RESULTS: The collapse was obvious at weeks 2 and 6 after modeling, with only few granulation tissues and fibrous tissues. No chondrocytes were observed. At weeks 12 and 18, fibrous tissues were found at the defect site, and bone tissue and cartilage proliferation was observed at the bottom and margin. However, it was greatly different from normal cartilage. Moreover, there was still no chondrocytes.

结果：实验兔后腿关节的股骨髁关节面的关节软骨缺损后，在制模后2，6周缺损部位凹陷明显，只有少许肉芽组织和纤维组织生长填充，未见软骨细胞爬行生长，12，18周见缺损部位以纤维组织增生为主，底部及边缘可见骨组织和软骨增生，但是镜下形态仍与正常软骨组织区别较大，表面仍未见软骨细胞爬行生长。

Result］ The new buds and petals of I.germanica was proper to be used as explant of tissue culture, with lower browning rate compared with other positions. The well situated tissue block of explant (3 mm×3 mm×3 mm) might reduce browning rate. Adding 0.1%Vc or 0.3% active carbon in medium had obvious effect on preventing tissue browning. The exogenous hormone 2 mg/L 6-BA was easy to lead to brownness turning, and 2 mg/L Kt and 2 mg/L NAA brought less effect to tissue browning. When using disinfectants of alcohol, its dipping time of 5～10 s for explant was the best, with the minimum browning rate. The culture under full illumination condition was liable to induce brownness turning, compared with other cultured conditions, the brownness turning could be restrained effectively by using imitating artificial climate condition for culture.

结果］ 鸢尾的新生芽和花瓣较适宜作组织培养的外植体，与其他部位相比褐变率较低；外植体组织块适中（3 mm×3 mm×3 mm）可降低褐变率；在培养基中加入0.1%Vc或0.3%活性炭对防止组织褐化效果明显；外源激素2 mg/L 6-BA易引发褐变，2 mg/L Kt和2 mg/L NAA对组织褐变影响不大；使用乙醇消毒剂时，对外植体浸泡时间以5～10 s为佳，褐变率最低；全光照条件培养容易诱发褐变，采用仿人工气候条件培养较其他条件培养可有效抑制褐变的发生。

It was observed that the influence of bud and its position on the healing process of grafting unions using hardwood sectioning method, and found that:(1) bud of a scion and its position to the graft unions have no influence on callusing formation of the isolation layers, and the early differentiation of cambia;(2) a scion bud closer to the grafting union would promote the formation of more compacted callus cells and the earlier formation of callus bridges;(3) conduit differentiation in grafting unions of lower-bud scions was later than that of other treatments.

利用滑动切片法对芽子有无及芽位对嫁接愈合过程的影响进行了观察，发现：(1)接芽有无及芽位对隔离层及愈伤组织形成和形成层的早期分化没有影响；(2)芽或去芽的芽位距接口近，可以促进愈伤组织的紧密连接，即愈伤组织桥的形成；(3)低位芽枝嫁接体中导管分化较晚，而其它处理导管的分化几乎是同时发生的，而且导管首先出现于愈伤组织薄壁细胞中。

The results showed with the increasing of NaCl concentration, the phenomenon of carnification could be found in the leaves of Arabidopsis thaliana . Under the stress of NaCl lower than 100 mmol/L, the tissue structure of root had no distinct difference from the control. When the concentration of NaCl was higher than 250 mmol/L, the stained color of safranine was deep but not even, and color in fibrovascular tissue and peripheral cortex cell were deeper than that around fibrovascular tissue.

当NaCl浓度小于100 mmol／L时，根组织结构与对照未表现出明显的差异；但当NaCl浓度达到250 mmol／L时，根组织结构对番红染料的着色较深而且不均匀，维管组织和外围皮层细胞着色较深，而维管组织周围的细胞着色较浅。

Content of flavone is the hightest in browning callus. It is supposed, that suppose which the last pace of synthesize flavone is in brovwing callus. SDS-PAGE cataphoresis electrophoresis show protein has big change from differentiating callus to browning callus. The change may has relation with change of enzymire activiation of control flavone synthesize.

黄酮含量的变化以褐化愈伤组织中含量最高，因此我们推测：黄酮合成的最后几步，可能发生在褐化愈伤组织中，从可溶性蛋白质的 SDS-PAGE 平板电泳结果表明，分化愈伤组织与褐化愈伤组织相比蛋白质谱带发生了明显的变化，这可能与控制黄酮合成的有关酶的活性变化有关。
