纤维细胞

Under light microscope, on H-E stain section, four types cells can be distinguish: cells with weak basophilic fibrillar elements; cells with acidophilic granular substance; cells with strong basophilic fibrillar elements and ciliated cells. In the basal lamina region under gland epithelium, there are a few connective tissue; Surface view of the hypobranchial gland could be see by scanning electron microscope, there are cilia and different kinds of secretions distributed. Ultrastructure of the hypobranchial gland could be understand by transmission electron microscope, supporting cells, sensory cells and seven types gland cell were observed to form the glandular epithelium; cells with much rough endoplasmic reticulum,smooth muscle fiber and nerve endings were found beneath glandular epithelium, among basal lamina region.

光镜下H-E染色切片中腺上皮区仅可以区分出四种类型的细胞：弱嗜碱性纤维样细胞、强嗜碱性纤维样细胞、嗜酸性颗粒分泌细胞和纤毛细胞等；腺上皮下的基底膜区有少量结缔组织存在；扫描电镜下可以观察到鳃下腺表面的纤毛及腺细胞的分泌物等情况；透射电镜下观察到腺上皮中有支持细胞、感觉细胞和7种类型的腺细胞；近基底膜区观察到富含粗面内质网的细胞；基底膜间为薄层疏松结缔组织，内含肌细胞及神经末梢等结构。

结果表明：除檫木外，其它木材的管孔分布均为散孔材，具较高的单孔率；导管分子穿孔板兼有单穿孔和梯状穿孔2种或者仅具有单穿孔；导管间纹孔式为互列；导管－射线间纹孔式类型丰富，主要为刻痕状和大圆状。木射线有单列和多列射线2种类型，单列射线稀少、短，多列射线数量多；射线组织主要为异形Ⅲ和异形Ⅱ；轴向薄壁组织以傍管状为主，少数有带状或轮界状。油细胞和粘液细胞普遍存在于射线薄壁细胞或轴向薄壁细胞中。木纤维由韧性纤维和纤维管胞组成，部分树种具分隔纤维。

The results were as follows: The pituitary of Long-snout catfish included adenohypophysis and neurohypophysis. The adenohypophysis have six-cell types in common,i.e adrenocorticotro-phs,lactot-rophs,somatotrophs,gonadotrophs, thyrotrophs, and Melanotroph cells. In neurohypophysis, there were three types of neurosecretory fibres, types A1,A2 and B.There were different types'granules in type A neurosecretory fibres axone.In type B neurosecretory fibres axone, there were many small transparent vesicles.

结果表明：长吻魷脑垂体包括腺垂体和神经垂体两个部分，腺垂体组织中有六种分泌细胞即促肾上腺皮质激素分泌细胞、催乳激素分泌细胞、生长激素分泌细胞、促性腺激素分泌细胞、促甲状腺激素分泌细胞、促黑色素细胞刺激激素分泌细胞；神经垂体组织中存在A型(A_1、A_2)和B型神经分泌纤维，A型分泌纤维轴突中具有不同类型的分泌颗粒，B型分泌纤维轴突中含有许多透明小囊泡。

Results Trunk neural crest stem cells were successfully isolated and cultured. The immuocytochemcal result showed that the cells were nestin-and P75-positive. Trunk neural crest stem cells grew through adhering to chitosan fiber after inoculation. Under an electon microscope, the spindle-shaped trunk neural crest stem cells were proliferated and migrated along the fiber. The cells arranged side by side or linked by heads and tails in processes. The ends of S-100 positive cells were flat and expanded, adhering to the fiber as claw-shaped pseudopodium.

结果 采用神经管植块法培养的神经嵴干细胞，免疫细胞化学染色呈现nestin及P75双阳性，将其接种在壳聚糖纤维支架材料上，可见神经嵴干细胞贴附于壳聚糖纤维上生长；S-100免疫细胞化学染色显示，S-100阳性细胞突起的末端呈扁平状膨大；扫描电镜显示，壳聚糖纤维上的躯干神经嵴干细胞为梭形，呈现"端对端"、"肩并肩"排列，伸出爪形伪足贴附于壳聚糖纤维上。

The pcDNA〓-apoE〓 plasmid was transfected to SK-N-SH Neuroblastoma cell by liposome. It could be screened by G〓. The immunohistochemical assay was shown that neuroblastoma cell transfected by pcDNA〓-apoE〓 expressed the recombinant apoE protein. The intracellular expressed recombinant protein could inhibit the death of neuroblastoma cell induced by beta amyloidal peptides 25-35 fragment.

