Objective It was reported that rALR (rat augmenter of liver regeneration) distribute in most of tissue and organ of rat, no-tissue specility; mRNA expression of rALR was identified variedly in different tissue, strongest in testis. But nowadays there was not some study on ALR in rat testis. Here to confirm the expression and localization of ALR in infancy,adult and old rat testis,comparing the change of ALR expression degree before per 70 PH and after per 70 PH. In order to evaluate expression principle of ALR in rat testis and know if ALR has tight connection with spermatogenesis and development of testis, to explore the relation between ALR and reproduction, to discover the possible expression character and functional methods of ALR.

目的：文献报道rALR分布于各组织器官，无组织特异性，但其mRNA在各组织的表达强度有差异，在睾丸组织表达最强，目前对大鼠睾丸ALR表达尚无深入的研究；本实验研究不同年龄段大鼠睾丸组织中肝再生增强因子的表达水平、表达部位、以及表达的方式，进而探讨睾丸ALR与睾丸发育、精子生成以及生殖活动的关系；同时探究睾丸组织ALR表达是否参与了肝脏的再生过程中，进一步了解睾丸ALR表达的特性及作用方式。

Based on pre-experiment foundation of the earlier period, 60 male patients among 45-65 year old were chosed as the clinical case according to the PADAM diagnosis standard, and were divided into 3 groups according to the block stochastic principle, the observation group (acupuncture combined with BUSEHN capsule), group 1, group 2 (takes orally BUSEHN capsule group), each group had 20 cases, and each group of patients was measured by the method of radio-immunity for the serum level of testosterone, luteinizing hormoha, follicle-stimulating hormone before and after the treatment. By the contrast of the grade of PADAM symptom among three groups before and after treatment ,we can reveal the whole funcamental characteristic of acupuncture combined with herb ,and discuss a new way to enhance the curative effect of acupuncture ,and summarize the endocrinous adjustment of acupuncture for the PADAM's patients and the orchidic improvement by the treatment of acupuncture.

在前期预实验的基础上，选择符合诊断标准的PADAM诊断标准的45-65岁男性患者临床病例60例，按照区组随机原则分为3组，观察组、对照组1、对照组2，每组各20例，采用放射免疫法分别测定各组患者治疗前后血清中血清睾酮、黄体生成素、卵泡刺激素的血清浓度的水平；通过三组及三组间治疗前后PADAM症状评分指数的对比研究，揭示针药结合的整体调节作用特色，探讨提高针灸疗效的新途径；总结出针刺对中老年雄激素缺乏综合征患者的内分泌的调节及针刺对睾丸的改善程度，探讨针药结合治疗本病的机制及针灸增效剂的应用前景，为今后进一步研究打下坚实基础。

Results The expression of the Mahoganoid protein and its mRNA was showed by brown staining and observed in the Leydig cells, spermatogonia, primary spermatocytes, spermatids, Sertoli cells, and peritubular myoid cells of testicular tissues and principal cells, basic cells and epithelial cells of epididymal tissues.

结果1。免疫组化法检测Mahoganoid蛋白的实验结果显示的棕色阳性信号可见于各天龄段大鼠睾丸的生精细胞、支持细胞、间质细胞和管周肌样细胞以及附睾输出管的高柱状、低柱状上皮细胞和附睾管的上皮细胞、主细胞、基细胞。其中Mahoganoid蛋白主要表达于胞膜和胞浆。

During the first two weeks after birth, the growth performances were not significant between the mutant mice and the normal ones, but after that, the mutant mice grew much slower. The differences were significant in the indexes of eyes, lungs, thymuses, and adrenal glands between the genders. At the same time, the differences were significant in the organ indexes, such as lungs, adrenal glands, uterine and testicles between the normal phenotype mutant mice and the abnormal ones. And these two kinds of mice showed different amount of hemoglobin and red blood cell.

结果：B6-Co小鼠不育率22%、平均产仔数5.62±2.55只、离乳成活率56.59±41.22显著低于正常的B6小鼠，而胎间隔29.50±10.50天比正常B6小鼠短。B6-Co小鼠出生后2周内生长发育与正常B6小鼠基本一致，2周后，其生长发育慢于正常B6小鼠。B6-Co小鼠雌雄间的眼球、肺、胸腺、肾上腺的脏器指数差异有统计学意义。B6-Co表型异常小鼠与B6-Co表型正常小鼠相比，肺、肾上腺、子宫、睾丸的脏器系数差异有统计学意义。B6-Co小鼠表型异常小鼠在血红蛋白和红细胞数量上异于B6-Co表型正常小鼠。

The results showed that (1) COX-2 mRNA and protein existed in the mature male rat testis. COX-2 was localized in spermatocytes by immunohistochemistry.(2) COX-2 also existed in adult man testis. COX-2 protein was positively stained in spermatocytes and Leydig cells.(3) 2 weeks after administration of rofecoxib, the level of testosterone in the whole testis reached its lower values, being only 50% of control values. However, testosterone level recovered during 4 weeks. After such treatment, histologic examination of these testes showed atrophy of the seminiferous tubules and maturation arrest of spermatogenesis.(4) 2 weeks post-EDS, expression of COX-2 decreased significantly (P.005), in comparison with vehicle-treatment control. 4 weeks after treatment, a new generation of fetal like Leydig cells repopulated in the testicular interstitium resulting in COX-2 expression partially recovered. Although the expression of COX-2 mRNA and protein are enhanced at 8th week after using external testosterone, it wasn't significant higher than control group.

实验研究证明：(1)正常成年雄性大鼠睾丸组织中COX-2在mRNA和蛋白质水平均存在表达，免疫组织化学结果显示COX-2定位于曲细精管内的生精细胞；正常成年男性睾丸组织中同样存在COX-2表达，免疫组织化学结果显示COX-2定位于曲细精管内的生精细胞和Leydig细胞；(2)服用特异性COX-2酶抑制剂rofecoxib 2周后，实验组大鼠睾丸组织内睾酮的含量减少，为正常对照组的50％；持续用药4周后睾丸组织内睾酮浓度逐渐恢复至正常水平；COX-2酶活性降低后病理组织切片显示睾丸内曲细精管萎缩，生精紊乱，持续用药4周时影响最明显；(3)注射特异性Leydig细胞杀灭剂EDS 2周后，实验组大鼠睾丸组织内睾酮浓度降至极低水平时，COX-2的蛋白和基因表达水平也显著低于正常空白对照组（P.005）；使用EDS后第4周，睾丸组织中的睾酮浓度逐渐回升，同样组织中COX-2蛋白表达和mRNA表达水平也相应提高接近正常水平；使用EDS后4周开始给予外源性睾酮，6周和8周时COX-2表达水平的绝对值虽有提高，但与正常大鼠空白对照组比较并无显著性差异，说明外源性雄激素刺激睾丸合成COX-2的作用并不明显。
