纤维原细胞

Methods HPDLFs were primary cultured from tissue explants, and the cells of the 5th to 8th passages were used after immunohistochemical identification of keratin and vimentin expressions. The cells were divided into 5 groups and treated with TP at 1, 0.5, 0.25, 0.125, and 0.0625 mg/ml, respectively, with another group without TP treatment as the blank control group. Cell counting and MTT colorimetric assay were performed to assess the cell proliferation, and flow cytometry was employed to determine the DNA content of the HPDLFs.

采用组织块培养法培养原代人牙周膜成纤维细胞并传代，经免疫组化SABC法检测角蛋白和波形丝蛋白鉴定，取5~8代细胞用于实验；按茶多酚不同浓度分1mg/ml、0.5mg/ml、0.25mg/ml、0.125mg/ml、0.0625mg/ml组和空白对照组，采用细胞计数法、MTT法检测细胞增殖情况，流式细胞术检测细胞DNA含量。

Most endothelial cells fell off, elastic fibre autolyze; the number of SMC decreased, some SMC nucleus contracted completely or depressed, typical apoptosis body were seen in later stage, muscle silk autolyzed and disappeared mostly; in outer layer saw much collagenic fiber, arranged unorderly; inflammation cells were seen in the aneurysm wall(hypertrophy cell et al.).

多数瘤壁的内皮细胞溶解脱落，不连续，弹力纤维自溶；动脉瘤血管壁均可见SMC明显减少，较多SMC染色质边聚、固缩，部分SMC核全部固缩或出现不规则凹陷，SMC核胞浆或胞核部分出现脱落或裂解成碎片，晚期可见典型的凋亡小体出现，肌丝大都自溶消失：外膜可见大量的胶原纤维，排列较紊乱；瘤壁还可见炎性细胞浸润。

Results:Wound healing rate in group A was higher than that in group B and the wound healing time was 1～3 days earlier in group A.More active proliferation of epidermic cells around wounds showed in group A.The function of fibroblasts in wounds,collagen synthesis and secretion,were more vigorous in group A than those in group B.

结果：A组创面愈合率高于B组，其创面愈合时间较B组提前1～3 d；创周表皮细胞增殖也较B组活跃，创面成纤维细胞较B组呈现出更旺盛的合成与分泌胶原纤维的功能。

Objective To observe the ultrastructure of supra interspinous ligament of ankylosing spondilitis,to evaluate the role of fibroblasts in ossification of ligament,and to discuss if disorderly arrangement of collagen and sedimentation of calcium granule in AS are induced by fibroblasts.

目的 观察强直性脊柱炎棘上棘间韧带的超微结构和探讨成纤维细胞在韧带骨化中的作用。找出AS韧带中胶原紊乱排列和钙颗粒沉积是否由成纤维细胞活动引起。

The positive signal of ER and ER mRNA distributed in parenchymal and non-parenchymal hepatic cells, especially near the hepatic centrolobular and periportal areas. Ovariectomy decreased ER level and ER mRNA expression significantly (P＜0.05). Hepatic ER and ER mRNA concentration were elevated after treatment with estrogen in both ovariectomy and sham ovariectomy fibrotic groups (P＜0.05), and the elevation of ER was correlated with a marked decrease in hepatic damage and fibrosis.

5肝脏ERα及其mRNA基因的表达分布于肝细胞及肝脏非实质细胞，特别以肝脏中央静脉和门脉周围为主；卵巢切除减少ERα及其mRNA基因的表达（P＜0.05）；雌二醇治疗在肝纤维化和卵巢切除大鼠均提高肝脏ER及其mRNA基因的表达（P＜0.05），其增加程度与肝脏组织学损伤、纤维化指标的降低程度以及肝脏胶原沉积减少相平行。

RESULTS: The extract of Aml could reduce the abnormally high levels of ALT and AST. Examined by light microscope , the degeneration and necrosis level of hepatocytes and the distribution of collagen fibrosis were markedly increased in the model group. The pathological changes and the c ollagen fibrosis associated with hepatic fibrosis decreased significantly in the prophylactically treated groups.

结果： 蓍草提取物预防给药能降低 CCl 4所致大鼠血清中异常增高的 ALT 、 AST 含量，光镜下观察肝纤维化模型组大鼠肝细胞严重变性、坏死，胶原纤维明显增加，蓍草预防各组大鼠肝细胞病理损伤得到明显改善。

Results: The cultured cells showed typical shape and features of fibroblast cell ,which were successfully dissociated and purified.

结果：原代培养的成纤维细胞特征典型，并通过传代培养得到了纯化的成纤维细胞。
