MATERIALS AND METHODS: Arg to Ser mutation was introduced into the 249 position of the p53 gene by knock-in method. These ES cells with this mutation were selected according to the homologues-recombination with PCR and Southern blot. The positive ES cells without a selection marker were injected into blastocysts recovered from Hprt(superscript -/-) mice, which were derived from Hprt-deficient ES cells. The injected blastocysts then were implanted into pseudopregnant females.

材料与方法：利用基因打靶技术在小鼠胚胎干细胞p53基因249编码子中引入点突变，使编码子249由精氨酸变成丝氨酸，然后将含突变的ES细胞显微注射到Hprt小鼠囊胚中，将注射过的囊胚植入假孕的雌性小鼠子宫，到第14d取小鼠胚胎纤维母细胞，用含HAT的培养液筛选出从ES细胞分化而成的鼠EF细胞，经测序证实细胞含有由249Arg到Ser的突变。

Coadministration of a subeffective dose of L-NNA (3 pmol) or ODQ (10 nmol) with morphine prevented its antiedematogenic effect, but sildenafil (0.15 nmol) rendered a subeffective dose of morphine effective (18 nmol).

亚效应剂量 L-精氨酸(3 pmol)或 ODQ(10 nmol)与吗啡同时注射则会抑制后者的抗水肿作用。

On the normotensive rat, bilateral microinjection of L-arginine (16 n mol/0.1 ul/site), a precursor of nitric oxide into the rVLM, caused a decrease in BP, HR and the pressor response induced by the dPAG stimulation, while microinjection of NG-methyl-Larginine (L-NMMA, 16 n mol/0.1 ul/site), a blockade for NO to form, into the rVLM bilaterally, caused an increase in BP, HR and the pressor response induced by the dPAG stimulation. There was no effect of both drugs mentioned above on the tachycardiac response induced by the dPAG stimulation.

五、在正常血压大鼠，于双侧rVLM微量注射一氧化氮前体(16nmol/0.1μl/site)可引起BP降低、HR减慢、以及电刺激dPAG诱发的防御性升压反应减弱；而于同区微量注射生成一氧化氮的拮抗剂N〓-甲基-L-精氨酸(16nmol/0.1μl/site)则可引起BP升高、HR加快、以及电刺激dPAG诱发的防御性升压反应增强；而上述两药对电刺激dPAG诱发的防御性加快心率反应均无明显作用。

The result indicted that the RSD of precision and reproducibility were less than 3%,conforms to the related regulation;In conclusion that this method is accurate,repeatable and provides a scientific basis for controlling the quality of Pinellia ternate.

结果表明，该方法精密度和重复性RSD均小于3％，符合有关规定；半夏清制和姜制后，其所含精氨酸与半夏生品相比均有不同程度的下降，而且指纹图谱的主峰数目也有所变化。

目的：研究硝基精氨酸赖氨酸三肽对组成型一氧化氮合酶和诱导型一氧化氮合酶的抑制程度。

After administration of L-N〓-nitro-Arginine, a specific antagonist of NOS, it was found that the basal and stimulated NO release of blood vessels (aorta, internal carotid, renal and mensenteric artery) was decreased further. The increase of coronary blood flow stimulated by Ach was also reduced further, while the platelet aggregation degree and erythrocyte aggregation index increased significantly. Pathological examination indicated that aortic endothelium was destroyed, the blood vessels in cerebral, cardiac and renal tissues were hypertrophied further and some were almost occluded. The above fin dings resulted in SHR feeded with L-NNA having high incidence of stroke rate and high degree of renal cirrhosis when compared to control SHR. The infarct and hemorrhagic focuci in cerebral tissues, infarct focuci in cardiac tissues, atrophy and hyalinedegeneration of renal glomeruli and degeneration even necrosis of renal tubuli in renal tissues were al so found. The increase ratio of brain and heart to body weight and decrease ratio of kidney to body wei ght further demonstrated that the cerebral, cardiac and renal injuries were aggravated.

在上述结果的基础上进一步给予SHR NOS特异性拮抗剂左旋亚硝基精氨酸后，血管(胸主、颈内、肾及肠系膜动脉)NO基础释放及激动剂刺激释放进一步减少，Ach刺激冠脉流量的增加和胸主动脉内膜NOS活性也进一步减少，血小板聚集程度及红细胞聚集指数显著上升，病理切片显示胸主动脉内皮细胞破坏明显，脑心肾组织内的血管进一步增厚甚至几乎阻塞，导致饲L-NNA的SHR缺血性及出血性脑卒中发生率显著高于对照SHR，脑体重比显著升高；心肌内出现明显的缺血梗塞疤痕灶，心体重比显著升高；肾小球明显萎缩、玻璃样变性，肾小管也变性甚至坏死，使肾硬化程度显著增加呈颗粒性固缩肾，肾体重比也显著减少。

The vasomotion was induced by noradrenaline in vivo, then the changes of vasomotion after injecting SI and Na-monomethyl-L-arginine （L-NMMA, a NO synthase inhibitor） were measured respectively on a TV monitor using a TV camera mounted on the microscope, and the influence of L-NMMA on effect of SI was also observed.

Methods Eighteen Wistar rats were randomized into 3 groups, namely the CLP group (n=6) in which the rats were subjected to cecal ligation plus puncture to induce severe abdominal infection, L-Arg group (n=6) where the rats received 300 mg/kg peritoneal L-Arg injection following CLP establishment, and the control group (n=6) where the rats underwent ventrotomy only.

Methods Male SD rats underwent WIR stress for 2,4 and 8 hours. Ulcer index of stomach was chedsed by Guth method, nitrite production was measured by Greiss assay, nitric oxide synthase activities and L-arginine transport rate were determined by isotope tracer method.
