1、抗原的提取：收集新疆不同地区分离得到的利什曼原虫前鞭毛体，经培养，用0.01mol/L pH7.2的磷酸盐缓冲液漂洗，超声粉碎，比色法测定其蛋白质含量，间接ELISA法滴定出抗原的工作稀释度。2、检测：用提取的三株抗原，分别进行间接ELISA法测定黑热病病人血清和疫区健康人血清抗体，并以肺结核病人血清、非疫区健康人血清做对照，三株抗原所用检测方法一致，血清工作稀释度均为1∶100。
