developed for 14h) were isolated. After the reverse transcription and PCR reaction, two distinct differential fragments were acquired., fragment A was from KAx-3 cells and fragment C was from AK127 cells. After retriving and reamplifying the differentially expressed fragments, white-blue plaqueselection, the fragments were purified. Northern blot proved that fragment A was from KAx-3 cells and fragment C was from AK127 cells. The results of sequencing and researching for NCBI database have been showed: part sequence of fragment A shows 91% similarity to the gene encoding DhkA, 92% similarity to the gene encoding DhkF, 91% similarity to the gene encoding STATc, 97% similarity to the homoeobox gene encoding protein. These genes play important part in controlling cell differentiation and cell proportion in Dictyostelium discoideum.

本研究通过提取盘基网柄菌发育14小时的野生型KAx-3细胞和突变型AK127细胞的总RNA，运用mRNA差异显示法分离出了两条明显的差异表达片段，其中片段A来自野生型KAX-3细胞，片段C来自突变型Ak127细胞；并通过凝胶回收差异片段、对差异片段进行再次PCR、蓝白斑筛选克隆、提取质粒、酶切电泳纯化差异片段；接着进行Northern杂交的结果表明，片段A只与野生型KAx-3细胞的总RNA有杂交信号，片段C只与突变型AK127细胞的总RNA有杂交信号，这就排除了差异片段假阳性的可能；最后通过测序，搜索NCBI BLAST数据库发现：片段A的小部分序列与编码组氨酸激酶DhkA基因中一段序列的相似性高达91%，与编码组氨酸激酶DhkF基因中的一段序列相似性高达92%，与编码STATc蛋白基因的一段序列相似性达91%，以及与编码同源框蛋白的基因中的一段序列相似性达97%，这些基因在盘基网柄菌细胞分化和细胞比例调控过程中起着相当重要的作用，这些数据进一步说明了突变细胞不能完成发育的原因。
