The results showed that RSAP used two primers of 18 nucleotides in which starting at the 3' end of each primer were restriction site sequences (4-6 bases),followed 12-14 bases of arbitary sequence, the differences between two primers were restriction sites and filler sequence; PCR am-plification was run for the first 5 cycles with an annealing temperature of 35℃, followed by 35 cycles with an annealing temperature of 48℃; the PCR reaction of 25 μL included 20ng DNA templates, 2.5mmol/L of Mg(superscript 2+), 0.2 mmol/L of dNTP, 1.5U of Taq DNA polymerase, 600nmol/L of each primers. RSAP is a new maker technique with simplicity, moderate throughput ratio and reliability. RSAP is of good reprodicibity and broad application.

结果显示，RSAP技术的引物为2条长度均为18bp的引物，引物的3'端为4~6个碱基的限制性酶切位点序列，接着是12~14个碱基的随机序列，2条引物的限制性位点和随机序列不同：PCR扩增的前5个循环采用35℃的退火温度，随后的35个循环采用48℃的退火温度；在25μL反应体系中，模板DNA用量为20ng，Mg(上标 2+)浓度为2.5mmol/L dNIPs浓度为0.2 mmol/L，Tap DNA聚合酶用碳为1.5U，2条引物浓度均为600mmol/L；RSAP技术重复性好，适用性广泛，是一种操作简便、产率中等、稳定可靠的DNA标记技术。

Calcium carbonate and soluble glass and so on were confected as alkaline antifouling agent. The Infrared Spectrometry results showed that 2275-2250 cm~(-1) diagnostic absorb wave crest in part A and 3390-3200 cm~(-1) in part B and 1730 cm~(-1) in coating film strongly proved polymers reacted just as the reacting mechanism, that means polysiloxane which offer low-surface-energy property linked Polyurethane system by —OH groups reacting with —NCO groups. Relative molar mass of part A and part B was determined by Gel Permeation Chromatography and the results were appropriate. Surface-energy was determined and results were between 33.6-47.17 mN/m and it increased when alkaline antifouling agent was put into the coating. Alkaline antifouling agent was releasing steadily when the coating film was dipped in seawater. As time gone, the pH value of seawater was increasing. Solubility parameter and results showed that there was no corrasive phenomenon between the polyethylene strand and the coating.

红外光谱分析结果表明，出现在A组分中的2275～2250cm~(-1)处的异氰酸酯特征吸收峰、B组分中的3390～3200cm~(-1)处硅羟基的特征吸收峰及出现在固化后涂膜中的1730cm~(-1)处氨基甲酸酯基团的特征吸收峰证明了涂料成膜物质交联固化符合本文的设计要求，即赋予涂膜低表面能特性的有机硅组分通过—OH基团与—NCO基团的反应已经链接到聚氨酯体系中；凝胶渗透色谱结果表明，成膜物质的A、B组分均为低分子量，符合预聚物的要求；涂膜表面能的测算结果表明，涂膜的表面能在33.6～47.17mN／m之间；加入碱式防污剂后涂膜的表面能略有升高；碱式防污剂的释放平稳，随着涂膜在海水中浸泡时间的增长，pH值有增加的趋势；用溶解度参数原则来评价涂料与网绳的涂装，结果表明，本试验研制的防污涂料对聚乙烯网绳无溶蚀作用。

Experimental results indicated that the contaminants generally existed in mussel, such as α-HCH,β-HCH,γ-HCH,δ-HCH, o·p-DDT, p·p-DDT, p·p-DDD, were degraded completely under the above conditions, whereas DDE, aldrin and dieldrin were partly degraded. No detectable degradation of PCB 1242 and PCB 1248 were observed. Thus the conventional preparation of the contaminants using the columns packed with Florisil or SiO〓 could be left out.

All index proved that the P5776 cDNA library constructed in this research had high quanlity and could be used for further ESTs sequencing and analyzing. The amplified library of P5776 was screened on white-blue plates, and more than 5,000 white-clones were selected to culture in 96-well plates, then the high quanlity plasmids of white-clones were extracted by traditioal alkaline-lysis method using the Vitagene?96-easy plasmid extract kit. Through PCR primered by T3( the vector has T3 RNA polymerase promoter ), the target fragments of inserts were get. Then the inserts were taged by four different fluorescence-dye, and fractionated by capillary in ABI Prism? sequencer. More than 5,000 5′-ESTs had been sequenced and through rough selecting, 4,747 ESTs were remained for further research. Among the remained ESTs, more than 90% have longer lenth than 500bp, and there are less than 5% unable-read bases in every sequence.

将扩增好的P5776 cDNA文库铺布蓝白斑筛选平板，挑取其中的白斑经活化培养后，运用Vitagene 96-easy质粒DNA制备试剂盒以碱裂解法共提取出58 板(96孔板)，即5，000 余个重组子克隆的高质量质粒DNA；提取好的质粒以T3(连接载体上具有该引物RNA聚合酶启动子)为引物经PCR(Polymerase Chain Reaction，聚合酶链式反应)扩增出重组子插入片段，标记上荧光染料后在ABI Prism 3100 测序仪上经毛细管电泳完成了5，000 余条5′端EST 序列的测定工作，初步筛选出4，747 条质量较好的EST序列，经筛选得到的EST中90%以上具有大于500bp 的可读序列，每一EST序列中不可读碱基数小于5%。

Results:Berberine inhibited HUVEC adhesion for lymphocyte when pretreated HUVEC and antagonised IL-1 and TNF-induced upregulation of HUVEC adhesion for lymphocyte.Meanwhile,IL-1-stimulated lymphocyte adhesion to HUVEC was diminished in the presence of berberine.The main molecular mechanism of berberine's function may be downregulating the expression of ICAM-1 on the HUVEC .

结果：小檗碱能抑制静息的及IL-1、TNF激活的内皮细胞与淋巴细胞间的粘附，其主要分子机制为下调内皮细胞表面粘附分子ICAM-1的表达；小檗碱能抑制IL-1激活的淋巴细胞与内皮细胞的粘附，对淋巴细胞表面粘附分子CD18的表达无影响。

RESULTS:Berberine inhibited IL-1, TNF-induced HUVEC adhesion for PMN when pretreated HUVEC and antagonised IL-1, TNF-induced upregulation of ICAM-1 on HUVEC. Meanwhile, TNF-stimulated PMN adhesion for HUVEC and CD18 upexpression on PMN was diminished in the presence of berberine.

结果：小檗碱与IL-1或TNF共同处理HUVEC，能抑制IL-1、TNF诱导的PMN与HUVEC间的粘附增强，且能下调由IL－1、TNF诱导的HUVEC表面粘附分子ICAM-1表达的增高；小檗碱与TNF共同处理PMN，能抑制TNF诱导的PMN与HUVEC的粘附增强，亦能降低TNF诱导的PMN表面粘附分子CD18的表达。

After experiments, we got some results as the following:1.Induced by semi-conductor laser, solid DNA changes as below: 1the deoxyribose-phosphate backbone vibrations in DNA decreasing, these bands are at 884~890cm~(-1),1112cm~(-1) and 1168cm~(-1). 2 deoxyribose and C-O stretch vibration in d increasing, there are new bands occurring at 1436cm~(-1) and 1072cm~(-1), the intensities of these bands at 1466cm~(-1) and 1030cm~(-1) increase.
