According to the result, we believed that the reason resulted in the low sensitivity was that the marker residue compound MQCA structure has changed after conjugated with protein and the antibody induced by the conjugation could not recognize the free MQCA in the ci-ELISA. Therefore we designed some hapten which could reserve the marker residue structural speciality mostly. After synthesis route selection and optimum, the hapten 7-Hydroxymethyl-3-methyl-quinoxaline-2-carboxylic acid was synthesized, used 4-methyl-2-nitroaniline as the start material via six steps, such as oxidation cyclization, bromize reaction, hydrolysis, Beirut reaction, reduction, hydrolysis. This work established basis for producing the high affinity antibody and developing the sensitive ELISA residue detection method.

Methods: 2-ethyl aniline was used as starting material of the synthesis and was processed by acetic anhydride acylation, strong nitric acid nitraction at low temperature, and desacyl synthesis to produce 2-ethyl-5-nitro aniline. The product was then synthesized to 3-methyl-6-nitro-lH-indazole by using ring-closing reaction in the presence of NaNO2, and then ethylated to obtain 2, 3-diethyl-6- nitro-2H-indazole. The latter compound was then reduced by SnC12/Hcl to get target compound 2, 3-diethyl-6-nitro-2H-indazole.

We have synthesized 4,4'-dinitro-3 4''-dimethyl triphenylamine,(1), in 84% yield in an one-step condensation process between 3,4-dimethyl aniline and 4-fluoro-nitrobenzene. After the auto-oxidation of (1) and the subsequent hydrogenation of the nitro-groups, an AB2 type triphenylamine polyimide intermediate bearing two amine groups and one acid-anhydride group each on individual phenyl ring,{AB2 PI Intermediate}, was prepared successfully. The yields of the autoxidation and the hydrogenation steps are 81% and 90% respectively.
