In this thesis preliminary study of genes relevant to symboiosis of prototrophic revertant 10NH-1 was carried out by molecular cloning.

The tryptophan auxotrophs and the kanamycin resistant prototrophic revertants have been proved to contain a copy of Tn5 inserted in the chromosome by Southern Blot analysis with DIG-labeled 3. 5 kb HindⅢ fragment of pkan2 as the Tn5 probe. Four typies of prototrophic revertants could be found by the above assay.

用Tn5探针进行的Southern分子杂交证明Tn5在色氨酸营养缺陷型菌株及部分Km抗性原养回复突变型菌株中均插入在染色体DNA上，并且对菌株本身具有的内源性质粒的数目及大小没有带来影响。

Twenty-eight prototrophic revertants of the tryptophan auxotrophs were obtained through spotaneous back mutation from colonies grown on synthetic medium plates or nodules of plants. Most of the revertants have resumed the nodulation abilities and enhanced nitrogen fixation activities as evidenced by darker green color, higher dry weight and more nitrogen contents in shoot than those of the wild type strain 8-1-4A did in pot experiments. However, noodulation abilities decreased to some extent and no significant differences were found in the mass of nodules compared with the parent strain.

通过自发回复突变，从培养平板和根瘤中分离到28个色氨酸原养回复突变型菌株，对其中25株进行植物盆栽试验证明其中24株菌的结瘤功能得到了恢复，而且固氮酶活性普遍比出发菌有所提高，其中41％达到极显著水平，18％达到显著水平，植株地上部分干重及含氮量也有不同程度的提高，唯有根瘤数比出发菌有所降低，其中23％降低达极显著水平，11％降低达显著水平。

OBJECTIVE: To construct a novel adenoviral eukaryotic expression vector that can co-express mutant hypoxia-inducible factor-1 alpha (HIF-1α) target protein and humanized Renilla reniformis green fluorescent protein reporter molecule under normoxic conditions.

To get more information of the function of human Era on cell cycle, the complete control inducible mammalian expression system was used to express human Era mutant and analyzed cell cycle of uninduced (add 1% ethanol) and induced (10μM Pon A) cell lines by flowcytometer, the primary results demonstrated human Era mutant could inhibit apoptosis induced by ethanol, and Era S36N may stimulate the expression of Bcl-2 as detected by Western Blot.
