磷酸

A process based upon solvent extraction has been developed for the recovery of phosphoric acid from a leaching liquor obtained by decomposition of phosphate rock with sulfuric acid.

本文采用二丁基亚砜作为萃取剂，对于用硫酸分解磷矿而得的粗磷酸进行多级逆流萃取及反萃取，以获得高纯度的磷酸和磷酸盐。

Results Before taking medicine, model rats' cAMP/cGMP and ACTH were lower than normal rats', and they showed a trend of rise with adding dose. Before taking medicine, model rats' GLU was lower than normal rats'. With adding dose, model rats' GLU increased, however, normal rats' GLU reduced. When taken medium dose white prepared lateral root of aconite, model rats' CHO was higher than normal rats'. Taking medicine made both groups ALT rise, but normal rats' went up more quickly than model rats'. Model rats' BUN was higher than normal rats', and taking low dose or little dose medicine could lead to incline of reduction.

结果 给药前模型组环磷酸腺苷/环磷酸鸟苷和促肾上腺皮质激素低于正常组，随着给药剂量的增加，两者有升高的趋势；给药前模型组血糖低于正常组，随着给药剂量的增加，模型组GLU有升高的趋势，而正常组有下降的趋势；中剂量给药时模型组总胆固醇显著高于正常组；随着给药剂量的增加，2组丙氨酸氨基转移酶均有升高的趋势，且正常组ALT升高的幅度更大；给药前模型组尿素氮高于正常组，低、小剂量给药有降低模型组BUN的趋势。

After amplying a 2.2kb fragment form the PPV-SC1 RF-DNA,we clone the fragment into pMD 18-T,named pTNSl.The whole sequence which is 1989 bp long was determined by sequencing, including the complete ORF of PPV-SC1 NS1 which encoding 662 amino acids.Alignment of pairs of sequence indicates that there are 98% and 99% similary with other porcine parvovirus strains Kresse and NADL-2, respectively. Multiple sequence alignment discloses that there are a few difference between ppv-scl nsl gene and other ppv nsl gene: A-G at 39nt,T-C at 153nt,A-G at 175nt, A-C at 1117nt, A-C at 1535nt .Alternative codon in ppv-scl nsl have distinctly different frequentfy by codonbias analysis at EMBOSS(http://genopole.toulouse.inra.fr/bioinfo/emboss). Thereis not distinct hydrophobicity and transmenbrane helices in ppv-scl nsl protein. Struction domain anslysis of PPV-SC1 NS1 protein indicate that there are a ATP/GTP-binding site motif A at 398-405,16 Protein kinase C phosphorylation site,21 Casein kinase II phosphorylation site,and 3 cAMP/cGMP-dependent protein kinase phosphorylation site.At the same time ,there is a same motif between ppv-scl nsl and Poxvirus D5 protein-like which may share in the same fuction which is necessary during virion duplication.

将PPV-SC1 NS1序列与其他PPV NS1基因进行多序列比对，结果显示，PPV-SC1 NS1与其他的PPV NS1的同源性较高，仅存在个别的差异，分别是第39位A→G，第153位T→C，第175位A→G，第1117位A→C，第1535位A→C；同源搜索比较表明，PPV-SC1与PPV NS1同源性可达98％、99％，与其他的细小病毒NS1基因也存在很大的保守性；密码子偏向性分析结果表明PPV-SC1 NS1基因在同一氨基酸的不同密码子的选择上存在一定的偏向性；PPV-SC1 NS1蛋白总体上说具有亲水性不存在明显的疏水性区段，用swiss TMPRED软件预测PPV-SC1 NS1的跨膜区，返回的结果并没有得到有显著意义的跨膜区的存在；根据基于motif数据库的结构域预测，PPV-SC1 NS1的第393-415位氨基酸残基存在潜在的ATP／GTP结合位点，该蛋白还存在16个蛋白激酶C磷酸化位点，21个酪蛋白激酶2磷酸化位点，3个cAMP-／cGMP依赖蛋白激酶磷酸化位点，PPV-SC1 NS1蛋白与POX_D5(痘病毒D5蛋白)具有一致的保守结构域，推测NS1可能与POX_D5有类似的功能。

The experimental results show that: DNA of normal gastric mucosa has stable phosphate backbone; Since the peak intensity at 1090 cm^(-1) is lower than that at 1050 cm^(-1), phosphate backbone in DNA of intestinal metaplasia becomes unstable, while the rest spectra peaks are similar to those in normal gastric mucosa; In terms of adenocarcinoma DNA, double peaks at 1090 cm^(-1) whose intensity are higher than 1050 cm^(-1), are observed, which indicates that DNA chain is broken and re-forms a double stabilized phosphate backbones. In addition, peaks at 950 cm^(-1), 1010 cm^(-1), 1100~1600 cm^(-1) also exhibit obvious difference compared with normal gastric mucosa, which shows that deoxyribose and bases are changed due to broken of DNA.

实验结果表明，胃粘膜正常组织DNA中有稳定的磷酸骨架；肠化组织基因组DNA拉曼特征峰在1090 cm^(-1)处谱峰强度低于1050 cm^(-1)处谱峰强度，表明其磷酸骨架变得不稳定，其它位置的谱峰与正常组织相似；腺癌组织基因组DNA拉曼特征峰在1090 cm^(-1)附近出现双峰，其相对于1050 cm^(-1)处谱峰强度更强，提示DNA有可能出现断链并再次形成了稳定的磷酸骨架，在950 cm^(-1)、1010 cm^(-1)、1100~1600 cm^(-1)波段的特征谱峰与正常组织DNA相比变化也较大，说明脱氧核糖和碱基可能由于DNA断链也随之改变。