用pcDNA〓—apoE〓真核表达载体与脂质体共转染神经成纤维胶质瘤细胞，用G〓选择压力筛选，细胞免疫荧光检测表明，pcDNA〓—apoE〓转染的神经成纤维胶质瘤细胞表达了apoE〓重组蛋白，这种内源性的apoE〓重组蛋白对25μM Aβ25—35多肽诱导的神经成纤维胶质瘤细胞的死亡具有拮抗作用。

Results Among the 52 cases, 30 cases were confirmed to fibrosarcomas, which were identical with primary dingnoses;22 cases were revised to malignant fibrous histocytoma (8 cases), malignant nerve sheath tumor (4 cases), monophasic spindle cell synovial sarcoma (3 cases), leiomyosarcoma(3 cases), rhabdomyosarcoma(2 cases) and abdominal desmmoid tumor (2 cases). Of the 18 cases, only two cases were misdiagnosed and were revised to malignant fibrous histocytoma and spindle cell synovial sarcoma, respectively.

结果：1980年前52例中维持原诊断30例，修改诊断22例(42.3%)，分别为恶性纤维组织细胞瘤8例、恶性神经鞘膜瘤4例、单相性梭形细胞滑膜肉瘤3例、平滑肌肉瘤3例、横纹肌肉瘤2例、韧带状纤维瘤2例；1980年后纤维肉瘤18例中，维持原诊断16例，修改诊断2例(11.1%)，分别为恶性纤维组织细胞瘤和滑膜肉瘤各1例。

The cells,in the same population,were unanimous on the migrating direction and speed.There were two kinds of the shape of SCs on the stereoframe,spherical cells or long olivary cells;Cells contacted each other too.

Compared with SCs on the plane,SCs migrated easily on the stereoframe.The long olivary cell of SC was still trending to wrapping fibre up without neurons.

Objetive:To investigate the histopathological features and detect the mutation of JAK2V617F in PV、ET and IMF;to analyze the effects of the stages of myelofibrosis and the mutation on the number of leucocyte in PV and ET patients;Methods: the morphological features of 69 patients who dignosed in the first time were studied by HGF and Gomori stains ; the mutation of JAK2V617F was detected in 38 blood or marrow samples of PV、ET and IMF patients by real-time PCR;the effects of the stages of myelofibrosis and the mutation on leucocytes were analyzed;Results: Different stages of reticulin were observed in 15 PV and 15 ET patiens'marrow sections,and the cytosis、the pleomorphism and the bone trabecula side location of megakaryocytes were observed in all PV、ET and IMF patients;the means of leucocyte in patients with and without JAK2V617F were 20.4×109/Land 11.4×109/L respectively;the P value was 0.367 as comparing the leucocytes among different stages of reticulin increasing.

目的：探讨PV、ET、IMF患者的骨髓病理特点并且检测JAK2V617F突变的发生率，分析骨髓中纤维增生程度以及JAK2V617F对PV、ET患者白细胞数的影响；方法：69例初诊患者的骨髓切片经HGF染色以及Gomori染色后进行形态学研究；并利用real-time PCR法测38例患者的骨髓或者外周血的JAK2V617F突变；分析骨髓切片中纤维组织增生程度以及JAK2突变对患者白细胞数的影响；结果：15例PV和15例ET患者的骨髓切片中可以观察到不同程度的纤维组织增生，所有PV、ET、IMF患者骨髓病理中可见到巨核细胞增多、巨核细胞多形性改变以及巨核细胞定位于骨小梁旁；12例PV、4例ET、4例IMF中检测到JAK2V617F；有突变和无突变患者发病时白细胞数均值分别为20.4×109/L和11.4×109/L，比较不同纤维增生程度的PV、ET患者的白细胞数均值，P值为0.367。

The histopathologic changes included lymphocyte and monocyte infiltration,hyperplasia of synovial cells and small vessels,interstitial fibrosis,hyaline degeneration and cartilaginous metaplasia.The immunohistochemical observations showed that the high expressions of IL-1 and IL-6 were significantly different between the pathologic plicae and the control groups（P＜0.01）.The positive expressions of IL-1 and IL-6 were the synovial cells and monocyt-lymph cells in the pathologic synovial plicae.The positive expressions of MMP-1 and TIMP-1 have significant difference between the experiment and control group（P＜0.01,P＜0.05）.The expression of MMP-1 was positive in synovial lining cell,monocyte,fibroblast,endothelial cell in small vessel and chondrocyte.The TIMP-1 expression was detected in the synovial lining cells and a small quantity fibroblast.

结果 正常滑膜皱襞和病理性滑膜皱襞在滑膜细胞增生及小血管增生、间质纤维化及玻璃样变、软骨化生组织学改变方面，差异均有显著性（P＜0.01）；IL-1、IL-6在病理性滑膜皱襞内的增生滑膜细胞、单核及淋巴细胞和在正常滑膜皱襞内的表达差异均有显著性（P＜0.01）； MMP-1、TIMP-1在病理性滑膜皱襞和正常皱襞内的阳性表达，差异具有显著性（P＜0.01，P＜0.05），MMP-1在增生滑膜衬里层细胞、单核和纤维母细胞、血管内皮细胞和软骨化生的软骨细胞呈阳性表达；TIMP-1只在滑膜衬里层细胞和少量纤维母细胞有表达。
