This experiment adopted organic solvent extraction step by step,following deproteinization,dialyzation,alcohol-precipitation and layer-separation of dextran,to reveal the unknown effective ingredients in pumpkin,which have influence to blood sugar in normal mice or model mice of diabetes mellitus.

为探寻南瓜中对正常及糖尿病模型小鼠血糖有影响的未知有效成分，本实验依次采用有机溶剂分步萃取、除蛋白、透析、乙醇沉淀及葡聚糖柱层析等分离手段，以小白鼠血糖值作为筛选活性成分的指标，在分离的每一阶段对分离所得各个组分进行活性定量评估，并追踪活性最强的部分，最后分离纯化出降血糖活性最强的组分G———南瓜多糖。

Nepalensis wall polysaccharide was prepared with hot water extraction,ethanol precipitation,dialyzation,and lyophilization.the crude polysaccharide was separated by deae-cellulose chromatography and purified by sephadex g-200 chromatography.the inhibit effect of the polysaccharide on lipid peroxidation of mice liver was conducted by tba method.the model of liver-injury induced by carbon tetrachloride（ccl4was used to validate the variety of the realated ast.results onefold ingredient polysaccharide was obtained after isolated and purified.conclusion the ingredient can remarkably inhibit lipid peroxidation of rat liver and significantly reduce activity of ast.

用热水提取，乙醇沉淀提取粗多糖，再经sevag法去除蛋白，透析去除小分子物质，冷冻干燥得楮头红多糖粗品；采用deae-52纤维素柱层析和sephadex g-200凝胶柱层析进行分离纯化；用tba法考察多糖的抗脂质过氧化作用；采用ccl4急性肝损伤模型，测定其血清中ast变化。结论获得一个楮头红多糖组分。tba法表明该组分对小鼠自发性脂质过氧化具有显著的抑制作用；在保肝试验中，给药组能明显抑制ast的升高，对肝损伤有一定的保护作用。

To investigate the expression, location and function, long distance PCR was used to expand the targets containing open reading frame. The expressive vectors of prokaryotic and eukaryotic cells were constructed after product purification and connecting with vectors. The prokayotic vectors were induced to express the protein by IPTG and the protein was seen by denaturalization PAG electrophoresis and dyeing. The eukayotic expressive vectors were transfected into culture cells and the protein was found in the nulceolus under the observation of the confocal microscope. The transfected cells were chosen by the geneticin (G418). The cell cyele was examined by flow cytometer and the cell numbers were reduced obviously in the stage of G〓-G〓 and G〓-M, but increased distinguishably in the S stage.
