In this paper, we studied M. meretrix ferritin, cathepsin B and caspase genes, which are involved in clam larval shell formation, nutrition, metabolism and apoptosis, respectively. We have cloned the three genes, investigated the temporal and spatial expression profile both at gene and protein level in trochophore (L1), D-veliger (L2), pediveliger (L3) and postlarvae (L4). The potential roles of these proteins were analyzed with specific inhibitors during larval development. Firstly, embryos were found developed into trochophore-like larvae with no shell if cultured at gastrula stage in artificial seawater without iron. Shell-like structures were formed only in the presence of iron. The larvae which had been transferred at L1 stage into ASW developed normal shell. This indicated that iron and iron associated protein are important for larval shell formation. The EST sequence which is homologous with ferritin, which is a principal iron metabolic protein, was selected from the M. meretrix cDNA library. The full-length of ferritin subunit cDNA was cloned by RACE. The results of real-time PCR revealed that the MmeFer mRNA expression changed before and after the larval shell formation.
本论文以文蛤幼虫为研究对象,分别对文蛤幼虫发育过程中贝壳形成相关的铁蛋白、营养及变态相关的组织蛋白酶B及变态过程中细胞凋亡相关的caspase三个基因进行了克隆,分析了基因及编码蛋白在担轮幼虫期(L1)、D形幼虫期(L2)、壳顶幼虫期(L3)和稚贝期(L4)的时空表达特征,解析了其可能的功能,并研究了相应酶类的特异性抑制剂作用对幼虫发育过程的影响,进行了目标蛋白的功能验证,详述如下:研究结果显示,在文蛤胚胎发育到原肠胚时放入不含铁离子的人工海水中培养,发育成无壳的畸形,随着人工海水中铁离子添加浓度的升高,幼虫长出壳状组织接近正常状态;而发育到L1期幼虫放入不含铁离子的人工海水中培养却可以发育出正常的壳,推测铁和铁代谢相关蛋白在幼虫贝壳初始形成有重要的作用。

To obtain more messages from Ardisia, some studies, such as characters of seeds germination and storage, factors influencing callus induction and growth, monitoring bergenin in callus of Ardisia spp and plant regeneration of Ardisia spp, were carried out. As a result, calli were obtained from nine species of Ardisia and bergenin, the major pharmaceutical element of Ardisia, was proved to exist in six of them when checked by HPLC. Plant regeneration systems were established from three species of Ardisia too. These works are all reported for the first time.

Fourthly, the power of social physical educators in Guangxi universities was so weak that it couldn't satisfy the need of specialty development. For example, the number of special teacher and the practicing ability of teachers are deficiency. The teachers that master special theoretic well and have practicing experience are deficiency especially. Fifthly, social physical practice base have not been building in Guangxi universities. And the necessary equipments of gymnasium are deficiency at the same time. Sixthly, the whole sports technology levels of new social physical students are low and hard to fit for social needing. Seventhly, the professional ideology of students is not stable enough. For example, the special concept and cognition of students are dim. The attitude for learning is not right. The most of students want to or have trend to change their profession after graduating. Eighthly, the ability of students fostered by social physical education in various Guangxi universities has a gap with the ability that students should hold for adapting the need of Guangxi employer. The special technical levels of social physical students are weak. Ninthly, in investigation of social physical personnel being demanded by Guangxi area employers, we found that few social physical personnels were demanded by business unit, the state-operated factory and mining enterprise because of being influenced by economical environment. And the more social physical personnels were demanded by social physical directors, fitness centers and each kind of sports association and so on.

The hFCECs were cultured by sticking tissues piece method that the cornea were divided into the limbus and central tissue piece and digestive method,respectively.For digestive method,the digestive juice of 5mg/ml DispaseⅡ+0.25%trypsin was chosed.D/F12 with 10%fetal bovine serum and 100 U/ml penicillin and streptomycin were used in this study,the culture condition was 37℃and 5%CO2,and culture medium changed every three days2、Passage of hFCECsAfter more than 80%confluenced,cells from corneal limbus hFCECs were digested for 5-15min with different concentrations of trypsin/EDTA at 37℃,and then passaged at a ratio of 1:2.The cells were subcultured on empty plates,mouse 3T3 fibroblast feeder layer,fetal corneal stromal cell feeder l ayer or HTK feeder layer respectively.And D/F12, mouse 3T3 fibroblast conditioned medium,fetal corneal stromal cells conditioned medium or HTK conditioned medium with 10%FBS and 100 U/ml penicillin and streptomycin were used respectively as the culture medium.And the culture medium were changed every three days as well.
方法一、人胎儿角膜上皮原代和传代培养1、原代培养严格无菌操作获取人胎儿角膜片,采用组织块贴壁法、酶消化法原代培养人胎儿角膜上皮细胞。2、传代培养角膜缘部的细胞生长融合达80%以上,不同稀释浓度的胰酶/EDTA消化液消化传代分别接种于空板、小鼠3T3成纤维细胞饲养层、胎儿角膜基质细胞饲养层及HTK饲养层,培养液分别采用D/F12、小鼠3T3成纤维细胞条件培养液、胎儿角膜基质细胞条件培养液、HTK条件培养液。
