Methods The target cDNA was amplified by PCR and PCR products were indigested by restriction enzyme Nco Ⅰ and Hind Ⅲ. Then the indigested products were recombined into pET-BBH and pET-BBH was expressed in E. coli BL21 (DE3) by IPTG inducing. Next, the expressed proteins were identified by SDS-PAGE, western blot and enzyme activity test. Finally, fluorescence quantitative PCR was used to test its expression quantity in different worm stages.

The results showed that the free-living marine nematodes were the dominant group the percentage of which in the total number of meiofauna ranging betwean 76.1%~96.3%. The abundance of meiofauna in old mangrove district was the least and in bare beach where waste water flows was the most. There were a total of 37 putative species in the samples. The total number of recorded species was the lowest in the new mangrove district, and highest in the old mangrove district. The species diversity indexes, species evenness indexes and richness indexes in old mangrove district were all higher than those in new mangrove district. The dominant species and feeding types were different in three types of habitats.

By suspending cell culture from the callus of P. thugbergii and by fluorescent observation, a new method, which is easy and sensitive to observe cell activity and to test the phytotoxicity of various bacteria, was used. This method can be utilized to study the bacterial toxins. Cultured filtrates of some strains of the nematode-carrying bacteria were bioassayed for their toxicity to suspending cultured cells from P. thugbergii callus. Strong toxic effects were observed by the filtrates of Pseudomonas spp., P. fluorescence, and Sphingomonas spp., these bacteria species are coincide with those in the early stage of pine wilt disease.
